It is also used for cosmetological skin treatment. PDT is generally considered to have a low risk of carcinogenicity. However, instances of nonmalignant human tumors turning malignant have been linked to PDT. In this study, we used 5-aminolevulinic (ALA) Acid and 3 water soluble photosensitizers-PP(Arg)2, PP(Ser)2Arg2, PP(Ala)2Arg2, all Diamino Acid derivatives of protoporphyrin IX-to treat benign papillomas in FVB/N mice induced by 7,12-dimethylbenz(a)anthracene (DMBA)-12-O-tetradecanoyl-phorbol-13-acetate (TPA). Of these drugs, ALA and PP(Arg)2 were found the most efficient. PDT reduced the number of papillomas, but with increasing effectiveness of the drugs, the risk of malignant transformation of the papillomas into squamous cell carcinomas increased. The underlying mechanisms are not clear and further investigations are needed. © 2009 UICC

Diamino Acid derivatives of PpIX as potential photosensitizers for photodynamic therapy of squamous cell carcinoma and prostate cancer: In vitro studies

The search for new compounds which might become effective PSs is the major direction for PDT development. In the present work we have studied the dark toxicity, intracellular localization and photodynamic properties of four potential, water soluble, second generation PSs – PP(Arg)2, PP(Ser)2Arg2, PP(Ala)2Arg2, PP(Phe)2Arg2, all Diamino Acid derivatives of protoporphyrin IX. Human prostate cancer (DU-145) and squamous carcinoma (A431) cells were used as experimental model. Among investigated compounds PP(Ser)2Arg2 exhibited the lowest dark toxicity and the highest PDT effectiveness towards both cell lines. Fluorescence microscopy revealed the time-dependent changes in intracellular localization of the PS which were related to the phototoxicity. The results show that PP(Ser)2Arg2 may be a potential PS for PDT.

Interactions of peripheral-type benzodiazepine receptor with Diamino Acid derivatives of protoporphyrin IX.

B Biology, 2008Co-Authors: Aneta Bombalska, Alfreda GraczykAbstract:Abstract The aim of this study was to examine and determine binding affinities of the peripheral benzodiazepine receptor (PBR) with Diamino Acid derivatives of protoporphyrin IX with general structure PP(AA)2 in a function of environmental pH changes. Fluorescence intensity was used as a measure of protein association constant. Decrease in protein fluorescence, after titration with increasing concentration of porphyrins, gave evidence of interactions between PBR and examined ligands. Experimental data were fitted to theoretical curves, assuming different models of interactions. All examined ligands were best fitted to weak binding model. Increase in environmental Acidity resulted in changes in association constants. For all examined derivatives association constants were at least twice higher in pH 5.5 as compared to pH 7.4 and 6.5.

Influence of Diamino Acid derivatives of protoporphyrin IX on mouse immunological system: preliminary results.

Diamino Acid derivatives of protoporphyrin IX are promising photosensitizing agents that are intended to be the components of new anti-tumor drug. The influence of three derivatives – PP(Ser)2Arg2, PP(Ala)2Arg2, PP(Phe)2Arg2 and a mixture of these compounds called Sensyphyrine® on mouse immunological system was investigated where animals were exposed and unexposed to the laser irradiation. Porphyrins solutions were injected intravenously, mice were irradiated with the red diode laser at λ = 632 nm. Cells and blood samples were taken at time intervals after irradiation. The levels of interleukin-6, interleukin-1β and the production of reactive forms of nitrogen by macrophages were determined. The results show that all of the Diamino Acid derivatives of protoporphyrin IX indicate an immunological response when the light is applied. Each of the porphyrin revealed different impact on mice immunological system. The most potent stimulant properties disclosed PP(Phe)2Arg2 derivative for which the highest values of IL-1β, IL-6 and NO 2 - were noticed. The weakest immunological activation revealed PP(Ser)2Arg2 derivative.

The water-soluble hematoporphyrin derivatives diarginate (HpD[Arg]2) and 1-arginin di(N-amino Acid)-protoporphyrinate used in this study are effective photosensitizers in tumor photodynamic therapy. Depending on the amino Acid substituent, the porphyrin derivatives differ in their affinity for yeast cells. It is shown that HpD(Arg)2 and PP(Met)2 (Arg)2 penetrate into the yeast cell and are metabolized. Both compounds sensitize yeast cells to photodamage but have no mutagenic effect on nuclear or mitochondrial genomes.

