    Understanding human pre-implantation Development has important implications for assisted reproductive technology (ART) and for human Embryonic stem cell (hESC)-based therapies. Owing to limited resources, the cellular and molecular mechanisms governing this early stage of human Development are poorly understood. Nonetheless, recent advances in non-invasive imaging techniques and molecular and genomic technologies have helped to increase our understanding of this fascinating stage of human Development. Here, we summarize what is currently known about human pre-implantation Embryo Development and highlight how further studies of human pre-implantation Embryos can be used to improve ART and to fully harness the potential of hESCs for therapeutic goals.

    Abstract:

    Abstract Objective : To determine the effect of hydrosalpinx fluid-containing medium on murine Embryo Development and implantation. Design : The Development of one-, two-, and four-cell mouse Embryos in medium containing 5%, 10%, and 20% of human hydrosalpinx fluid was observed. Implantation rates of mouse Embryos transferred into the uterine horn with hydrosalpinx fluid-containing media were determined. Setting : Private hospital-based fertility center and IVF program. Main Outcome Measure(s) : Percentage of Embryos continuing cell division and implantation rates after ET. Result(s) : Hydrosalpinx fluid in culture medium affected Embryo Development in a dosedependent fashion. The injection of hydrosalpinx fluid-containing medium into the uterine horn did not affect Embryo implantation. Conclusion(s) : Hydrosalpinx fluid negatively affects murine Embryo Development, but its presence in the uterine horn at ET did not affect implantation.

    Abstract Atmospheric oxygen (∼20%) in culture significantly impairs preimplantation Embryo Development. However, it is not known whether all stages of preimplantation Embryo Development are susceptible to oxygen toxicity. This study investigated the temporal responses of preimplantation Embryos to oxygen conditions in vitro . Mouse Embryos were cultured in atmospheric (∼20%) or lower (5%) oxygen concentrations for the first 48h, followed by culture in the same or reciprocal oxygen concentrations for another 48h: group 1 (control, 5 and 5%); group 2 (5 and 20%); group 3 (20 and 5%); and group 4 (20 and 20%). Time-lapse microscopy was performed with imaging of individual Embryos at 15-min intervals.
