    Abstract In continuation of a research project aiming at introducing ionic liquids (ILs) as an alternative to the widely applied for the recovery of natural products of industrial interest conventional molecular solvents, we developed a concise procedure for isolation of the biologically active alkaloid S -(+)-glaucine from IL-based aqueous Crude Extract. To this end, a comparative study of the behavior of 1 M [C 4 C 1 im][Ace]-aqueous solution and methanol in a series of consecutive Extractions with the same Extractant was conducted. The results obtained proved the better performance of the IL-based system in the solid–liquid Extraction step, since the latter showed constantly higher Extraction efficiency (ca. 35% enhanced) compared to methanol. The above procedure allows glaucine accumulation from at least ten successive Extractions, while simultaneously reduces the total solid–liquid ratio from 1:40 to 1:7.2, without loss of efficiency. Furthermore, the loss of IL into the matrix pores after Extraction was also considered, suggesting the need for IL recycling by posttreatment of the residual biomass. To recover glaucine from the Crude IL-based aqueous Extract, a series of non-miscible with water molecular solvents were tested. As a result, optimal conditions for quantitative Extraction into chloroform were found, from which, after solvent removal and subsequent crystallization from ethanol, the target compound was isolated as a hydrobromide salt, the latter being the marketed form of glaucine.

    Abstract In this work, development of a new microplate-based high-resolution profiling assay using recombinant human aldose reductase is presented. Used together with high-resolution radical scavenging and high-resolution α-glucosidase assays, it provided the first report of a triple aldose reductase/α-glucosidase/radical scavenging high-resolution inhibition profile – allowing proof of concept with Radix Scutellariae Crude Extract as a polypharmacological herbal drug. The triple bioactivity high-resolution profiles were used to pinpoint bioactive compounds, and subsequent structure elucidation was performed with hyphenated high-performance liquid chromatography–high-resolution mass spectrometry–solid-phase Extraction–nuclear magnetic resonance spectroscopy. The only α-glucosidase inhibitor was baicalein, whereas main aldose reductase inhibitors in the Crude Extract were baicalein and skullcapflavone II, and main radical scavengers were ganhuangemin, viscidulin III, baicalin, oroxylin A 7- O -glucuronide, wogonoside, baicalein, wogonin, and skullcapflavone II.

    Abstract:

    The Crude methanol Extract of Pueraria lobata was investigated by dual high-resolution α-glucosidase inhibition and radical scavenging profiling combined with hyphenated HPLC-HRMS-SPE-NMR. Direct analysis of the Crude Extract without preceding purification was facilitated by combining chromatograms from two analytical-scale HPLC separations of 120 and 600 μg on-column, respectively. High-resolution α-glucosidase and radical scavenging profiles were obtained after microfractionation of the eluate in 96-well microplates. This allowed full bioactivity profiling of individual peaks in the HPLC chromatogram of the Crude methanol Extract. Subsequent HPLC-HRMS-SPE-NMR analysis allowed identification of 21 known compounds in addition to two new compounds, i.e., 3′-methoxydaidzein 8-C-[α-d-apiofuranosyl-(1→6)]-β-d-glucopyranoside and 6″-O-malonyl-3′-methoxydaidzin, as well as an unstable compound tentatively identified as 3′-de-O-methylpuerariafuran.

    Abstract The leaves of Moringa oleifera Lam (Moringaceae) are used by the Indians in their herbal medicine as a hypocholesterolemic agent in obese patients. The scientific basis for their use in hypercholesterolemia was therefore examined. It was found that administration of the Crude leaf Extract of Moringa oleifera along with high-fat diet decreased the high-fat diet-induced increases in serum, liver, and kidney cholesterol levels by 14.35% (115–103.2 mg /100 ml of serum), 6.40% (9.4–8.8 mg/g wet weight) and 11.09% (1.09–0.97 mg/g wet weight) respectively. The effect on the serum cholesterol was statistically significant. No significant effect on serum total protein was observed. However, the Crude Extract increased serum albumin by 15.22% (46–53 g/l). This value was also found to be statistically significant. It was concluded that the leaves of Moringa oleifera have definite hypocholesterolemic activity and that there is valid pharmacological basis for employing them for this purpose in India.
