In model experiments Corn grits were spiked with FB1 (2 mg/kg) and d-glucose (50 g/kg) or sucrose (50 g/kg) and manufactured into extrusion Products at various temperatures (160−180 °C) and moisture levels (16−20%). A liquid chromatography/electrospray ionization−mass spectrometry method using isotopically labeled fumonisin FB1-d6 as an internal standard was developed for the determination of NCM-FB1. For sample cleanup solid-phase C18 cartridges were used. The detection limit achieved with this method was 10 ng/g (signal-noise ratio = 3:1) using the protonated molecule [M + H]+ signal of NCM-FB1 (m/z 780) in the selected ion monitoring mode. Liquid chromatography/electrospray mass spectrometry in combination with selected ion monitoring (SIM) was used for unambiguous detection of FB1 and HFB1 after extraction of samples with acetonitrile/methanol/water (25:25:50) and solid-phase C18 cleanup. Quantitation was carried out using labeled fumonisin FB1-D6 as an internal standard. For the quantitative determination of fumonisin FB1 in Corn Products, a rapid method without any additional clean-up or purification of the sample extract was developed utilizing high-performance liquid chromatography-electrospray mass spectrometry. Using labelled fumonisin FB1–D6 as an internal standard the quantification limit for FB1 was set at 20 ng/g sample with a signal-to-noise ratio of 10 : 1.

With deuterated fumonisin FB_1-d_6 as internal standard the routine quantitation limit for FB_1 was set at 400 pg (0.8 ng g^−1 sample) with a signal-to-noise ratio of 10∶1. Almost all Corn grit and Corn meal samples under study were found to be contaminated with fumonisin FB_1 and FB_2 in concentrations ranging from 1 to 200 ng g^−1. Lower fumonisin levels were found in Cornflakes, popCorn and baby food, while no fumonisins were detected in canned sweet Corn.

Journal of Cereal Science, 2013Co-Authors: Saima Majeed, Mazhar Iqbal, Shahzad Zafar IqbalAbstract:Abstract In present study, a total of 275 retail cereal samples including rice, Corn and Corn Products was analyzed for the presence of aflatoxins and ochratoxin A (OTA) using HPLC equipped with a fluorescence detector. The data has shown that 38 out of 68 samples of rice, 37/105 of Corn and 43/102 samples of Corn Products have been found contaminated with AFs, average level of AFB 1 and total AFs, 8.23 and 19.54, 7.90 and 12.08, and 5.47 and 7.85 μg/kg, respectively. Furthermore, 34, 28 and 26 numbers of rice, Corn and Corn product samples were found to be contaminated with OTA with an average level of 12.94, 5.29 and 3.69 μg/kg, respectively. The samples of rice, Corn and Corn Products found above the permissible European Union (EU) limit for AFB 1 and total AFs were 18, 13 and 14 and 28, 14 and 20%, respectively; however, the samples of rice, Corn and Corn Products above the EU limit were 40, 14 and 15%, respectively.

Development and Validation of a Cost-Effective HPLC-FLD Method for Routine Analysis of Fumonisins B_1 and B_2 in Corn and Corn Products

Therefore, regulatory limits for food and feed were established in many countries. Herein, a sensitive, valid, cost-effective, and easily transferable analytical method for quantitative determination of the relevant fumonisins B_1 (FB1) and B_2 (FB2) in Corn and Corn Products was developed. The final method is based on reversed-phase high-performance liquid chromatography with fluorescence detection (RP-HPLC-FLD). Derivatization of analytes was simplified by establishing an automated precolumn derivatization method by using an autosampler. Strong anion exchange (SAX) cartridges were used instead of widespread immunoaffinity chromatography (IAC) approaches. Analytes and matrix signals were chromatographically separated in a total run time of 25 min by means of a C_8 column and a gradient elution using methanol and phosphate buffer. The limits of detection (LODs) for FB1 and FB2 were 29.2 and 17.5 μg/kg, respectively. Obtained recoveries ranged from 79.4 to 94.5 % for FB1 and from 79.9 to 98.0 % for FB2. Positive confirmation of the developed method was achieved by participating in an interlaboratory proficiency testing. Nineteen samples of sweet Corn and Corn Products were analyzed to demonstrate the applicability of the method.

A survey of aflatoxin B_1 and total aflatoxin contamination in baby food, peanut and Corn Products sold at retail in Indonesia analysed by ELISA and HPLC

This research was undertaken to determine the degree of aflatoxin contamination in different Indonesian foodstuffs. A preliminary survey was carried out to evaluate the level of total aflatoxin (AfT) and aflatoxin B_1 (AfB_1) contamination of baby foods, peanut Products, and Corn Products, which were purchased from traditional markets and supermarkets in Indonesia during the year 2001-2002. Eighty two peanut Products, 12 baby foods Products, and 11 Corn Products from different brands were analysed for AfT and AfB_1 using the Enzyme-Linked Immunosorbent Assay (ELISA) method. The results indicate that, of the brands analysed, 35% of the peanut Products were contaminated with aflatoxins at various levels (range 5 to 870 μg/kg). Peanut-chilli sauces had the highest percentage of AfT contamination 9/12 (75%), which was followed by traditional snacks 5/11 (45%), peanut butter 4/11 (40%), flour egg coated peanut 6/16 (37%), and peanut cake 3/10 (30%). Fried peanuts and roasted peanut were found to contain aflatoxin at relatively lower percentages of 9% and 8%, respectively. From the 12 analysed baby food samples, on the other hand, no sample was found to be contaminated with aflatoxins. Two of 11 samples (18%) of Corn based Products were contaminated with AfT, ranging between 5.8 and 12.4 μg/kg. Additionally, 30 selected samples in different concentration ranges were further analysed to verify the correlation between ELISA and HPLC techniques and results were compared.

Recovery rate of ZON from spiked Corn samples was between 80% and 128%. The cross-reactivities of the anti-ZON mAb against ZON, β-zearalenol, α-zearalanol, β-zearalanol, deoxynivalenol and aflatoxin B1 were 100%, 24.1%, 189.1%, 43.9%, 50 ) of the anti-ZON mAb was 1.79 ng/g. In total, 137 samples of Corn, powdered Corn, and processed Corn Products were collected from farms and supermarkets in northeastern China. Of 42 Corn samples tested, 4 (9.5%) samples contained ZON 5–60 ng/g, 4 (9.5%) samples contained 60–500 ng/g, In 34 (81%) samples ZON was not detected. Of 15 powered Corn samples, only 1 (6.7%) sample contained 5–60 ng/g, and no ZON was detected in 80 processed Corn Products.
