Sustained speed of kill and repellency of a novel combination of fipronil and permethrin against Ctenocephalides canis flea infestations in dogs

La nouvelle combinaison topique de fipronil et de perméthrine (Frontline Tri-Act®/Frontect®, Merial) a été développée pour lutter contre puces, tiques, moustiques, phlébotomes et mouches piqueuses chez le chien. Etant donné les propriétés répulsives et insecticides de la perméthrine et l’action insecticide du fipronil, l’efficacité de la combinaison contre les puces, dont C. canis , devrait être rapide. L’étude a été conduite afin de mesurer l’efficacité à 1 heure, 6 heures et 24 heures, mais aussi le caractère répulsif de la combinaison fipronil-perméthrine chez les chiens traités par rapport aux chiens non traités. Méthode 12 chiens de race Beagle ont été répartis de manière aléatoire dans l’un des deux groupes sur la base des résultats d’une infestation par les puces avant traitement et durant la phase d’acclimatation. Les chiens du groupe 1 ne sont pas traités alors que les chiens du groupe 2 sont traités une fois à J0. Chaque chien est infesté par 100 puces C. canis adultes à jeun, aux jours 2, 7, 14, 21 et 28. Les chiens sont peignés à la recherche de puces 1 et 6 heures après chaque infestation. Après cet examen, les puces qui restent sur le revêtement de sol de chaque cage sont collectées et comptées. Après les peignages-comptages à 1 heure et 6 heures, toutes les puces vivantes sont replacées sur les chiens. Un dernier comptage par peignage est réalisé pour chaque chien 24 heures après infestation. Résultats Les chiens traités ont eu significativement moins de puces (p ≤ 0,01) que les chiens non traités, à chaque point de comptage. L’efficacité était ≥ 99,1% à 6 h et 24 h après chaque infestation hebdomadaire durant un mois. L’efficacité à 1 heure était également élevée avec 96,5,%, 98,9%, 92,0%, 70,2% et 55,7% aux jours 2, 7, 14, 21 et 28, respectivement. L’efficacité “répulsive”, évaluée sur le revêtement de sol à 1 h, était de 86,5%, 94,9%, 79,5%, 58,4% et 43,9% aux jours 2, 7, 14, 21 et 28, respectivement. Conclusions Cette étude démontre l’effet bénéfique de la combinaison de fipronil et perméthrine contre C. canis , qui apporte une action à la fois insecticide et « répulsive » dès 1 h post infestation, et >99,1% d’efficacité à 6 h pendant un mois. Kaltsatos, Emilie Bouhsira, Michel FrancAbstract:This study was conducted to evaluate the efficacy of a new topical ectoparasiticidal spot-on containing 4.95 % dinotefuran (w/w), 36.08 % permethrin (w/w) and 0.44 % pyriproxyfen (w/w) (Vectra 3D™, Ceva, Libourne, France) against Portuguese strain of Phlebotomus perniciosus and a French strain of Ctenocephalides canis in dogs. Twelve beagle dogs were exposed for 1 h to 100 P. perniciosus on day 6 for allocation in two groups. One group was treated on day 0, and the other group was the control group. The dogs were exposed for 1 h to 100 P. perniciosus on days 1, 7, 14, 21 and 28. After each sandfly challenge, the same dogs were infested with 100 C. canis. Counts of living fleas were determined 48 h after infestation on days 4, 3, 9, 16, 23 and 30. For sandflies, the anti-feeding effect was 96.9, 99.7, 98.7, 83.5 and 87.0 % on days 1, 7, 14, 21 and 28, respectively. The mortality effect was 97.8, 99.8, 73.7, 27.5 and 39.6 % on days 1, 7, 14, 21 and 28, respectively. At each challenge point, the mortality and anti-feeding effects on sandflies were significantly different between the control and treatment groups (p < 0.05). The adulticidal effect on C. canis remained above 99 % throughout the study period. The results indicate that a combination with dinotefuran, permethrin and pyriproxyfen may be used as an effective part of an overall flea and sandfly control strategy in dogs for monthly use.

