Phylogeny and Classification of Euglenophyceae: A Brief Review
Frontiers in Ecology and Evolution, 2016Co-Authors: Carlos Eduardo De Mattos Bicudo, Mariângela MenezesAbstract:Euglenophyceae (phototrophic euglenids) are an important lineage within the Euglenida, Euglenozoa. Most of the approximately 3,000 described species are free-living, phototrophic, unicellular flagellates with one to several plastids of secondary origin, three bounding membranes and chlorophylls a and b; but, the lineage also includes colorless species that lost their photosynthesizing capability. They show a typical cell membrane consisting of parallel proteinaceous strips and microtubules located underneath the plasma membrane, and discoid mitochondrial cristae. Euglenozoa are a monophyletic group that includes, besides Euglenida, the Kinetoplastidea, Diplonomea and Symbiontida. Since the class Euglenophyceae was proposed, its classification system has undergone several revisions, mainly after the adoption of molecular techniques. This article summarizes recent advances in the phylogeny and classification of the phototrophic euglenids, aiming at understanding the situation of the group within the phylum Euglenozoa, as well as its evolutionary relationships and the changes in its taxonomic classification. The current status of the group, as well as the limitations derived from the lack of inclusion of tropical strains in phylogenetic studies is briefly discussed.

Taxonomic and ultrastructural survey of Trachelomonas (Euglenophyceae) of a shallow subtropical reservoir in the municipality of Triunfo, southern Brazil

The reservoir covers approximately 15 ha and has a maximum depth of 2 m. Samples were collected monthly at two stations during 14 consecutive months, from February 1995 to March 1996, using a 25-μm mesh plankton net. Of a total of 26 Trachelomonas Ehr. emend. Defl. taxa previously identified, 15 were presently also studied in detail, including ultrastructure using a scanning electron microscope. The range of limnological conditions under which each taxon was collected is given, as well the distribution of each taxon in Brazil. Most of the 15 taxa are distributed widely throughout the world.

We sampled 10 stations in lotic and lentic environments, between 2011 and 2013. The 34 samples, resulting from sampling, were collected with a plankton net (30 µm), and preserved with 4% formalin. Nine taxa are new records for Rio Grande do Sul State, found in only three of the 10 stations sampled. Among these, Trachelomonas bernardii Wolosz. f. major Defl., T. caudata Defl. f. pseudocaudata (Defl.) Pop., T. hirta Cunha var. hirta, and T. torosa (Bal.) Tell & Conf. are first reports for the country.

Hoehnea, 2015Co-Authors: Sandra Maria Alvesdasilva, Isabele Corino KleinAbstract:This work is the result of a taxonomic study of three genera of pigmented Euglenophyceae in the Area de Protecao Ambiental do rio Ibirapuita, located in the southwest of the Pampa Biome, where the climate is subtropical. Four samplings were carried out between March 2011 and May 2013 covering 10 sites, six at Ibirapuita River and four in surrounding environments. Qualitative analysis resulted in the identification of 33 specific and infraspecific taxa, one of Cryptoglena, two of Monomorphina, and 30 of Phacus. We recorded Phacus formosus and P. triqueter, which are first reports for Rio Grande do Sul State. Despite the high diversity and species richness in some places, most taxa had infrequent distribution (61%). Phacus curvicauda Swir, P. hamatus Pochm, P. longicauda (Ehr.) Duj. var. longicauda, P. orbicularis var. orbicularis Hubner, and P. tortus (Lemm.) Skv. were frequent (totaling 15%), while the others were sporadic.

