    Program Kemitraan Masyarakat (PKM) which performed on a group of cattle fattening is not qualified yet to solve the problems 1) rice straw as animal feed 2) agricultural waste and industrial waste as feed without forage 3) processing of rice straw into straw tape 4) processing agricultural waste and industrial waste into animal feed without forage 5) processing Growth Promotor derived from empon-empon and retrieval medicinal plants, and 6) making of cow manure waste into environmentally friendly fertilizer. The purposes of PKM are (1) to process of rice straw into straw tape as a nutritious animal feed (2) without forage feed processing (complete feed) (3) processing of Growth Promotors (4) local cattle fattening using tape straw and feed without forage as well as Growth Promotor. Benefits of PKM is to develop human resources and increase income of fattening local cattle farmers with lower production costs. The method used in PKM activities are education and training of local cattle fattening using tape straw feed and feed without forage plus Growth Promotor, processing waste into environmentally friendly fertilizer on UKMK of local cattle fattening members in Tuban.     Abstract:

    Scope: Zearalenone (ZEN) and α-zearalanol (α-ZAL, zeranol) were studied in differentiated Caco-2 cells and in the Caco-2 Millicell® system in vitro to simulate their in vivo intestinal absorption and metabolism in humans. Methods and results: In addition to metabolic reduction/oxidation, extensive conjugation with glucuronic acid and sulfate of the parent compounds and their phase I metabolites was observed. The positional isomers of the glucuronides and sulfates were unambiguously identified: Sulfonation occurred specifically at the 14-hydroxyl group, whereas glucuronidation was less specific and, in addition to the preferred 14-hydroxyl group, involved the 16- and 7-hydroxyl groups. Using the Caco-2 Millicell® system, an efficient transfer of the glucuronides and sulfates of ZEN and α-ZAL and their phase I metabolites into both the basolateral and the apical compartment was observed after apical administration. The apparent permeability coefficients (Papp values) of ZEN, α-ZAL and the ZEN metabolite α-zearalenol were determined, using an initial apical concentration of 20 μM and a permeation time of 1 h. Conclusion: According to the Papp values, the three compounds are expected to be extensively and rapidly absorbed from the intestinal lumen in vivo and reach the portal blood both as aglycones and as glucuronide and sulfate conjugates in humans.

    Abstract:

    α-Zearalanol (α-ZAL, zeranol) is a macrocyclic resorcylic acid lactone, which is highly estrogenic and used as a Growth Promotor for cattle in various countries. Little is known about the phase I metabolism of α-ZAL. We now report that α-ZAL and its major metabolite zearalanone (ZAN) are extensively monohydroxylated at the aromatic ring by microsomes from human liver in vitro. This novel pathway leads to catechols, the chemical structures of which were unambiguously established by the use of deuterium-labeled α-ZAL and ZAN, and by the synthesis of authentic standards. The aromatic hydroxylation of α-ZAL is almost exclusively mediated by the human cytochrome P450 (hCYP) 1A2 isoform. The catechol metabolites of α-ZAL and ZAN are unstable and readily oxidized to quinones, which could be detected among the metabolites of α-ZAL and ZAN generated by human hepatic microsomes and hCYP1A2. Furthermore, the quinone metabolites are able to form covalent adducts with N-acetylcysteine (NAC), as several of such adducts were found in microsomal incubations fortified with NAC. Aromatic hydroxylation of α-ZAL was also observed with bovine, porcine and rat hepatic microsomes. Further studies are needed to demonstrate the catechol pathway of α-ZAL in vivo and to assess its toxicological significance.

