Induction of Apoptosis in an Intestinal Epithelial Cell Line, IEC-6

We have already reported that, in FCS culture, these Cells exhibited signs of differentiation after reaching confluence, represented by the induction of alkaLine phosphatase (ALP) activity. The IEC-6 Cells then became detached from the culture plates, apoptosis being induced in the Cells. The sequential events in in vitro culture of IEC-6 Cells closely resemble the in vivo differentiation of small intestinal Epithelial Cells. In this study, we examined the nature of the signals involved in the induction of apoptosis in IEC-6 Cells. Apoptosis was induced in an IEC-6 culture by stimulating with ionomycin. The addition of TPA inhibited apoptosis, inducing a proliferate response. However, TPA enhanced the induction of apoptosis when added together with ionomycin. The effects of other reagents were also examined, and the results suggest that the stimuli inducing apoptosis and those inducing proliferation were closely related.

Cells which became detached from the culture plates after differentiation showed DNA fragmentation and chromatin condensation, indicating that the Cells had undergone apoptosis. Cell death could be induced in subconfluent, undifferentiated IEC-6 Cells when they were stimulated with ionomycin and heat-shock, or when calcium was removed from the culture medium. However, apoptosis was not induced in these Cells. Cell differentation was induced in IEC-6 cultures by adding retinoic acid, and stimulation of these Cells resulted in apoptosis. These results show that when IEC-6 Cells became differentiated, they were susceptible to apoptotic signals. Our system provides evidence that apoptosis is the final step of differentiation in the small intestine.

After IEC-6 Cells had reached confluence, alkaLine phosphatase (ALP), a differentiation marker, was introduced and its activity increased thereafter. Histochemical staining of the Cells indicated that most of the ALP-positive Cells did not proliferate. These results indicate that the Cells differentiated during culture, which accompanied the end of proliferation. Furthermore, we found that the Cells which became detached from the cultured plates had undergone apoptosis (programmed Cell death). The differentiation of IEC-6 Cells in the culture closely resembled the in vivo differentiation of small intestinal Epithelial Cells.

characterization of a porcine lung Epithelial Cell Line suitable for influenza virus studies
Journal of Virology, 2001Co-Authors: Olga Goloubeva, Richard J Webby, Robert G WebsterAbstract:We established a porcine lung Epithelial Cell Line designated St. Jude porcine lung Cells (SJPL) and demonstrated that all tested influenza A and B viruses replicated in this Cell Line. The infectivity titers of most viruses in SJPL Cells were comparable to or better than those in MDCK Cells. The propagation of influenza viruses from clinical samples in SJPL Cells did not lead to antigenic changes in the hemagglutinin molecule. The numbers of both Sia2-3Gal and Sia2-6Gal receptors on SJPL Cells were greater than those on MDCK Cells. Influenza virus infection of SJPL Cells did not lead to apoptosis, as did infection of MDCK Cells. No porcine endogenous retrovirus was detected in SJPL Cells, and in contrast to MDCK Cells, SJPL Cells did not cause tumors in nude mice.