(pigmented Euglenophyceae) undertaken in two areas located in the northern portion of Patos Lagoon - Lake Casamento and associated ecosystems (30o03' - 30o34'S; 50o25' - 50o47'W) and ecosystems near Butiazal de Tapes (30o23' - 30o38'S; 51o16' - 51o29'W), revealed 37 specific and infra-specific taxa for this genus. The study aim was to increase knowledge of this genus in Rio Grande do Sul's coastal zone. The local climate is humid subtropical. The qualitative samples were collected in autumn and winter spring of 2003, on the banks of ponds, lakes, and marshes. Of the taxa identified, nine are new citations for the state while Phacus agilis Skuja var. inversa Bour., Phacus asymetricus Sokoloff, Phacus elegans Pochm., Phacus orbicularis Hubner f. communis Pop. and Phacus rostafinskii Drez. were reported for the first time in Brazil. All taxa are accompanied by maximum and minimum variations in air and water temperature, pH, and conductivity at the site where each taxon occurred.

A sample collected from rice-fish fields ( Oriza sativa L. and Cyprinus carpio Linnaeus, 1758) in shallow water(20 m 2 × 0.30 m deep) in the Itajai River Valley region, Santa Catarina (26o53’33’’S and 48o49’41’’W) revealed 48 morphospecies fromthe class Euglenophyceae represented by the genera Euglena , Lepocinclis , Phacus , Strombomonas and Trachelomonas . Trachelomonas was the best represented genus with 26% of all taxa. The reddish water at the time of sampling was due to the high number of Euglenacaudata Hubner and Euglena sanguinea Ehr. individuals per plate (bloom). Twenty eight taxa are new records for the state of SantaCatarina. Descriptions, dichotomous keys, size variation, length/width relationship (Rc/l), illustrations and geographic distri bution areprovided for all taxa. diversity and ecological considerations on pigmented Euglenophyceae in the state park of the jacui delta rio grande do sul southern brazil
Biociências (On-line), 2007Co-Authors: Sandra Maria Alvesdasilva, Guilherme Hermany, Maria Angelica OliveiraAbstract:Qualitative samples were taken from both lotic and lentic environments, in the State Park of the Jacui Delta, located in the vicinity of the capital of Rio Grande do Sul State (29°56’ e 30°03’S e 51°12’e 51°18’W), collected between May 1998 and September 1999, along the shore of an island, five bays (“sacos”) and one canal. The analysis revealed a high diversity of pigmented Euglenophyceae, with 130 taxa identified to genus, species infra-species levels. Amongst these, 42 taxa were new citations of the genus Trachelomonas for the Park. Twinspan classification analysis identified four groups. The study of the pigmented Euglenophyceae population composition and its relationship with environmental variables revealed a group of taxa (group B) with preference for habitats strongly changed by human action, with high levels of organic contamination and ammonium salts, as well as nitrite ions and orthophosphate, indicating moderately polluted waters (β-α mesossaprobic). Additionally, the environmental preferences of species never before described in the literature as characteristic of the conditions listed above, are given for the first time in Brazil. fatty acid composition as biomarkers of freshwater microalgae analysis of 37 strains of microalgae in 22 genera and in seven classes

FA analyses of 7 freshwater algal classes and 37 strains showed clear sim- ilarity within classes and strong differences amongst classes. Class was a dominant factor (66.4%) explaining variation in FA signatures of microalgae. The 7 algal classes comprised 4 separate groups according to their FA profiles: (1) Chlorophyceae and Trebouxiophyceae, (2) Bacillario- phyceae, (3) Cryptophyceae, Chrysophyceae, and Raphidophyceae, and (4) Euglenophyceae. Each group had a characteristic FA composition, although the proportional abundance of individ- ual FAs also differed between species and with environmental conditions. FAs found to be partic- ularly representative for each group (i.e. diagnostic biomarkers) were as follows: 16:4ω3 and 16:3ω3 for Chlorophyceae and Trebouxiophyceae; 16:2ω7, 16:2ω4, 16:3ω4, 16:4ω1, and 18:4ω4 for Bacillariophyceae; 22:5ω6 and 18:4ω3 for Cryptophyceae and Chrysophyceae (Synurales), 16:3ω1 for Chrysophyceae (Ochromonadales), 16:2ω4, 16:3ω4, 16:3ω1, and 20:3ω3 for Raphidophyceae; and 15:4ω2, 20:4ω3, 20:2ω6, 20:3ω6, and 22:4ω6 for Euglenophyceae. FAs thus offer a powerful tool to track different consumer diets in a lacustrine food web. Based on the 20:5ω3 (eicosapen- taenoic acid) and 22:6ω3 (docosahexaenoic acid) content among the investigated freshwater algal classes, Chlorophyceae, Trebouxiophyceae, and Chrysophyceae are of intermediate food quality for zooplankton, and Cryptophyceae, Bacillariophyceae, Euglenophyceae, and Raphidophyceae should be excellent resources for zooplankton.

