Compréhension et généralisation du mécanisme de protection de peptides hormonaux par conjugaison à l'albumine
2007Co-Authors: Parisien MichèleAbstract:Le glucagon-like peptide 1 (GLP-1) et surtout l'exendin-4 sont de puissantes hormones insulinotropiques dépendantes du glucose dont le potentiel pourrait être grandement exploité dans le traitement des personnes atteintes du diabète de type II (diabète non-insulinodépendant mellitus). Le GLP-1 est sécrété par les cellules intestinales de type L, à partir du gène du préproglucagon. Cependant, il est rapidement dégradé dans La circuLation par des enzymes protéolytiques, particulièrement par une Endopeptidase, La dipeptidyl peptidase IV (DP IV), qui l'inactive complètement. Par conséquent, l'utilisation thérapeutique du GLP-1 est limitée. L'exendin-4, un agoniste du récepteur du GLP-1, est sécrété par les gLandes salivaires du lézard GiLa Monster et possède un potentiel biologique supérieur au GLP-1 grâce à sa stabilité accrue en circuLation. Une pLate-forme technologique telle La conjugaison à l'albumine (DAC™ et PC-DAC™) pourrait améliorer le profil pharmacodynamique et pharmacocinétique de ces molécules thérapeutiques natives en augmentant leur stabilité face à La dégradation enzymatique et en diminuant le potentiel immunogénique de molécules exogènes, tel l'exendin-4. La présente étude vise donc à mieux comprendre, à analyser et à évaluer le pouvoir de protection que La conjugaison à l'albumine confère aux peptides hormonaux et, plus spécialement, à l'exendin-4, afin de mettre au point un agent thérapeutique plus puissant et plus stable qui servirait à traiter le diabète de type 2. Une série d'analogues du GLP-1 et de l'exendin-4 ont été synthétisés sous forme de DAC™ et les dérivés résultants ont été bioconjugués in vivo ou in vitro à La cystéine en position 34 de l'albumine, soit de rat (RSA) ou humaine (HSA). L'expérimentation in vivo chez La souris diabétique a permis d'évaluer le potentiel insulinotropique des différents dérivés à l'étude. L'activité biologique et La stabilité des DAC™ de l'exendin-4 lorsque bioconjugués in vivo sont de beaucoup supérieures à l'exendin-4 native et l'effet peut être observé jusqu'à 24 heures suivant le test oral de tolérance au glucose. Le profil pharmacocinétique démontre bien cette stabilité conférée par La conjugaison à l'albumine. Lorsque les peptides DAC™ sont conjugués in vitro à l'albumine, le potentiel insulinotropique est de beaucoup supérieur à celui des DAC™ seuls et des molécules natives évidemment. L'évaluation de l'immunogénicité de l'exendin-4 chez le rat Sprague Dawley a permis de démontrer que La conjugaison à l'albumine de rat diminuait le potentiel immunogénique de cette molécule lors de La phase thérapeutique et de La première phase d'immunisation. Ces résultats tendent donc à démontrer l'efficacité de La conjugaison à l'albumine de manière in vitro, plus que in vivo afin d'augmenter l'activité biologique et La stabilité en circuLation des molécules thérapeutiques natives. La poursuite de cette étude dans le but de mieux comprendre le mécanisme et les propriétés de La conjugaison à l'albumine devrait permettre de mettre au point non seulement un agent thérapeutique efficace pour traiter le diabète de type 2, mais aussi de mettre au point plusieurs autres agents thérapeutiques qui pourraient profiter de cette technologie.\ud ______________________________________________________________________________ \ud MOTS-CLÉS DE L’AUTEUR : diabète, exendin-4, GLP-1, albumine, conjugaison

Production d'un peptide antigénique par l'insulin-degrading enzyme
2012Co-Authors: Parmentier NicolasAbstract:Most antigenic peptides recognized by CD8 T lymphocytes are produced through degradation of intracelluLar proteins by the proteasome. However, some antigenic peptides are produced by a proteasome-independent pathway, which is poorly characterized. MAGE-A3168-176 is a tumor antigenic peptide presented by HLa-A1 and widely used for vaccination of meLanoma patients. We observed that proteasome and tripeptidyl peptidase inhibitors failed to block presentation of the antigen by tumor cells. However, processing of this peptide occured in the cytosol because TAP-inhibition prevented its presentation. To characterize the cytosolic peptidase producing MAGE-A3168-176 we setup an in vitro digestion assay using a 20-mer precursor peptide encompassing the sequence of the antigenic peptide. We observed that only the cytosolic fraction was able to produce the antigenic peptide from this precursor. This production was abolished by treating the cytosolic fraction with o-phenanthroline, a broad-spectrum inhibitor of metallopeptidases. This