  • the mechanism of atorvastatin on the prevention of Heart Muscle Ischemia reperfusion injury
    Proceeding of Clinical Medicine, 2009
    Co-Authors: Ma Jie
    Abstract:

Objective:To observe the role of two ATP-sensitive K+channels in the protective effect of atorvastatin(ATV)on myocardial Ischemia-reperfusion injury(IRI).Methods:Thirty-two rabbits were randomly divided into four groups:control group,ATV group,ATV+5-HD(A1),ATV+HMR1883(A2).All rabbis were subjected to 30 min Ischemia and 30 min reperfusion. The max rate of rise of left ventricular pressure(±dp/dtmax) and left ventricular end-diastolic pressure(LVEDP) were detected during the experiment. Results:Compared with the control group, the indexes of Heart function were improved significantly(P0.05) in the ATV group and the A2 group. And there was no statistically significant between the control group and A1 group(P0.05).Conclusion:The mitochondrial KATP channel is likely involved in the protective mechanism that against myocardial IRI in rabbits of ATV.

    Abstract:

Objective:To observe the effect of atorvastatin(ATV) on myocardial Ischemia-reperfusion injury and the role of two ATP-sensitive K+ channels.Methods:Thirty-two rabbits were randomly divided into four groups, control group,ATV group,ATV+5-HD(A1) group,ATV+HMR1883(A2) group. All rabbits were subjected to 30 min Ischemia and 30 min reperfusion. The max rate of rise of left ventricular pressure(±dp/dtmax) and left ventricular end-diastolic pressure(LVEDP) were detected during the experiment. At the end of reperfusion, the area of necrosis and area of Ischemia were determined by Evans blue and TTC staining. Results:Compared with the control group, the indexes of Heart function were improved significantly and infarct size(AN/AAR) were reduced(P0.05) in ATV group and A2 group. And this effect was blocked by 5-HD.There was no statistically significant between the control group and A1 group(P0.05).Conclusion:Atorvastatin exerts an early cardio protective effect against myocardial Ischemia reperfusion in rabbits and mitochondrial KATP channel is likely involved in this protective mechanism.

  • The effects of fluvastatin on the prevention of Heart Muscle Ischemia reperfusion injury in rabbits
    Journal of Chinese Physician, 2006
    Co-Authors: Li Jian
    Abstract:

Objective To investigate the effects and mechanism of different dosage of fluvastatin on the prevention of Heart Muscle Ischemia reperfusion injury in rabbits.Method Thirty-five rabbits were randomly divided into 5 groups with 7 rabbits in each: sham group, myocardial Ischemia reperfusion control group, low dosage of fluvastatin pretreatment group (2 mg/kg,Group F1),middle dosage of fluvastatin pretreatment group(5 mg/kg,Group F2) and large dosage of fluvastatin pretreatment group(20 mg/kg,Group F3).The left ventricular systolic pressure(LVSP),the max rate of rise of left ventricular pressure(±dp/dt_(-max)) and left ventricular end-diastolic pressure(LVEDP) were detected during the experiment. At the end of reperfusion, the infarct size and area at risk were defined by Evans blue and TTC staining, and the levels of myocardial nitrogen monoxidum(NO) and nitricoxide synthase(NOS) were measured.Result Compared with the Ischemia reperfusion group, the indexes of Heart function improved significantly, the level of myocardial NO was increased significantly and the myocardial infarct size was decreased significantly in the groups F2 and F3.There was no significant difference between the group F1 and Ischemia reperfusion group. Conclusion Fluvastatin exerts a cardioprotective effect against myocardial Ischemia reperfusion injury in rabbits.NO is likely involved in this protective mechanism.

  • an analysis of efficiency of atorvastatin on the prevention of Heart Muscle Ischemia reperfusion injury
    China Modern Doctor, 2011
    Co-Authors: W U Yunliang
    Abstract:

Objective Evaluation of atorvastatin on myocardial Ischemia reperfusion injury process, improvement of cardiac function and myocardial protective effects. Methods 30 healthy male rabbits were randomly divided into 3 groups:sham operation group,Ischemia reperfusion group(I/R group),atorvastatin group(treatment group),1 hour before surgery with atropine treatment group were fed saline with 10mg/kg atorvastatin, left anterior descending coronary artery ligation after 30 minutes, open 1 hour to make rabbit model of Ischemia-reperfusion;by measured using physiological recording cardiac function,by Evans blue + 1% triphenyl tetrazolium chloride(TTC) staining, double isolated necrosis in ischemic myocardium. Results Sham perfusion before and after the LVSP,LVEDP,± dp/dtmax,were no significant differences(P0.05),I/R group and treatment group were significantly different(P0.05);compared with the sham operation group,I/R group and treatment group LVEDP increased significantly(P0.05),LVSP and ± dp/dtmax decreased significantly(P0.05);I/R group compared with the treatment group decreased LVSP and ± dp/dtmax was no significant difference(P0.05),but LVEDP increased and myocardial infarction rates declined significantly(P0.05).Conclusion Effect of atorvastatin on myocardial Ischemia and reperfusion injury.