Pigmented Euglenophyceae of a lentic environment at Upper Paraná River Floodplain.

Material and Methods: Quarterly samplings in the limnetic region of a floodplain lake between February 2000 and March 2009, numbering 30 samples obtained from both vertical and horizontal trawls using plankton net of a 15 µm-mesh aperture and subjected to Transeau-solution preservation. Temporary slides were prepared and analyzed in order to have the organisms identified, measured and photographed through digital camera, directly using an Olympus binocular optical microscope with a 400x/1000x magnification. The analysis included the calculation of Constancy (C) for the sample of each species. In order to summarize the environmental variability, a Principal Component Analysis was applied regarding the physical and chemical data of the environment collected concomitantly to the phytoplankton samplings. Results and Discussion: 36 taxa were identified. Genus Trachelomonas Ehr. Emend. Defl had the best representation, with 20 taxa. Among the taxa surveyed, 53% (16 taxa) constituted new records of occurrence for Upper Parana River Floodplain. The taxa recorded as frequent (33.3%) are considered cosmopolitan and presenting wide geographical distribution throughout Brazil; however, most of the taxa surveyed (66%) was classified as sporadic or rare, which is probably related to the high environmental variability observed along the study period. Conclusion: Taxa Monomorphina pyrum , Phacus contortus , Ph. longicauda var. major , Ph. rodriguesiae , Strombomonas fluviatilis var. levis , S. triquetra , Trachelomonas acanthophora , T. amphoriformis var. granulosa , T. armata var. armata fa. inevoluta , T. bernardii , T. decora , T. hispida var. crenullatocollis , T. megalacantha , T. molesta , T. obtusa , and T. woycickii var. pusilla expanded the records of biodiversity for Upper Parana River Floodplain and constituted new records of geographical distribution for Brazil.

The collections of biological material were performed monthly at two points (24o55’11”; 24o55’05” S and 53o29’09”; 53o29’14” W) from March 2011 to February 2012, totaling in 36 samples. Six taxa of chlorophytes of the order Chlorococcalesand fi ve taxa of euglenoids of the order Euglenales were identifi ed, thus being the total taxa recored for both orders; four are new records for the state of Parana: Golenkiniopsis solitaria (Korshikov) Korshikov, Euglena agilis Carter, E. clara Skuja, and E. geniculata Dujardin emend. Diel responses of phytoplankton of an Amazon floodplain lake at the two main hydrological phases

METHODS: Samples were taken with a Van Dorn bottle, at three depths (surface, middle and bottom), in two periods (low waters and high waters), for seven consecutive days in two schedules, night and morning. RESULTS: Phytoplankton was represented by 198 taxa and the Class Euglenophyceae, with 62 taxa, was the best-represented group in both periods. There were abrupt changes in the community during the high waters (potamophase). During this period, in conditions of partial atelomixis, we recorded higher values of phytoplankton biovolume and dominance of Gonyostomum semen (Ehr.) Dies. (Raphidophyceae). CONCLUSIONS: In the low waters (limnophase), conditions in the lake were more stable, the diversity and rate of change in the community were lower, and the frequent periods of stratification and mixing were responsible for the dominance of Geitlerinema sp. (Cyanobacteria)
