    Objective : To assess the Diuretic potential of water extract (WE) of leaves of Pongamia pinnata L. Materials and Methods : Different doses (500, 1000, 1500, and 3000 mg/kg) of WE of the leaves of P. pinnata or vehicle or furosemide, reference drug was orally administered to hydrated rats ( n = 6/group) and their cumulative urine output was monitored at hourly intervals for 6 h. Result : The WE of P. pinnata does not possess significant ( P > 0.05) Diuretic Activity. Conclusion : The WE of the leaves of P. pinnata did not possess Diuretic Activity in rats as was claimed in traditional and folkloric medicine.

    Objective: To assess the Diuretic potential of hot water extract (HWE) of a whole plant of Amaranthus viridis L. Materials and Methods: Different doses (25, 50, and 100 mg / kg) of HWE of the whole plant of A. viridis or vehicle or furosemide, reference drug, was orally administered to hydrated rats (n = six / group) and their cumulative urine output was monitored at hourly intervals for five hours. Result: The HWE of A.viridis does not possess significant ( P > 0.05) Diuretic Activity. Conclusion : The HWE of the whole plant of A. viridis did not possess Diuretic Activity in rats, as was claimed in traditional and folkloric medicine.

  • Oral Diuretic Activity of hot water infusion of Sri Lankan black tea (Camellia sinensis L.) in rats

    Abstract:

    Background: Black tea [Camellia sinensis (L.) O. Kuntze (family: Theaceae)] has been used by Sri Lankan traditional practitioners to promote diuresis. However, the type and grade of tea is not specified. Materials and Methods: This study investigates the Diuretic Activity of black tea infusion (BTI) in rats using Broken Orange Pekoe Fannings (BOPF) grade from major agroclimatic elevations: high-, mid-, and low-grown. Different concentrations of BTI, furosemide (positive control), and water (vehicle) were orally administered to starved (18 h) male rats (n = 9/group), then hydrated. Acute and chronic (28 days) Diuretic activities were assessed by measuring cumulative urine output at hourly intervals for 6 h. Electrolyte levels (Na + , K + , Ca 2+ , H + , Cl− , HCO3− ), pH, osmolarity of urine, and glomerular filtration rate (GFR) of treated rats were determined. Results: Administration of BTI induced a significant (P < 0.05) and dose-dependent Diuretic Activity, which varied with the tea produced in different agroclimatic elevations. Diuretic Activity had a rapid onset (1 st h), peaked at 2 nd h and maintained up to 4 th h (except the low dose). Furthermore, there was a dose-dependent increase in micturition frequency, which peaked at 2 nd h. A close association between the caffeine content of tea and Diuretic Activity was evident. BTI-induced diuresis was accompanied with an increased urine Na + level and GFR. The Diuretic Activity of BTI was mediated via multiple mechanisms: inhibition of both aldosterone secretion (with increased Na + /K + ratio) and carbonic anhydrase [with decreased Cl− /(Na + + K + ) ratio] and via thiazide type of Diuretic action (evaluated with increased Na + /Cl− ratio). Conclusion: The Sri Lankan BOPF grade black tea possesses mild oral Diuretic Activity whose efficacy differs with the agroclimatic elevation of production. Furthermore, it supports the traditional claim that the black tea acts as a Diuretic.

  • Diuretic Activity of sri lankan black tea camellia sinensis l in rats

    Abstract:

    The aim of this study was to evaluate the Diuretic potential of Sri Lankan black tea (Camellia sinensis L.). This was assessed in rats using high grown Dust grade No: 1 tea, which is consumed widely by the tea drinkers worldwide. Different doses of hot black tea brew (BTB) (84, 167, 501 or 1336 mg/ml respectively equivalent to 1.5, 3, 9 and 24 cups) were made and orally administered to previously starved (24 h) but subsequently hydrated (with 15 ml of isotonic saline) rats and their urinary output was monitored cumulatively at hourly intervals for 6h. The reference drug used was frusemide (13 mg/kg). The results showed that BTB induced significant (P < 0.05), mild to moderate and dose- dependent diuresis (starting from 167 mg/ml). This Diuretic Activity had a fairly rapid onset (within 2 h) and relatively short duration of action (3 h). BTB also significantly (P < 0.05) increased the overall urinary frequency. Further, the Diuretic Activity of BTB was less potent to frusemide (by 45%). Decaffeination of black tea almost completely abolished the diuresis. The diuresis of the BTB was solely due to increased (by 55 %) urinary Na + excretion (with no urinary K + loss). Further, the chronic daily administration of the BTB did not develop tolerance or induce toxicity (general, renal and hepatic). It is concluded that BTB made from Sri Lankan high grown Dust grade No :1 tea has safe, mild to moderate Diuretic Activity with rapid onset and relatively short duration of action. Further, this study supports the claim made by Sri Lankan indigenous physicians that it is a Diuretic.