inhibitor also blocked the presentation of MAGE-A3168-176 by tumor cells. Processing of the antigenic peptide was observed in HLa-A1 cells electroporated by the 20-mer precursor or by a precursor modified at the C- and N-terminus to exclude exopeptidase activity. With both precusors, the processing was inhibited by treating cells with o-phenanthroline concluding to a major role of a cytosolic metalloEndopeptidase. One such enzyme is insulin-degrading enzyme (IDE). We observed that depletion of IDE abolished the capacity of a cytosolic fraction to produce the antigenic peptide. Furthermore, recombinant IDE was able to produce the peptide in vitro from the 20-mer peptide. Using a siRNA approach we observed that a reduction of IDE expression in tumor cells resulted in a reduced capacity of MAGE-A3168-176 presentation. Lastly, half-life studies revealed that in meLanoma cells proteasome and IDE act as parallel ways for the degradation of MAGE-A3 contributing together to a different set of peptides.La majorité des peptides antigéniques reconnus par les lymphocytes T cytolytiques sont produits à partir de La dégradation des protéines celluLaires qui implique principalement le protéasome. Certains peptides antigéniques sont cependant produits par une voie qui ne dépend pas du protéasome. Le peptide antigénique dérivé de La protéine tumorale MAGE-A3 (EVDPIGHLY) et présenté par le HLa-A1 a été utilisé dans de nombreux essais de vaccinations chez des patients souffrant de méLanomes métastatiques. Au cours de ce travail, nous avons observé que ni l’inhibition du protéasome ni celle de La tripeptidyl peptidase II ne compromettait La présentation de l’antigène par des cellules tumorales. La présentation de l’antigène étant dépendante du transporteur TAP, ceLa indiquait que MAGE-A3/HLa-A1 était produit dans le cytosol. Nous avons caractérisé l’activité produisant le peptide à partir d’un précurseur de 20 acides aminés comprenant La séquence du peptide antigénique. Nous avons observé que seule La fraction cytosolique était capable de produire le peptide antigénique. La fraction cytosolique traitée avec un inhibiteur de métallopeptidases, l’orthophénanthroline, ne produisait plus le peptide antigénique. Ce même inhibiteur diminuait La présentation du peptide antigénique par des cellules tumorales. La présentation de l’antigène dans des cellules HLa-A1+/ MAGE-A3- à partir du précurseur de 20 acides aminés ou d’un autre modifié à ses deux extrémités indiquait que La production de l’antigène dépendait d’une Endopeptidase. La caractérisation de La présentation de l’antigène indiquait que l’insulin-degrading enzyme (IDE) était vraisembLablement La peptidase qui produisait le peptide. Nous avons ensuite observé qu’une IDE recombinante était capable de produire le peptide antigénique et qu’une déplétion de l’IDE abolissait La capacité de production du peptide antigénique par La fraction cytosolique. Le rôle de l’IDE dans La production du peptide antigénique a finalement été confirmé en observant une inhibition de La présentation de l’antigène dans des cellules transfectées avec des siRNA spécifiques de l’IDE. Mutations to a glycine loop in the catalytic site of human Lon changes its protease, peptidase and ATPase activities.

Eukaryotic Lon possesses three domains, an N-terminal domain, an ATPase domain and a proteolytic domain. It requires ATP hydrolysis to digest Larger, intact proteins, but can cleave small, fluorogenic peptides such as Glu-ALa-ALa-Phe-MNA by only binding, but not hydrolyzing, ATP. Both ATPase and peptidase activities can be stimuLated by the binding of a Larger protein substrate, such as β-casein. To better understand its mechanism of action, we have prepared several point mutants of four conserved residues of human Lon (G893A, G893P, G894A, G894P, G894S, G893A-G894A, G893P-G894A, G893A-G894P, T880V, W770A, W770P) and studied their ATPase, protease and peptidase activities. Our results show that mutations to Gly894 enhance its basal ATPase activity but do not change its β-casein-stimuLated activity. The loop containing Gly893 and Gly894, which fLanks Lon's proteolytic active site, therefore appears to be involved in the conformational change that occurs upon substrate binding. Furthermore, mutations to Trp770 have the same general effects on the ATPase activity as mutations to Gly893, indicating that Trp770 is involved in ATPase stimuLation. We have also established that this loop does not need to move in order to cleave small, fluorogenic peptides, but does move during the digestion of β-casein. Finally, we also noted that Lon's ability to digest small peptides can be inhibited by moderate ATP concentrations.
