    Epilobium sp. are commonly used in traditional medicine in the treatment of early stages of benign prostatic hyperplasia and inflammation. It is suggested that a dominating constituent, oenothein B, is responsible for the extracts therapeutic effects. Several bioactivities were established for extracts and oenothein B in various in vitro models, but due to the questionable bioavailability of this dimeric macrocyclic ellagitannin, their significance in the in vivo effects remains unresolved. We have thus focused our attention on a complex comparative investigation of the in vitro and in vivo activities of phytochemically characterized Epilobium angustifolium aqueous extract and oenothein B on prostate cancer cells proliferation. Incubation of different cell lines with E. angustifolium aqueous extract resulted in a significant reduction of proliferation of PZ-HPV-7 and LNCaP cells, which was partly associated with antiandrogenic activity. These effects were fully congruent with oenothein B, examined in parallel. Oral supplementation of rats implanted with LNCaP cells with E. angustifolium aqueous extract 50–200 mg/kg b. w. resulted in a reduction of the occurrence of prostatic adenoma up to 13 %. Oenothein B was not detected in the urine and feces of the E. angustifolium aqueous extract-treated group, however, conjugates of nasutins gut microbiota metabolites of ellagitannins were detected in the urine, while in human volunteers supplemented with Epilobium tea, only urolithin conjugates were present. Despite observing significant and consistent effects in vitro and in vivo, we were unable to point out unequivocally the factors contributing to the observed E. angustifolium aqueous extract activity, facing the problems of an unknown metabolic fate of oenothein B and interspecies differences in E. angustifolium aqueous extract gut microbiota metabolism.

    Abstract Ethnopharmacological relevance The Epilobium genus (willowherb) comprises of ca. 200 species of herbaceous plants distributed around the world. Infusions prepared form willowherbs have been traditionally used externally in skin and mucosa infections and in the treatment of benign prostate hyperplasia. Nowadays extracts from different Epilobium species are widely used by patients, however the lack of clinical studies does not allow to fully establish their efficacy. The present review summarizes published data on phytochemistry, ethnopharmacological use and pharmacological studies concerning willowherb species investigated throughout past few decades. Materials and methods Literature survey was performed using Scopus, PubMed, Web of Science and Reaxys databases looking for papers and patents focused on chemical composition and bioactivity of Epilobium species. Systematic research in ethnopharmacological literature in digitalized sources of academic libraries was also carried out. Result The chemical composition of different Epilobium species and their bioactivities are described. The detailed information on constituents isolated and detected by chromatographic methods is given. The studies show that polyphenols are main compounds occurring in Epilobium herb among which flavonoids, phenolic acids and tannins (oenothein B and oenothein A) are dominating constituents. The extracts and some isolated compounds from Epilobium sp. were shown to possess antimicrobial, anti-proliferative, anti-inflammatory, analgesic and antioxidative activities. Because many studies suggest that oenothein B as dominating constituent may be responsible for Epilobium sp. pharmacological effects, its documented bioactivities were also described. Conclusions The pharmacological studies performed on Epilobium justify the traditional use of this species in external and in gastrointestinal inflammations. As far as the treatment of benign prostate hyperplasia (BPH) is considered, in the literature, there are some reports indicating that Epilobium extracts have a beneficial effect for this disorder, but the number of in vitro studies is not sufficient and the in vivo studies are not conclusive or too preliminary to draw a final conclusion about the efficacy of Epilobium preparations. More in vitro, in vivo and clinical studies to confirm this mode of action are strongly needed. Epilobium’s extracts have also documented antioxidative and potential anti-inflammatory properties. Oenothein B can be considered as responsible for some of Epilobium pharmacological properties. Because of the lack of clinical data further studies are needed to provide an evidence base for traditional uses of plant materials belonging to the Epilobium genus.

    Extracts from Epilobium sp. herbs have been traditionally used in the treatment of prostate-associated ailments. Our studies demonstrated that the extracts from Epilobium angustifolium, Epilobium parviflorum and Epilobium hirsutum herbs are potent prostate cancer cells (LNCaP) proliferation inhibitors with IC50 values around 35 mg/ml. The tested extracts reduced prostate specific antigen (PSA) secretion (from 325.625.3 ng/ml to ~90 ng/ml) and inhibited arginase activity (from 65.21.1 mUnits of urea/mg of protein to ~40 mUnits of urea/mg protein). Selected constituents of extracts (oenothein B, quercetin-3-O-glucuronide, myricetin-3-O-rhamnoside) were proventobeactiveinrelationtoLNCaPcells.However,oenotheinBwasthestrongestinhibitorofcellsproliferation (IC50=7.80.8 mM), PSA secretion (IC50=21.93.2 mM) and arginase activity (IC50=19.22.0 mM). Additionally, ellagitannins from E. hirustum extract were proven to be transformed by human gut microbiota into urolithins. Urolithin C showed the strongest activity in the inhibition of cell proliferation (IC50=35.23.7 mM), PSA secretion (reduced PSA secretion to the level of 100.731.0 ng/ml) and arginase activity (reduced to the level of 27.93.3 mUnits of urea/mg of protein). Results of the work offer an explanation of the activity of Epilobium extractsand supporttheuseof Epilobium preparationsin thetreatment of prostate diseases. Copyright © 2013 John Wiley & Sons, Ltd.

