Constructed lentiviruses carrying siRNA against Cx37 significantly knocked down Cx37 mRNA and protein expression in vitro. A total of 60 mice with gastric cancer were randomly divided into the Cx37 siRNA group, the mock‑siRNA group and the control group. Cx37 siRNA, mock‑siRNA and saline were separately injected (with the lentiviruses transfected into the gastric cancer cells). Following six weeks, the Cx37 mRNA expression, Cx37 protein expression and tumor apoptosis were detected using semiquantitative reverse transcription‑polymerase chain reaction, western blot analysis and terminal deoxynucleotidyl transferase‑mediated dUTP nick end labelling, respectively. Six weeks following lentiviral transfection, the Cx37 mRNA levels in the Cx37 siRNA group, mock‑siRNA group and saline group decreased to 42, 63 and 67%, respectively (P<0.05). The mock‑siRNA group demonstrated no significant change in Cx37 levels compared with the control group. Western blot analysis revealed lower Cx37 protein levels in the Cx37‑RNAi group than in the other groups (0.21±0.07 vs. 0.65±0.06 vs. 0.54±0.07), and that the apoptotic index of the Cx37‑RNAi group was higher than those of the mock‑siRNA and control groups (19.7±5.1 vs. 9.8±6.4 vs. 10.5±7.2%, 11.1±6.9; P<0.05). In conclusion, it was demonstrated that Cx37 siRNA is correlated with gastric cancer. Interference of Cx37 effectively reduces Cx37 mRNA and protein expression and promotes tumour apoptosis.

Effects of small interfering RNA silencing of Connexin 37 on subcutaneous tumor of gastric cancer in mice
Chinese journal of experimental surgery, 2014Co-Authors: Yuan-ming Jing, Haixin Qian, Xiaoping Zhang, Ai-jing Sun, Feng TaoAbstract:Objective To construct lentiviruses targeting mice Connexin 37 (CX37) small interfeting RNA (siRNA) and modelling the gastric cancer with murine gastric cancer cell line.The lentiviruses were transfected into gastric cancer.Methods Lentiviruses carrying siRNA of CX37 were constructed,which knocked down mRNA and protein expression of CX37 significantly in vitro.Sixty mice with gastric cancer were randomly divided into CX37 siRNA group,mock-siRNA group and the control group,receiving injection of CX37 siRNA,mock-siRNA and normal saline respectively.After 6 weeks,the expression of CX37 mRNA and protein was detected by using semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting respectively.Apoptosis was examined by using TdT-mediated dUTP nick end labeling (TUNEL).Results Six weeks after lentivirus transfection,the results showed that the CX37 mRNA levels in CX37 siRNA group (42%) were significantly lower than in the mock group (63%)and control group (58 %) (P < 0.05).Western blotting revealed the expression of CX37 protein in CX37siRNA group was lower than in other three groups (0.21 ± 0.07 vs.0.65 ± 0.06 vs.0.54 ± 0.07).At the same time,the apoptotic index in CX37 siRNA group was higher than in mock-siRNA group and control group [(19.7±5.1)% vs.(9.8±6.4)% vs.(10.5±7.2)%,P<0.05].Conclusion CX37 siRNA can significantly inhibit the growth of the gastric cancer cells. Association between C1019T polymorphism in the Connexin 37 gene and Helicobacter pylori infection in patients with gastric cancer.

