Three groups of 6 or 7 calves were selected based on different egg excretion patterns. SDS-gel electrophoresis of adult Cooperia antigen under reducing conditions, followed by Western blotting, revealed that resistance of individual calves to C. oncophora might be related with antibody responses (42 days post infection) against at least 2 protein fragments (14–16 kDa and 27 kDa). The 14–16-kDa protein complex was bound, to some extent, by individual sera from all calves. The intensity of staining was negatively correlated with egg excretion on Day 42 p.i. Calves with high egg counts on Day 21 p.i. either did not or only weakly recognized the 27-kDa band. It has to be established whether the 14–16 kDa (or recombinant 14.2 kDa) provides a tool for immunodiagnostics and whether the 27-kDa fragment can help further unravel immune-mediated resistance to Cooperia .

SDS-gel electrophoresis under reducing conditions, followed by western blotting, revealed that a complex of 14–16 kDa protein fragments, a 27 kDa and a 31.6 kDa band of Cooperia oncophora adult antigen were recognized by sera from calves both after primary and trickle infection. A 19 kDa and 30 kDa protein fragment were additionally bound by sera from calves after trickle infection. The data suggest that the systemic humoral immune response of calves to Cooperia oncophora after trickle infection is more pronounced (14–16, 27, 31.6 kDa) and extended (19, 30 kDa) than after primary infection with Cooperia .

C. oncophora adults differed from the L 4 larvae at the 31 32 and 37 kDa level. Several adult and L 4 proteins were bound by biotinylated Concanavalin A, as was also true for L 3 proteins. A 31 32 kDa glycoproprotein of adult worms was recognized by a monoclonal antibody with specificity for phosphorylcholine. Using monoclonal antibodies in ELISA and Western blotting, the serum antibody response of C. oncophora -infected calves to adult worm antigen was almost entirely IgG1. Binding of the IgG1 antibodies was restricted to a complex of reduced 12–15 kDa protein(s) and a 27 kDa fragment of adult worms. The data suggest that the systemic humoral immune response of calves during a primary infection with C. oncophora consists mainly of an IgG1 response, and is directed to a non-glycoslyated Cooperia protein (molecular weight estimated at 12–15 kDa under reducing conditions and 18 kDa under nonreducing conditions). This protein is probably present in both L 4 larvae and adults. Since it was not bound by immune sera from calves mono-infected with several other nematodes, the 12–15 kDa protein complex may represent a Cooperia -specific component that can be used for serodiagnosis.

Acquired immunity against Cooperia spp and Ostertagia spp in calves: Effect of level of exposure and timing of the midsummer increase.

Kloosterman, F W RietveldAbstract:Abstract In two experiments, groups of calves were exposed to different levels and patterns of infection with Ostertagia spp. and Cooperia spp. The experimental design simulated the stereotypic pattern of herbage infestation, including both a normal and a delayed midsummer increase, under conditions of set-stocking. After this simulated ‘first grazing season’, calves were followed during the subsequent winter housing. At the end of that housing period some calves were collected with 100 000 L3 Cooperia spp. and 40 000 L3 Ostertagia spp. and slaughtered 23 days later. All previously infected calves were protected against the establishment of the challenge infection with Cooperia spp., but not against Ostertagia spp. For the latter a significant negative correlation was found between worm count and previous level of exposure to infection. During the simulated first grazing season, changes in the ratio of Cooperia to Ostertagia eggs in the faecal egg output and the genus-specific egg count were influenced by both the level of exposure and the timing of the midsummer increase. It is concluded that acquired immunity against both parasite genera develops depending on the level of exposure to infection during a first grazing season, and that delaying the midsummer increase results in a delay of the acquisition of an effective immunity as measured by faecal egg counts and the ratio of Cooperia to Ostertagia egg output.