    Objectives The aim of this work was to determine the effect of standardized aqueous extracts from Epilobium angustifolium L., E. parviflorum Schreb. and E. hirsutum L. herbs on the apoptosis of hormone-dependent prostate cancer cells (LNCaP). Methods The extracts were characterized using high-performance liquid chromatographydiode array detector coupled with mass spectrometry (HPLC- DAD-MS/MS). Apoptosis in the cells was analysed using Annexin V-fluorescein isothiocyanate, and mitochondrial potential, Dym, using JC-1 by flow cytometry. Caspase-3 activity was determined by enzyme-linked immunosorbent assay. Key findings Using the HPLC-DAD-MS/MS method, 38 constituents were char- acterized. Extracts contained significant amounts of oenothein B as well as flavo- noids and phenolic acids. Exposure of LNCaP cells to the extracts (20, 50 and 70 mg/ml) resulted in a significant increase in the level apoptotic cells, from 2.86 0.5% (for untreated cells) up to 86.6 1.0%. All extracts significantly decreased the mitochondrial potential, Dym, resulting in an increase in the activity of caspase-3 from 0.3 0.07 ng/mg of protein (for untreated cells) up to 1.26 0.32 ng/mg of protein. Conclusions This study demonstrated that Epilobium extracts are active against LNCaP prostate cancer cells and that their apoptotic activity is related to activa- tion of the mitochondrial pathway. The high oenothein B content may influence the biological activity of these plant materials.

    Willow herb tea or preparation are available and relatively popular in the European market, and claimed to be effective inter alia because of their anti-inflammatory activity. The present study is therefore aimed at comparing the anti-inflammatory and antioxidant activity of extracts of the three most popular Epilobium species (E. angustifolium, E. hirsutum and E. parviflorum) and at juxtaposing this activity against the dominating compounds from the following extracts: oenothein B (OeB), quercetin-3-O-glucuronide and myricetin-3-O-rhamnoside. The phytochemical analysis of the extracts has shown that OeB quantities vary between 20% and 35%, while flavonoids content does not exceed 2%. All extracts have inhibited the activity of hyaluronidase and lipoxygenase with IC₅₀ around 5 μg/ml and 25 μg/ml. The inhibition of hyaluronidase is related with the presence of OeB, a strong inhibitor of this enzyme (IC₅₀) 1.1 μM). Additionally, the extracts inhibited myeloperoxidase (MPO) release from stimulated neutrophils. OeB inhibited MPO release similarly to the anti-inflammatory drug indomethacin with IC₅₀ 7.7 μM and 15.4 μM, respectively. Tested extracts significantly reduced the production of reactive oxygen species (ROS) from f-MLP and PMA induced neutrophils with IC₅₀ 5 μg/ml and 25 μg/ml, respectively. The flavonoids content seems to exert little influence on extracts' activity, contrary to OeB, whose high concentration explains the activity of extract obtained from Epilobium. Tested currently marketed Epilobium preparations are often wrongly assigned, but we should stress that the level of OeB in all tested herbs was high and always exceeded 2% in raw material.

    An aqueous extract of Epilobium angustifolium and its main compound oenothein B (OeB), a dimeric macrocyclic ellagitannin, are specifically able to induce the neutral endopeptidase (NEP) in prostate cancer cells. The angiotensin-converting enzyme (ACE) is not influenced. Additionally, a weak but statistically significant inhibition of cell proliferation is observed. Simultaneous treatment of the cells with arabinosylcytosine and the extract as well as the OeB, leads to an additional enhancement of NEP activity. Taking into account the role of this peptidase in prostate cancer progression, our results might offer a pharmacological explanation for the use of Epilobium in folk medicine.

    The ability of Epilobium extracts and polyphenols to induce neutral endopeptidase (NEP) activity and to inhibit the proliferation in cell lines with high NEP expression (SK-N-SH) and with low NEP expression (PC-3) was investigated. Epilobium extracts enhanced in a dose-depend manner NEP activity in both cell lines with additional inhibition of cell proliferation. The sensitivity of cells depended on basal enzyme activity. SK-N-SK cells were much more sensitive than PC-3 cells. Oenothein B enhanced NEP activity at a concentration of 5-40 microM while quercetin-3-glucuronide and quercetin-3-O-(6"-gal-loyl) galactoside showed slight or no activity at a concentration of 100 microM. The comparison of activities of the extracts with oenothein B, a dimeric macrocyclic ellagitannin, suggests that the latter is mostly responsible for the observed effects. Taking into account the role of NEP in the homeostasis of signalling peptides, Epilobium angustifolium extracts may be a potential herbal remedy in diseases connected with the disturbed metabolism of signaling peptides caused by an unbalanced neutral endopeptidase activity.

    Willow herb (Epilobium angustifolium L.) extracts showed inhibitory activity against the metallopeptidases: neutral endopeptidase (NEP), angiotensin-converting enzyme (ACE), and aminopeptidase N (APN). A bioassay-guided fractionation led to the isolation of several flavonoids and phenolic acids and an ellagitannin. The dimeric macrocyclic ellagitannin oenothein B inhibited the neutral endopeptidases in a dose-dependent manner with IC50 = 20 microM. Other polyphenols showed weaker activity but their synergistic activity cannot be excluded. Taking into account the role of these peptidases in prostate diseases, the results may partly support and explain the use of Epilobium extracts in folk medicine.