Connexin 37 gene polymorphism 1019 site genotypes were determined by gene sequencing technology. Genotypes and alleles frequencies were compared. Results: (1) Connexin37 gene 1019 site distribution frequency (CC type, TC type, TT type) in the CSG group was 18.1%, 45.1% and 36.8%; in the stomach cancer group it was 35.1%, 45.9% and 19.%, conforming to the Hardy-Weinberg euilibrium. (2) In comparison with CSG group, the frequency of Connexin37 C allele was higher in the gastric cancer group (58.0% vs 40.7%, OR = 2.01, 95%CI = 1.58-2.57, P < 0.01). The prevalence of gastric cancer risk was significantly increased in the carriers of C allele (CC+TC) than in TT homozygote (OR = 2.47, 5%CI = 1.68- 3.610. (3) Gastric cancer patients complicated with Hp infection 211 cases, gastric cancer group of the male patients with HP positive patients with 187 cases, 40 cases of female patients with negative patients, 24 cases were HP positive, negative in 137 cases, control group male patients, 28 cases were Hp positive, negative in 95 patients, female patients with Hp positive 6 cases, 75 cases were negative. Compared with TT homozygotes, (CC+TC) genotype prevalence of gastric cancer risk increased significantly (OR = 2.96, 5%CI = 1.76-2.99 ). Conclusion: The T allele in the Connexin37 gene might not only be associated with gastric cancer but also with H. pylori infection.

Lentiviruses expressing small interfering RNA (siRNA) of Cx37 were constructed, and were shown to significantly knockdown the mRNA and protein expression of Cx37 in vitro. Sixty pigs on a high‑fat diet were randomly divided into three treatment groups of saline, mock or Cx37 siRNA, to induce plaque formation. The Cx37 lentiviral suspension was transfected into the abdominal aortic plaques of pigs. Plaque characteristics were detected by intravascular ultrasound and the expression of Cx37 mRNA was detected by semi‑quantitative polymerase chain reaction. The expression of Cx37 protein was analyzed by western blot analysis. Two months after lentivirus transfection, Cx37 mRNA levels were decreased by 38% in the Cx37 siRNA group, by 60% in the mock‑siRNA group and by 63% in the saline group (P<0.05). The mock group showed no significant changes in Cx37 expression as compared with the saline group. Cx37 protein expression was lower in the Cx37 siRNA‑treated group as compared with the other groups (0.21±0.07 vs. 0.65±0.06 vs. 0.54±0.07). The percentage of plaque necrosis at 10 months (two months following RNAi) was decreased in the Cx37 siRNA group as compared with that at eight months, prior to RNAi (5.26±2.11 vs. 7.83±1.03%, P<0.05). In the mock‑siRNA and saline groups, no differences (P=0.074, 0.061, respectively) were observed. In the Cx37 siRNA group, plaque volumes following 10 months decreased relative to those following eight months, prior to RNAi (21.03±6.24 vs. 31.23±10.23, P<0.01). By contrast, in the mock siRNA and saline groups, plaque volumes after 10 months were increased relative to those following eight months (38.54±13.56 vs. 32.12±11.21 mm3, 37.36±14.21 vs. 30.21±12.02 mm3, P=0.031, P=0.027). Atherosclerotic plaque formation was effectively decreased through the downregulation of Cx37 mRNA using Cx37 siRNA.

Methods A total of 1126 cases of EH were diagnosed in the People's Hospital of Wuxi City, China. A control group consisted of 874 healthy people, i.e., non-EH patients. All cases were genotyped by DNA sequencing. Results Polymorphism C1019T on the Connexin37 gene was found in the whole population. The distribution of three genotype frequencies in both groups was in accordance with the Hardy-Weinberg equilibrium. The frequency of the CX37C allele was higher in EH patients (57.4% vs. 42.1%, χ 2 =92.5, P 2 =49.0, P 2 =13.4, P 2 =38.7, P Conclusion The C allele in the CX37 gene might be associated with the susceptibility to EH in population of Wuxi, China.

The mock‑siRNA group demonstrated no significant change in Cx37 levels compared with the control group. Western blot analysis revealed lower Cx37 protein levels in the Cx37‑RNAi group than in the other groups (0.21±0.07 vs. 0.65±0.06 vs. 0.54±0.07), and that the apoptotic index of the Cx37‑RNAi group was higher than those of the mock‑siRNA and control groups (19.7±5.1 vs. 9.8±6.4 vs. 10.5±7.2%, 11.1±6.9; P<0.05). In conclusion, it was demonstrated that Cx37 siRNA is correlated with gastric cancer. Interference of Cx37 effectively reduces Cx37 mRNA and protein expression and promotes tumour apoptosis.