Evaluation of speed and duration of efficacy of spinosad tablets for treatment and control of Ctenocephalides canis (siphonaptera: pulicidae) infestations in dogs
Parasite, 2009Co-Authors: Michel Franc, Emilie BouhsiraAbstract:A controlled clinical trial was performed to determine the duration of efficacy of a new oral insecticide formulation of spinosad for the control of experimentally induced Ctenocephalides canis infestations in dogs. Twelve Beagle dogs (two groups of six) were used in the study. Dogs in the treated group received spinosad tablets per os on D0 at the commercial dosage. All dogs were infested with 100 fleas on Days – 7, – 1, 7, 14, 21, 28 and 35. The dogs were combed four hours after each infestation and fleas were counted and replaced on the coat. 24 hours after each infestation fleas were combed, counted and removed. The efficacy of the formulation was calculated four and 24 hours after the treatment and then four and 24 hours after each new infestation. The mean number of fleas on the control dogs was respectively between 65.1 and 83.3 at four hour counts and between 58.3 and 75.3 at 24 hour counts. The product was well tolerated. The treatment controlled the fleas already present on the skin with 81 % efficacy at four hours and 100 % efficacy at 24 hours. For the weekly infestations, the speed of action of the product was high: at four hours the efficacy was 100 % at D7, 96 % at D14, 74 % at D21, 42 % at D28, 12.90 % at D35 and 12.8 % at D42. The efficacy evaluated 24 hours after each infestation was approximately 100 % during three weeks then 90 % at D39, 81.4 % at D36 and 80.4 % at D43. A single dose of the new spinosad tablet formulation should control flea populations in dogs for four weeks as indicated in the claim (evaluation performed at 48 h for the registration). Spinosad tablet is the first product administered per os which acts so long and so quickly against adult fleas.

First blood meal of Ctenocephalides canis (Siphonaptera: Pulicidae) on dogs: time to initiation of feeding and duration.

Mallet, Michel FrancAbstract:Two experiments were conducted on dogs to evaluate interval to initiation and duration of the first blood meal of Ctenocephalides canis (Curtis). Percentage of fed male and female fleas was calculated for fleas held on dogs for 5, 15, 30, 60 min, 6, and 24 hr. Duration of first blood meal was also measured for individual fleas confined on dogs. When fleas were free in the hair coat, 21.2% had begun blood feeding within 5 min. After 1 hr, 72.5% of fleas had fed. After 6 hr, 95.2% of males and 100% of females had taken a blood meal, and 24 hr after deposition all fleas had fed. There was no significant difference between the 2 sexes. The mean delay between deposition and biting for fleas that began feeding within 15 min was 2 min 52 sec ± 3 min 2 sec for female fleas and 3 min 8 sec ± 2 min 45 sec for males. The mean duration of female and male meals was 5 min 3 sec ± 3 min 41 sec and 6 min 9 sec ± 6 min 8 sec, respectively. There was no significant difference between the 2 sexes. FrancAbstract:Abstract A novel spot-on formulation combining fipronil, amitraz and (S)-methoprene (CERTIFECT™, Merial Limited, GA, USA) was evaluated in adult Beagle dogs in a study to determine its adulticidal efficacy against the dog flea ( Ctenocephalides canis , Curtis, 1826). Sixteen dogs were randomly allocated to treatment groups: 8 dogs served as untreated controls, and 8 dogs were treated once. Treatment consisted of applying a new combination formulation to deliver at least 6.7 mg fipronil/kg body weight (bw), 8.0 mg amitraz/kg bw, and 6.0 mg (S)-methoprene/kg bw. The combination was designed to enhance the efficacy against ticks of the original fipronil/(S)-methoprene combination. Each dog was infested with 100 adult unfed dog fleas within 24 h prior to treatment and then at weekly intervals for 8 weeks after treatment. At 24 h after treatment or after each subsequent infestation, each dog was combed thoroughly to remove live fleas to be counted. Prevalence of Rickettsia felis in Ctenocephalides felis and Ctenocephalides canis from Uruguay.