    Abstract:

    Abstract Dexamethasone (DXM) is a synthetic glucocorticoid that is authorized for therapeutic use in veterinary medicine. The European Community (EC) fixed a maximum residue limit (MRL) at 2 ng g−1 for liver, 0.75 ng g−1 for muscle and kidney tissues and 0.3 ng ml−1 for milk, while its use as a Growth-Promotor is completely banned. We developed earlier a LC–APCI–MS/MS method capable of quantifying such low MRL levels in milk. Minor modifications—concerning only the extraction procedure—were sufficient to allow the quantification of these levels as well in tissue samples. Validation results according to EC requirements concerning linearity, accuracy and precision were satisfactory. Limits of quantification of 0.375 ng g−1 were obtained for muscle and kidney tissues and of 1 ng g−1 for liver tissue, i.e. half the MRLs. Limits of detection were 0.09, 0.13 and 0.33 ng g−1 for muscle, kidney and liver tissues, respectively. Decision limits (CCα) were 1.2 ng g−1 for muscle and kidney tissues and 2.2 ng g−1 for liver tissue, while detection capabilities (CCβ) were 1.8, 1.9 and 2.5 ng g−1 for muscle, kidney and liver tissues, respectively. A simple deproteinization step with concentrated trichloroacetic acid followed by an extraction with ethyl acetate was sufficient to quantify DXM in plasma samples with a limit of quantification of 1 ng ml−1. After intravenous injection of DXM to cattle, a distribution within 30 min was observed followed by a phase of slow elimination characterized by a half-life of 9.2 h. In muscle tissue, low levels of DXM were found and a quick elimination was observed, with the DXM level falling below the MRL within 4 days after administration. Higher DXM levels were found in liver tissue compared to kidney tissue up to 4 days after administration. Nevertheless, in liver tissue DXM was below the MRL after 8 days of withdrawal time, while it was still as high as 2.5 ng g−1 in kidney tissue.

The result is enhancement of cattle weight per cow per day on local cattle amounted to 0.578+0.054 kg, ongole crossbreed cattle 0.887+0.100 kg, hybrid limousine cattle for 1.588+0.084 kg and Simmental crossbreed cattle of 1,967+0.255 kg. Abstrak Iptek Bagi Masyarakat (IbM) yang dilakukan pada kelompok penggemukan sapi lokal belum memenuhi syarat untuk menyelesaikan masalah 1) jerami padi sebagai pakan ternak 2) limbah pertanian dan limbah industri sebagai pakan tanpa hijauan 3) pengolahan jerami padi menjadi pita jerami 4) pengolahan limbah pertanian dan limbah industri menjadi pakan ternak tanpa pakan ternak 5) mengolah Promotor pertumbuhan yang berasal dari empon-empon dan pengambilan kembali tanaman obat, dan 6) pembuatan limbah kotoran sapi menjadi pupuk ramah lingkungan. Tujuan IbM adalah (1) mengolah jerami padi menjadi pita jerami sebagai pakan ternak yang bergizi (2) tanpa pengolahan pakan ternak (pakan lengkap) (3) pengolahan promoter pertumbuhan (4) penggemukan sapi lokal dengan menggunakan jerami dan pakan pita tanpa pakan ternak serta Promotor pertumbuhan. Manfaat IbM adalah mengembangkan sumber daya manusia dan meningkatkan pendapatan penggemukan peternak sapi lokal dengan biaya produksi lebih rendah. Metode yang digunakan dalam kegiatan IbM adalah pendidikan dan pelatihan penggemukan sapi dengan pakan jerami dan pakan tanpa pakan ternak ditambah Promotor pertumbuhan, pengolahan limbah menjadi pupuk ramah lingkungan pada UKMK penggemukan sapi potong di Kabupaten Babat dan Sugio di Lamongan. Kegiatan program IbM melibatkan 8 siswa untuk meningkatkan semangat kewirausahaan. Hasilnya adalah peningkatan bobot sapi per ekor per hari pada ternak lokal sebesar 0,578 + 0,054 kg, sapi potong silang ongole 0,887 + 0,100 kg, ternak limusin hibrida 1,588 + 0,084 kg dan sapi potong silang Simmental 1.967 + 0.255 kg.
