    Abstract:

    Study design Randomised, controlled, single-blind, three-arm parallel-group trial set in general dental practice with a single general dental practitioner operator/assessor. Intervention Seventy-five adult patients, with basic periodontal examination scores of 0 in all sextants, good oral hygiene, at least one sensitive tooth (not diagnosed as pulpitis) and willing to comply with the trial regime were entered into the trial and randomised. Seventy-two participants completed the study. The three interventions were; non-desensitising toothpaste (Colgate Cavity Protection Regular, Colgate-Palmolive, USA), desensitising toothpaste (Colgate Sensitive Fresh Stripe, Colgate-Palmolive, USA) and Dentine Bonding agent (Seal and Protect, Denpsly, USA). The non-desensitising toothpaste and desensitising toothpastes were provided to the subjects for use at home but Dentine Bonding agent was applied in the surgery. Outcome measure Dentinal hypersensitivity was measured using a participant completed Visual Analogue Scale (VAS) at baseline, two weeks, three months and six months. At baseline and six months a standardised air blast to the buccal cervical root stimulus was used with the VAS. At two weeks and at three months participants self-completed the VAS at home with no stimulus. To determine whether glutathione (GSH) levels were important in the cytotoxicity of Dentine Bonding Agents, cells were pretreated with 2-oxothiazolidine-4-carboxylic acid (OTZ) to boost GSH levels or buthionine sulfoximine (BSO) to deplete GSH. Three replicates of each Dentine Bonding Agents were performed in each test. All assays were repeated three times to ensure reproducibility. Statistical analysis was by one-way analysis of variance (anova). Tests of differences of the treatments were analysed by Duncan’s test. Results  Clearfil SE Bond, PB, and SB were cytotoxic to pulp cells in a concentration-dependent manner (P   SB > CB. Addition of OTZ extracellularly protected the pulp cells from Dentine Bonding Agents–induced cytotoxicity (P < 0.05). Addition of BSO enhanced pulp cell death on Dentine Bonding Agents–induced cytotoxicity (P < 0.05). Conclusions  Dentine Bonding Agents have significant potential for pulpal toxicity. GSH depletion could be the mechanism for Dentine Bonding Agents–induced cytotoxicity.

    Abstract:

    AIM To investigate the cytotoxicity of five different Dentine-Bonding Agents on human pulp cells in vitro. METHODOLOGY Set specimens from Clearfil SE Bond (CB), Heliobond (HB), Prime & Bond NT (PB), Single Bond (SB), and Syntac Single Component (SC) were eluted with culture medium for 2 and 5 days. Cytotoxicity was judged using tetrazolium bromide reduction assay on human primary pulp cells. RESULTS Elutes from five Dentine-Bonding Agents were cytotoxic to primary human pulp cells (P PB > SC > HB > CB. CONCLUSIONS The influence of the cytotoxicity depended on the materials tested. Dentine-Bonding Agents have significant potential for pulpal toxicity.

    Abstract:

    AIM: To evaluate the cytotoxic effect of a new Dentine-Bonding Agents. METHODS: Using agar overlay test and tetrazolium-based colorimetric assay(MTT) in vitro,the cytotoxicity of three Dentine-Bonding Agents,U Bond(UB),Adper Prompt Self-etching Adhesive(AP),Prime Bond NT(PB),to L929 mouse fibroblast were evaluated.The test materials were applied on the Dentine discs derived from human third molars in the same way as in the clinical procedures recommented by each manufacturer. RESULTS: Cytotoxicity of the three Dentine-Bonding Agents was 1 grade according to MTT assay.In agar overlay test,parts of decolored zones and some dissolved cells were observed under all test Dentine discs. CONCLUSION: The new adhesive had slight cytotoxicity.Further study on the influence of the new dentin Bonding Agents towards pulp tissue must be taken.