OteoAbstract:Our aim was to determine the presence of Rickettsia spp. in 66 fleas from Uruguay. Rickettsial DNA was amplified using gltA and ompB PCR primers. Rickettsia spp. were found in 41% of the fleas (25 Ctenocephalides felis and 2 Ctenocephlides canis). Sequences resulted in the identification of Rickettsia felis and four genotypes closely related to this species (Rickettsia sp. TwKM03, California 2, Hf187, and RF2125). The presence of R. felis in fleas from Uruguay in was demonstrated. This is the second species of Rickettsia identified in Uruguay in the past 2 years using molecular approaches, and it is helping to clarify the etiology of rickettsial diseases in the region.

A total of 88 specimens were tested by polymerase chain reaction (PCR) using gltA and ompB genes as targets for Rickettsia spp., and 16S rRNA and ribC genes for Bartonella spp. Rickettsia felis-like (28.4%), Bartonella clarridgeiae (6.8%), and Bartonella henselae (3.4%) were detected in Ctenocephalides spp. Other Bartonella sp. different from B. clarridgeiae and B. henselae could also be present in fleas from La Rioja.

Identification of flea species using MALDI-TOF/MS

We measured the MS spectra from 38 flea specimens of 5 species including Ctenocephalides felis, Ctenocephalides canis, Archaeopsylla erinacei, Xenop- sylla cheopis and Stenoponia tripectinata. A blind test performed with 24 specimens from species included in a library spectral database confirmed that MALDI-TOF/MS is an effective tool for discriminating flea species. Although fresh and 70% ethanol-conserved samples sub- jected to MALDI-TOF/MS in blind tests were correctly classified, only MS spectra of quality from fresh specimens were sufficient for accurate and significant identification. A clus- ter analysis highlighted that the MALDI Biotyper can be used for studying the phylogeny of fleas.

Murine typhus, a typhus group rickettsiosis caused by Rickettsia typhi and transmitted by X. cheopis, is also known to occur in Algeria, but cases are poorly documented [2,3]. However, recently, fleas collected in Algeria, in the district of Oran, between July and September 2003 (Ctenocephalides canis from rodents, and Archeopsylla erinacei from hedgehogs) were tested by PCR for the presence of Rickettsia spp., and were shown to harbour an emerging pathogen, Rickettsia felis [4]. In this work, in an effort to identify the possible aetiological agents and vectors for rickettsiosis affecting humans in Algeria, we analysed more fleas collected from rodents trapped in peridomestic areas for evidence of rickettsial infection.

All fleas were tested by PCR to detect DNA of bacteria of the genera Rickettsia (gltA and ompB genes) and Bartonella (ITS and ftsZ genes). Sequencing of PCR-amplified products was done using gltA fragments for Rickettsia and ftsZ fragments for Bartonella. Two genotypes related to Rickettsia felis were identified in three Ctenocephalides canis and one C. felis specimen. Further, the following Bartonella spp. were detected: Bartonella henselae in two C. felis specimens; Bartonella clarridgeiae in three C. felis specimens; and a new Bartonella genotype in one Nosopsylla fasciatus specimen. Rickettsia and Bartonella may be frequently detected in fleas infesting peridomestic animals from the western border of Thailand.

Journal of Medical Entomology, 2001Co-Authors: Carlos Cruzvazquez, Eduardo Castro Gamez, Magdalena Parada Fernandez, Miguel Ramos ParraAbstract:Abstract The seasonal occurrence of Ctenocephalides felis felis (Bouche) and Ctenocephalides canis (Curtis) infestation on dogs and cats in Cuernavaca City in Mexico, was determined by examining 1,803 dogs and 517 cats at two veterinary clinics during 1995–1997. The overall flea infestation was 30.3 and 30.1% for dogs and cats, respectively. There were no significant differences (P > 0.05) in percentage of infestation among years for both hosts. The infestation was somewhat higher in spring, summer, and autumn than in winter, but no statistical differences was found among seasons (P > 0.05) for both pets. No relationship existed between percentage of flea infestation and temperature or rainfall among seasons. On dogs, 81.1% were infested with only C. felis felis, 16.8% with C. canis, and 2% had both flea species; whereas 92.3% of the cats were infested with C. felis felis and 7.7% with C. felis felis and C. canis.
