Protective Effect of Sambucus elbus Extract on Teratogenicity of Albendazole

The aim of present study was evaluation protective effect of Sambucus extract on albendazole induced- fetal malformation. Four groups of female pregnant Wistar rats (8 rats each group) were used. One group received normal saline (as control group). A single oral dose 30 mg/kg of albendazole was administered to rats on day 10 of gestation in group2. Rats in group3 received albendazole similar group2 and Sambucus extract at dose 600mg/kg. Rats in group4 received only Sambucus extract on day 10 of gestation. The rats were euthanatized on day 20 of gestation. The Fetuses were harvested and their weight and length and also the weight of placenta were measured. The skeletal malformation of Fetus was studied by stereomicroscope after staining by Alizarin red-Alcian blue. The length and weight of Fetuses were significantly decreased by albendazole but Sambucus extract prevent this effect. In group that received only Sambucus extract, the length and weight of Fetuses was similar to control group. Sambucus extract decreased albendazole effect on weight of placenta. There Fetus Resorption was decreased by Sambucus extract when co-administrated with albendazole. The incidence of skeletal malformations (mostly of the limbs, vertebrae and palate) decreased significantly by Sambucus extract when co-administrated with albendazole. Thus, Sambucus extract may have protective effect on albendazole teratogenicity; but this subject needs more detailed evaluation. To examine its safety in pregnancy condition, the influence of combination of ethanol extract of ginger and ethanol extract of noni fruit had been studied in pregnant Wistar rat. The ginger and noni extract were administered orally at dose level combination of 50 and 50 mg/kg BW , 500 and 500 mg/kg BW , 1000 and 1000 mg/kg BW on day sixth to fifteenth of gestation. The mother was sacrified on day nineteenth of gestation to observe life Fetus, Resorption, and growfail Fetus. One third of the Fetuses eviscerated for skeletal staining with alizarin red and two third of the Fetuses were hardened with Bouin’s fixation solution. No malformation were found in the eviscerated group. The combination at all doses did not cause any Fetus Resorption, growth failure and did not cause malformation in organ and skeleton. The combination dose of 500 and 500 as well as 1000 and 1000 mg/kg BW caused dark red to black color in the liver of rat Fetuses of 7.3 and 8.3 % respectively

Pengaruh Kombinasi Ekstrak Umbi Lapis Bawang Putih Dan Ekstrak Rimpang Kunyit Terhadap Janin Mencit Swiss-Webster

The garlic and turmeric extract were administered orally at dose level combination of 100:100, 500:500, and 1000:1000 mg/kg body weight on day sixth to fifteenth of gestation. The mother was sacrified on day eighteenth day of gestation to observe life Fetus, Resorption, and growth failure. One third of the Fetuses were eviscerated for skeletal staining with alizarin red and two third of the Fetuses were hardened with Bouin’s fixation solution. No malformation were found in the eviscerated group. Resorption of 1.11 % Fetuses and 1.11 % growfail Fetuses showed by extract combination of 100:100 mg/kg body weight. Growfail Growth failure of 4.59 % Fetuses showed by extract combination of 500:500 mg/kg body weight. Resorption of 0.96 % Fetuses showed by extract combination of 1000:1000 mg/kg body weight. The conclusion of this study, is that the combination of turmeric and garlic extract of 100:100 mg/kg body weight, which is equivalent to usual dose in human showed safe as herbal medicine, while in higher dose it is not recommended for pregnant woman.

Research on the role of NO in early embryo Resorption in mice
Chinese journal of veterinary science, 2002Co-Authors: Qifa Xie, Zepei Deng, Binggui SunAbstract:Intravenous injection of Lipopolysaccharide(LPS) induced Fetus Resorption on day 7 of gestation in mice.Variation of producton of NO and cytokines including IFN γ,IL 12 was measured both in serum and in homogenized uterus at 12,24,36 hours after administrated with LPS.Using immunohistochemistry,the changing expression of three NOS isolforms,nNOS,eNOS,iNOS, and Th2 cells in uterus were also detected at same time.Compared with normal pregnancy,there were significantly higher produciton ( P 0 01) of NO,IFN γ and IL 12 measured in homogenized uterus of mice administrated with LPS.The serum levels of NO were also higher ( P 0 01) than normal.The nNOS and iNOS were all expressed in uterus of mice treated with LPS,while the staining of eNOS was negative.Numerous positive cells of Th2 were only found in uterus of normal pregnancy.Intraperitoneal injection of sodium nitroprusside(SNP),a donor of NO,led to the early embryo Resorption.However,Aminoguanidine,a selective inhibitor of eNOS and nNOS,did not to reduce the early embryo Resorption of mice induced by LPS when given parenterally on day 6 to day 9 of gestation.The results presented strongly suggested that a role of NO derived from iNOS may act as an effector molecule in induction of the early embryo Resorption in mice.