Luethyella okanaganae gen. nov., sp. nov., a Novel Genus and Species of the Family Microbacteriaceae Isolated from the Insect Okanagana rimosa

Phylogenetic analysis employing 16S rRNA gene sequences demonstrate that “Corynebacterium okanaganae” is not a member of the genus Corynebacterium but related to members of the Microbacteriaceae being most closely related to, but distinct from, members of the genera Rathayibacter, Mycetocola and Curtobacterium . The bacterium is an aerobic, Gram-positive staining, rod-shaped actinobacterium with the cell-wall peptidoglycan based on 2,4, Diaminobutyric Acid as the diagnostic Diamino Acid. The predominant menaquinones are MK-10, MK-11 and MK-12, and the principle polar lipids are phosphatidylglycerol and diphosphatidylglycerol. The major fatty Acids consist of anteiso-C_15:0 and anteiso-C_17:0. Therefore, based upon the phylogenetic, biochemical, and chemotaxonomic information, the organism merits recognition as a novel species and genus in the family Microbacteriaceae , for which the name Luethyella okanaganae gen. nov. sp. nov. is proposed. The type strain is LBG B4405^T = CCUG 43304^T = NCIMB 702272^T.

microbacterium lindanitolerans sp nov isolated from hexachlorocyclohexane contaminated soil

Strain MNA2T showed 95 % 16S rRNA gene sequence similarity with Microbacterium halotolerans YIM 70130T. Phylogenetic analysis of 16S rRNA gene sequences showed that strain MNA2T belonged to a clade represented by the genus Microbacterium of the family Microbacteriaceae. Strain MNA2T contained anteiso-C15 : 0, anteiso-C17 : 0, iso-C16 : 0 and iso-C15 : 0 as the predominant fatty Acids and diphosphatidylglycerol, phosphatidylglycerol and two unknown polar lipids. The menaquinones were MK-12, MK-11, MK-13 and MK-10, in an approximate molar ratio of 45 : 40 : 13 : 3, respectively. The DNA G+C content was 65.3 mol%. The peptidoglycan was of the B type of cross-linkage with ornithine as the diagnostic Diamino Acid. The results of the phylogenetic, phenotypic and chemotaxonomic analysis indicate that strain MNA2T belongs to a previously unrecognized species of the genus Microbacterium, for which the name Microbacterium lindanitolerans sp. nov. is proposed. The type strain is MNA2T (=DSM 22422T =CCM 7585T).

georgenia ruanii sp nov a novel actinobacterium isolated from forest soil in yunnan china and emended description of the genus georgenia

The isolate contained chemotaxonomic markers that corresponded to those of its phylogenetic neighbour, Georgenia muralis, i.e. it possessed peptidoglycan type A4α with lysine as the diagnostic cell-wall Diamino Acid, the predominant menaquinone was MK-8(H4) and the major fatty Acid was ai-C15 : 0. The G+C content of the genomic DNA was 72.9 mol%. Strain YIM 004T exhibited a 16S rRNA gene sequence similarity of 97.3 % and a DNA–DNA relatedness value of 18 % with respect to G. muralis DSM 14418T. On the basis of the phenotypic and genotypic differences between the isolate and G. muralis, strain YIM 004T represents a novel species of the genus Georgenia, for which the name Georgenia ruanii sp. nov. is proposed. The type strain is YIM 004T (=CCTCC AB 204065T=DSM 17458T=KCTC 19029T). In addition, an emended description of the genus Georgenia is presented.

International Journal of Systematic and Evolutionary Microbiology, 2005Co-Authors: Axel Schippers, Klaus Bosecker, Cathrin Spröer, Peter SchumannAbstract:A taxonomic study of two crude-oil-degrading, Gram-positive bacterial strains, designated BAS69T and BNP48T, revealed that they represent two novel Microbacterium species. 16S rRNA gene sequence similarity to their closest phylogenetic neighbours was 98·5 % for BAS69T (Microbacterium paraoxydans DSM 15019T and Microbacterium saperdae DSM 20169T) and 99 % for BNP48T (Microbacterium luteolum DSM 20143T). Levels of DNA–DNA relatedness to the closest phylogenetic neighbours of both strains were between 11 and 38 %. According to phylogenetic analysis, the two strains are distinguishable from all recognized species of Microbacterium. Morphological and physiological characteristics of strains BAS69T and BNP48T were different from those of phylogenetically closely related Microbacterium species. The Diamino Acid in the cell-wall peptidoglycan of BAS69T is lysine and of BNP48T is ornithine. The major menaquinones are MK-11 and MK-12 for both strains. Based on their ability to degrade crude oil, the name Microbacterium oleivorans sp. nov. is proposed for strain BAS69T (=DSM 16091T=NCIMB 14003T) and Microbacterium hydrocarbonoxydans is proposed for strain BNP48T (=DSM 16089T=NCIMB 14002T).

Kroppenstedt, Erko StackebrandtAbstract:A gram-positive, aerobic actinomycete was isolated from soil. Spherical cells of this organism occur singly or form packets, which may cluster. The diagnostic Diamino Acid of the cell wall peptidoglycan is LL-Diaminopimelic Acid. The predominate menaquinone is MK-8 (H4), and the main fatty Acids are 13-methyl tetradecanoic Acid and 12-methyl tetradecanoic Acid. The diagnostic polar lipids are phosphatidylethanolamine and phosphatidylinositol. The DNA base composition is 73 mol% G+C. Comparison of 16S ribosomal DNA sequences showed that this isolate is a phylogenetic neighbor of Terrabacter tumescens and Intrasporangium calvum. Genotypic, chemotaxonomic, morphological, and physiological characteristics are used to describe a new genus and species, Terracoccus luteus gen. nov., sp. nov. The type strain is strain IMET 7848 (= DSM 44267).