Chinese journal of experimental surgery, 2014Co-Authors: Yuan-ming Jing, Haixin Qian, Xiaoping Zhang, Ai-jing Sun, Feng TaoAbstract:Objective To construct lentiviruses targeting mice Connexin 37 (CX37) small interfeting RNA (siRNA) and modelling the gastric cancer with murine gastric cancer cell line.The lentiviruses were transfected into gastric cancer.Methods Lentiviruses carrying siRNA of CX37 were constructed,which knocked down mRNA and protein expression of CX37 significantly in vitro.Sixty mice with gastric cancer were randomly divided into CX37 siRNA group,mock-siRNA group and the control group,receiving injection of CX37 siRNA,mock-siRNA and normal saline respectively.After 6 weeks,the expression of CX37 mRNA and protein was detected by using semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and Western blotting respectively.Apoptosis was examined by using TdT-mediated dUTP nick end labeling (TUNEL).Results Six weeks after lentivirus transfection,the results showed that the CX37 mRNA levels in CX37 siRNA group (42%) were significantly lower than in the mock group (63%)and control group (58 %) (P < 0.05).Western blotting revealed the expression of CX37 protein in CX37siRNA group was lower than in other three groups (0.21 ± 0.07 vs.0.65 ± 0.06 vs.0.54 ± 0.07).At the same time,the apoptotic index in CX37 siRNA group was higher than in mock-siRNA group and control group [(19.7±5.1)% vs.(9.8±6.4)% vs.(10.5±7.2)%,P<0.05].Conclusion CX37 siRNA can significantly inhibit the growth of the gastric cancer cells. Association between C1019T polymorphism in the Connexin 37 gene and Helicobacter pylori infection in patients with gastric cancer.

(2) In comparison with CSG group, the frequency of Connexin37 C allele was higher in the gastric cancer group (58.0% vs 40.7%, OR = 2.01, 95%CI = 1.58-2.57, P < 0.01). The prevalence of gastric cancer risk was significantly increased in the carriers of C allele (CC+TC) than in TT homozygote (OR = 2.47, 5%CI = 1.68- 3.610. (3) Gastric cancer patients complicated with Hp infection 211 cases, gastric cancer group of the male patients with HP positive patients with 187 cases, 40 cases of female patients with negative patients, 24 cases were HP positive, negative in 137 cases, control group male patients, 28 cases were Hp positive, negative in 95 patients, female patients with Hp positive 6 cases, 75 cases were negative. (4) The distribution frequency of C allele in patients with Hp infection was much higher than that in Hp negative cases in the GC group (64.5% vs 47.0%, OR = 2.05, 95%CI = 1.54-2.74, P < 0.01). Compared with TT homozygotes, (CC+TC) genotype prevalence of gastric cancer risk increased significantly (OR = 2.96, 5%CI = 1.76-2.99 ). Conclusion: The T allele in the Connexin37 gene might not only be associated with gastric cancer but also with H. pylori infection.

Expression of gap junction genes was assessed by immunohistochemistry and real-time polymerase chain reaction (PCR) in transplanted cryopreserved ovarian tissue compared with that of normal ovarian tissue. Apoptosis of ovarian cells was evaluated by using the terminal deoxynucleotidyl transferase-mediated biotinylated deoxyuridine triphosphates nick end-labeling method. After subcutaneous transplantation, Cx37 and Cx43 mRNA and protein expression were significantly lower in cryopreserved than in normal ovarian tissue. Apoptosis was increased in granulosa cells from antral follicles of the cryopreserved tissue. After cryopreservation and subcutaneous transplantation of ovarian tissue, proteins forming gap junctions between oocytes and granulosa cells are under-expressed compared with normal controls.