    A series of 1,6-dihydropyrimidine-2-amine derivatives and 1,6-dihydropyrimidine-2-thiol derivatives were synthesized by the reaction of substituted 1,3-diphenylprop-2-en-1-one (chalcones) with guanidine hydrochloride and thiourea, respectively. All the synthesized compounds were in good agreement with elemental and spectral data. The synthesized compounds were screened in vivo for Diuretic Activity. The four compounds 2d, 2e, 3d and 3e, which showed moderate to good Diuretic Activity, were evaluated for their toxicity studies and none of the compounds showed any toxicity of the liver as compared with control. However, compounds 3e and 3d showed Diuretic properties more than that of standard (acetazolamide) and were long acting. Overall, compound 3e, 6-(2,6-dichlorophenyl)-4-(pyridin-2-yl)-1,6-dihydropyrimidine-2-thiol, was found to be the most promising candidate of the series.

    Abstract Ethnopharmacological relevance Smilax canariensis is an endemic species of the Canary Islands, popularly known as “Zarzaparrilla sin espinas”. The rhizome, leaves and stem of this species has wide use in folk medicine practice on the islands, where they are habitually employed as Diuretic. Aim of the study In this paper we report on the Diuretic Activity in experimental animals of several fractions of the methanol extract of this species. Materials and methods Four fractions of the methanol extract of the rhizomes, leaves and stem of Smilax canariensis (50 and 100 mg/kg), Furosemide and Hydrochlorotiazide (10 mg/kg), were orally administered to laboratory animals to evaluate their Diuretic Activity. Water excretion rate, pH, conductivity, and content of Na+, K+ and Cl− were measured in the urine of saline-loaded mice. Results All the studied extracts showed an interesting increase of the diuresis, although the n-buthanol (27%; p  Conclusions These data, together with previous results on the aqueous and methanol extracts, reaffirm assertions made regarding the effectiveness of the extracts of this plant against urinary pathologies in the Canary Islands folk medicine.

    Abstract Aim of the study Withania aristata is an endemic plant used traditionally in Canary Islands as a Diuretic. In this paper, we report on this pharmacological Activity in several extracts of the dry vegetal material collected and the identification and Diuretic Activity of two withanolides, one of them previously not reported, isolated from the most active fraction. Material and methods Four Whitania aristata extracts at 100 mg/kg were orally administered to laboratory animals to evaluate their Diuretic Activity. From the most active fraction, two withanolides were isolated. Both and a mixture of them at 5 and 10 mg/kg were analyzed too as Diuretics. Water excretion rate and content of Na+ and K+ electrolytes were measured in the urine of saline-loaded animals. Results Whitania aristata water fraction, the two withanolides and the mixture of these compounds displayed high Diuretic Activity, with a significant excretion of sodium and potassium ions in laboratory animals. Conclusions This research supports the ethno-medicinal use of Whitania aristata as Diuretic. This Activity seems to be associated to the presence of a new type of natural Diuretic agents, such as withaferin A and witharistatin.

    Abstract A pharmacological evaluation for Diuretic Activity of infusions at 5, 10 and 15% of Artemisia thuscula Cav. was carried out. Urinary excretion of water, pH, density, conductivity and Na + , K + and Cl − content were investigated in saline-loaded rats. The infusions showed a dose-dependent decrease Diuretic effect, but augmented significantly with respect to the control group for the urinary excretion of water and sodium. Furthermore, a potassium-sparing effect at 5 and 10% was showed. The Diuretic effect does not seem to be related to the potassium content of the starting material. The results justify the use of Artemisia thuscula as Diuretic agent by the canary traditional medicine.