Age resistance in calves to Ostertagia ostertagi and Cooperia oncophora

FrankenaAbstract:Abstract Calves were infected repeatedly during a period of 6 weeks with Ostertagia ostertagi and Cooperia oncophora, at an age of 3, 6 or 9 months. The inoculations were performed during three periods, February–March, May–June and August–September, to account for possible seasonal effects or effects of larval batches. Observations were done on faecal egg output, antibody titres and weight gains. Calves were slaughtered for post mortem examinations 9 weeks after the start of infections. The influence of age on worm populations and egg output was significant for C. oncophora but not for O. ostertagi. The effect of season or larval batch on worm populations was significant for O. ostertagi but not for C. oncophora. The correlations between worm numbers and several other parameters found for Cooperia were strongly indicative of a process of worm expulsion taking place at the stage of infection (9 weeks after the start of infections) when post mortem examinations were done. Such correlations were absent for Ostertagia. It is concluded that within the range of ages examined here (the range to which first season grazing calves belong), there is no influence of age on Ostertagia populations but a clear effect of age on Cooperia. This difference strongly influences the total faecal egg output of grazing calves and its interpretation.

To this end, serum and mucus antibody responses were measured in animals primary-infected with 30000 or 100000 L3, treated with anthelmintics and subsequently challenged with 100000 L3. A detailed phenotypic and functional analysis of B cells was done in animals infected once or twice with 100,000 L3. Based on the similarity in parasitological variables of animals primed with 30000 or 100000 L3, we concluded that with these doses priming conferred protection in a dose-independent way. Upon challenge significant increases in Cooperia-specific serum and mucus IgG1 and IgA and total serum IgE titres were induced in primed animals in a dose-independent way. In contrast, intermediate and low responders differed in the onset of the production of Cooperia-specific serum IgG1. Furthermore, not only the onset but also the level of total serum IgE significantly differed between intermediate and low responders. Phenotypic and functional analysis of B lymphocytes revealed that (i). priming induced the generation of memory B cells which upon challenge readily differentiated into antibody secreting cells; (ii). sensitised B cells were more efficiently recruited to the intestinal effector sites; (iii). based on the expression of CD62L and CD86 two distinct B cell subpopulation could be differentiated. CD62L(+)CD86(-) B cells that were likely lymphocytes not yet activated and with an enhanced recirculation capacity, and CD62L(-)CD86(+) B cells that were activated B cells with a reduced recirculation ability; and finally (iv). the increased expression of CD86 and subsequent correlations with parameters of the T helper 2 immune response induced by C. oncophora, suggested that CD86- interactions are involved in the generation of protective immunity against Cooperia.

For this, 66 animals naturally infected, four to 36 months of age, raised in pasture, were selected. The results of necropsy revealed the presence of seven genera and 12 species with the following prevalence and mean count: Haemonchus contortus: 100.0% (2947.2); Trichostrongylus colubriformis: 90.9% (3048.8); Cooperia curticei: 56.0% (256.5); Oesophagostomum columbianum: 48.4% (36.0); Cooperia punctata: 30.3% (94.5); Trichostrongylus axei: 22.7% (26.5); Strongyloides papillosus: 19.6% (83.0), Haemonchus contortus (L4): 7.5% (17.2), Cooperia pectinata: 10.6% (12.9), Trichuris ovis: 10.6 % (0.6); Cooperia spatulata 4.5% (0.3); Capillaria bovis: 4.5% (0.1). The mean parasitism of helminthswas 6524.7 per animal. Haemonchus contortus (adults and L4) and Trichostrongylus colubriformis corresponded to 45.4% and 46.7% of the average worm burden totally, respectively. Based in the results obtained in this study, can be concluded that the two most abundant species of helminths and important, the micro-region of Jaboticabal are Trichostrongylus colubriformis and Haemonchus contortus, and these two species amounted to 92.1% of the distribution percentage of helminths collected from all animals. These results demonstrate the importance of conducting a counts of eggs per gram of feces (EPG) in the herds of this region when FAMACHA is used on a particular property, since this method control does not allow to diagnostic the damage/clinical signs in animals infected by T. colubriformis, because this specie does not have hematophagism habit on animals

Fauna helmintológica de ovinos provenientes da microrregião de Jaboticabal, estado de São Paulo, Brasil

