This study was aimed at evaluating the antiasthmatic, antitussive, and expectorant activities of the water extracts from the three plants in vivo and analyzing their chemical components by HPLC-DAD. At the medium and high doses, the water extracts of three Elaeagnus leaves significantly prolonged the preconvulsive time (P < 0.01) in guinea pigs, lengthened the latent period of cough (P < 0.01) and decreased the cough frequency caused by aqueous ammonia in mice (P < 0.01), and enhanced tracheal phenol red output in mice (P < 0.01). There were no significant differences in the pharmacological actions between the three Elaeagnus leaves. Moreover, there was more similarity on overlap peaks in the range of retention time from 10 to 40 min by HPLC and many peaks that belonged to flavonoids compounds. It suggested that the main constituents of the three Elaeagnus leaves were flavonoid for the pharmacological activities. These effects were the important evidence for the traditional use of E. henryi leaf and E. lanceolata leaf as well as E. pungens to treat asthma and chronic bronchitis.

(Family Elaeagnaceae) has been documented as an effective herb for the treatment of asthma and chronic bronchitis in traditional Chinese medicine. In the past years, only a few of preliminary studies reported the chemical constituents and pharmacology effects of the herb, but their action on the tracheal relaxation has not been investigated. Aim of the study To investigate the relaxing effect and mechanism of the extracts from Elaeagnus pungens leaves on guinea pig tracheal smooth muscle and bronchi smooth muscle cells. Materials and methods Four fractions of different polarities from Elaeagnus pungens leaves were tested to the tracheal strips on the resting tension or pre-contracted by histamine (20 μM) and acetylcholine (20 μM). Inhibitory effects of the 1-butanol fraction (400 mg/ml) on cumulative histamine and acetylcholine (0.2–20 μM) induced contraction were measured. In order to determine the mediators on the 1-butanol fraction effect, the relaxing effect of the 1-butanol fraction was evaluated in the absence and presence of β-adrenoceptor antagonists (1 μM propranolol), K + channels-blockers (4-aminopyridine (2 mM), tetraethylammonium chloride (5 mM) or glibenclamide (10 μM)), the cyclooxygenase inhibitor (indomethacin, 10 μM), nitric oxide synthase inhibitor (Nω-nitro- l -arginine methyl ester, 100 μM) or l -type Ca 2+ channel inhibitor (nifedipine, 1 μM). Moreover, [Ca 2+ ] i in bronchi smooth muscle cells was analyzed by measuring the fluorescence intensity with confocal system. Results 1-Butanol fraction induced the highest relaxant effect among four fractions of different polarities from Elaeagnus pungens leaves, and significantly relaxed the tracheal strip in the concentration-dependent manner on the resting tension and pre-contracted by histamine phosphate and acetylcholine. It also produced an unparallel rightward shift of the cumulative concentration-response curve of histamine or acetylcholine. Furthermore, the relaxant effect of 1-butanol fraction was not affected by propranolol, glibenclamide, tetraethylammonium chloride, 4-aminopyridine, indomethacin and Nω-nitro- l -arginine methyl ester. However, 1-butanol fraction-induced relaxation decreased after adding nifedipine. It also concentration-dependently inhibited CaCl 2 -induced contraction in the Ca 2+ -free, 60 mM K + -containing solution. Additionally, [Ca 2+ ] i in the BSMCs significantly reduced after administration of the 1-butanol fraction. Conclusions The 1-butanol fraction from Elaeagnus pungens leaves resulted in a relaxation in the non-precontracted and pre-contracted tracheal strips. The relaxant effect was not related to K + channels, NO, cGMP or β-adrenoceptors, but related to the inhibition of Ca 2+ influx through l -type Ca 2+ channels.

Study on Annul Growth Dynamics of Elaeagnus pungens Thunb seedlings
Journal of Anhui Agricultural Sciences, 2020Co-Authors: Peng ChengAbstract:[Objective] The research aimed to study the annul growth rules of seedlings height and ground diameter of 1-year-old seedlings of Elaeagnus pungens Thunb.[Method] The regular observation with seedling height and ground diameter of Elaeagnus pungens Thunb has been carried out.The growth process of seedling height and ground diameter was simulated by the Logistic equation of mathematic model.[Result] The growing period of Elaeagnus pungens Thunb Seedlings has been divided into four stages:seedling stage(before April 23),preliminary stage of seedling growth(from April 24 to May 23),prosperous stage of seedling growth(from May 23 to September 23)and later stage of seedling growth(after September 24).The growth rules of seedling height and ground diameter were in accordance with the growth curve of "S" type.The actual value was mainly coincident with the model predicted value.The growth of seedling height and ground diameter can be predicted by the Logistic equation.[Conclusion] The finding provides the theoretical basis for the cultivation,introduction domestication and management of Elaeagnus pungens Thunb.

Determination of Total Polyphenols in Different Parts of Elaeagnus angustifolia Fruit Growing in Xinjiang
Food Science, 2020Co-Authors: Hu Junping, Yang Jianhua, Wang Xinling, Muaitaer Nuermaimaiti, Rena KasimuAbstract:Gallic acid was used as a standard substance to determine total polyphenol contents in different parts of Elaeagnus angustifolia fruit growing in Xinjiang Autonomous Region by Folin-Ciocalteu method.The calibration curve of gallic acid presented good linearity within the concentration range of 2-12 μg/mL(r = 0.9991).The average of three replicates of spike recovery for total polyphenols at three levels was 98.80%,with a relative standard deviation of 1.81%.Elaeagnus angustifolia seeds exhibited significantly higher total polyphenol content than Elaeagnus angustifolia flesh,with a 3-to 5-fold increase.The method demonstrates the advantages of convenience,rapidity and accuracy,thereby providing an effective approach for the quality control of Elaeagnus angustifolia fruit.

Extraction and Purification of Total Polyphenols from Elaeagnus angustifolia Seeds
Food Science, 2020Co-Authors: Rena KasimuAbstract:Objective:To study the respective processes for the extraction and purification of total polyphenols from Elaeagnus angustifolia seeds.Methods:Mixtures of methanol and water at different volume ratios were used to extract total polyphenols from Elaeagnus angustifolia seeds,and 40% aqueous methanol solution was selected.Reflux extraction presented higher both yield and purity of total polyphenol extract when compared to ultrasonic extraction and shaking extraction.Subsequently,orthogonal array design method was used to optimize the process for the reflux extraction of total polyphenols using 40% aqueous methanol solution.In the final section of this study,the purification of the crude total polyphenol extract from Elaeagnus angustifolia seeds was investigated by macroporous resin adsorption or ionic precipitation.Results:The optimal extraction process of total polyphenols from Elaeagnus angustifolia seeds was based on the use of 30-fold volume of 40% aqueous methanol solution to extract the material three times for 1 h each time,followed by vacuum condensation.Macroporous resin adsorption was found more suitable for the purification of the crude total polyphenol extract from Elaeagnus angustifolia seeds than ionic precipitation.An optimal purification was achieved through the steps of adsorption of the target products onto AB-8 resin,washing with distilled water until no saccharides could be detected in the eluate,elution with 2-fold bed volume of 40% aqueous methanol solution,harvesting of eluate fractions,condensation and drying.The preparation of total polyphenols from Elaeagnus angustifolia seeds was performed in triplicate,and an average value of product purity of 52.17% was obtained.Conclusion:The preparation method integrating the above optimized extraction and purification processes has the benefits of good repeatability and stability..