Para tanto, foram utilizados 66 animais naturalmente infectados, de quatro a 36 meses de idade, criados em regime extensivo. Os resultados necroscópicos revelaram a presença de sete gêneros e 12 espécies, com a seguinte prevalência e a média de parasitismo: Haemonchus contortus: 100,0% (2947,2); Trichostrongylus colubriformis: 90,9% (3048,8); Cooperia curticei: 56,0% (256,5); Oesophagostomum columbianum: 48,4% (36,0); Cooperia punctata: 30,3% (94,5); Trichostrongylus axei: 22,7% (26,5); Strongyloides papillosusi: 19,6% (83,0); Haemonchus contortus (L4): 7,5% (17,2); Cooperia pectinatai: 10,6% (12,9); Trichuris ovis: 10,6% (0,6); Cooperia spatulata 4,5% (0,3); Capillaria bovis: 4,5% (0,1). Haemonchus contortus (Adultos e L4) e Trichostrongylus colubriformis corresponderam a 45,4% e 46,7% da carga parasitária média total, respectivamente. Pode-se concluir que as duas espécies de helmintos mais abundantes e importantes da microrregião de Jaboticabal/São Paulo foram Trichostrongylus colubriformis e Haemonchus contortus, sendo que essas duas espécies perfizeram 92,1% da distribuição percentual dos helmintos recolhidos de todos os animais. Tais resultados demonstram a importância em se realizar um monitoramento das contagens de ovos por grama de fezes (OPG) dos rebanhos desta região, quando o método FAMACHA for empregado em uma determinada propriedade, uma vez que este método de controle, geralmente, não permite diagnosticar os danos/sinais clínicos desencadeados nos animais pelo T. colubriformis, em função de essa espécie não possuir hábito de hematofagismo sobre os hospedeiros

Prevalence and intensity of infection by helminths in bovines from the dairy basin of São Carlos, SP, Brazil

Do material coletado do tubo digestivo, pulmões e pâncreas, foram observadas as espécies de helmintos com as seguintes prevalências e intensidades médias de infecção: Haemonchus contortus, 100% e 988,06; Haemonchus similis 43,24% e 50,01; Trichostrongylus axei, 24,32% e 16,00; Cooperia punctata 100% e 3649,45 ;Cooperia pectinata 44,59% e 436,70; Bunostomum phlebotomum, 9,45% e 3,17; Oesophagostomum radiatum, 86,48% e 138,35; Trichuris discolor, 21,62% e 7,44;Agriostomum wryburgi, 2,70% e 0,22; Dictyocaulus viviparus 13,51% e 51,62; Eurytrema coelomatlum, 14,86% e 27,67.On a preliminary study of bovine helminths on dairy herds of São Carlos, SP, Brazil, that includes the counties of São Carlos, Ribeirão Bonito and Dourado, 74 holstein-zebu mixed breeding animals, 8 to 20-month old, were necropsied. Helminth fauna of bovines from the Central-Western region, Minas Gerais State, Brazil

Os resultados necroscópicos revelaram a presença de nove gêneros e 16 espécies de helmintos, com a seguinte prevalência e média de intensidade de infecção: Haemonchus placei (100,0%; 3895,5); Haemonchus similis (29,0%; 159,6); Cooperia punctata (100,0%; 5595,0); Cooperia spatulata (32,9%; 137,8); Cooperia pectinata (34,2%; 1010,5); Trichostrongylus axei (69,7%; 239,2); Trichostrongylus colubriformis (10,5%; 10,8); Trichostrongylus longyspicularis (2,6%; 0,5); Ostertagia ostertagi (2,6%; 3,1); Ostertagia lyrata (2,6%; 1,5); Ostertagia trifurcata (1,3%; 0,3); Oesophagostomum radiatum (94,7%; 470,9); Trichuris discolor (47,4%; 32,5); Strongyloides papillosus (1,3%; 0,1); Capillaria bovis (9,2%; 10) e Bunostomum phlebotomum (2,6%; 0,3). A carga parasitária média foi de 11.558,5 helmintos por animal. Dos 76 bovinos necropsiados, 92,1% estavam infectados por três a sete espécies de helmintos. Apenas 7,9% dos hospedeiros mostravam-se parasitados por oito espécies diferentes de helmintos. Este estudo mostra o primeiro relato das espécies Ostertagia lyrata e Ostertagia trifurcata no Estado de Minas Gerais. É importante ressaltar que, no presente trabalho, por meio da identificação dos helmintos colhidos nas necropsias, foi possível observar que está ocorrendo uma inversão na intensidade parasitária média, com uma diminuição no número de Cooperia e um aumento nos valores de Haemonchus, em comparação com os valores relatados na literatura.Seventy-six naturally infected bovines, males and females of mixed breed, aged 8 to 12 months-old, were necropsied.
