Botanical Journal of the Linnean Society, 2010Co-Authors: Barbara łotocka, Ewa OsińskaAbstract:The anatomy and ultrastructure of internodes, leaves and petals were compared in Hypericum elegans, H. inodorum, H. olympicum, H. forrestii and two genotypes of H. perforatum. Internode anatomy was variable between species with respect to the structure of the cortical and pith parenchyma, including the presence of secretory reservoirs. Also, the secondary growth was more extensive in shrubs, i.e. H. inodorum and H. forrestii. In leaves, phloem secretory reservoirs were formed in all species, mesophyll secretory reservoirs were absent only in H. elegans and internal nodules were present only in H. elegans and H. perforatum. The petals differed between species in the mesophyll structure and the occurrence and location of secretory structures. The phloem secretory reservoirs lacked sheaths, whereas these were distinct in the mesophyll reservoirs. Other ultrastructural traits of the reservoirs were similar in all the species studied, with the exception of the leucoplast ultrastructure. In internal nodules, the inner cells vs. sheath cells differed in the number of vesicles and other membranous structures and plastid ultrastructure. © 2010 The Linnean Society of London, Botanical Journal of the Linnean Society, 2010, 163, 70–86.

The IC 50 resulted between 0.062 and 1 mg/ml.

Molecular Phylogenetics and Evolution, 2013Co-Authors: Nicolai M Nurk, Mark A Carine, Santiago Madrinan, Mark W Chase, Frederick R BlattnerAbstract:Abstract Phylogenetic hypotheses for the large cosmopolitan genus Hypericum (St. John’s wort) have previously been based on morphology, and molecular studies have thus far included only a few species. In this study, we used 360 sequences of the internal transcribed spacer (ITS) region of nuclear ribosomal DNA (nrDNA) for 206 species representing Hypericum (incl. Triadenum and Thornea) and three other genera of Hypericaceae to generate an explicit phylogenetic hypothesis for the genus using parsimony and model-based methods. The results indicate that the small genus Triadenum is nested in a clade within Hypericum containing most of the New World species. Sister to Hypericum is Thornea from Central America. Within Hypericum, three large clades and two smaller grades were found; these are based on their general morphology, especially characters used previously in taxonomy of the genus. Relative to the most recent classification, around 60% of the sections of Hypericum were monophyletic. We used a Bayesian approach to reconstruct ancestral states of selected morphological characters, which resulted in recognition of characters that support major clades within the genus and a revised interpretation of morphological evolution in Hypericum. The shrubby habit represents the plesiomorphic state from which herbs evolved several times. Arborescent species have radiated convergently in high-elevation habitats in tropical Africa and South America.

Cladistic analysis of morphological characters in Hypericum (Hypericaceae)
Taxon, 2010Co-Authors: Nicolai M Nurk, Frederick R BlattnerAbstract:Hypericum is a worldwide-distributed genus with almost 500 species, including the medically used, facultative apomictic species H. perforatum. It is one of the few large plant genera for which alpha taxonomy has been completed and most species have been described. To conduct a formal cladistic analysis of the genus, we coded 89 morphological characters for all described taxa and analyzed the data for the species using parsimony and Bayesian methods. The obtained trees indicate that Hypericum is monophyletic if the monotypic genus Santomasia is included, and that Lianthus is its sister. The arrangement of the remaining genera of Hypericaceae included in the analysis is in congruence with molecular phylogenies. Within Hypericum the cladistic analysis revealed a basal grade containing Mediterranean species and three big clades containing most of the diversity of the genus. In contrast to earlier assumptions, we found no indication for an African origin of Hypericum, but assume that the genus evolved in what today is the Mediterranean area. Our phylogenies indicate a shrubby habit to be the ancestral state within Hypericum from which species with tree-like and herbaceous habit evolved, and that apomixis originated at least three times independently within the genus.

Conversion of protohypericin to hypericin was achieved by exposing samples to light for 30 min immediately before HPLC analysis. External standard calibration was used to quantify hypericin. Leaves showed the highest hypericin content of 0.36% w/w. Total aerial parts contained 0.43% w/w of hypericin. The relatively high hypericin content of the H. triquetrifolium in this study encourages the introduction, cultivation, and biological evaluation of hypericum specie in Jordan.

Journal of Ethnopharmacology, 2017Co-Authors: Ali Yucel, Erdem Yesilada, Onat AkinAbstract:Abstract Ethnopharmacological relevance Topical formulations such as oily extracts or ointments prepared with the flowering aerial parts of St. John's wort ( Hypericum perforatum L., Hypericaceae) have been used in the management of a wide range dermatological problems including superficial wounds and burns, bruises, contusions and many others in the worldwide traditional medicines. Aim of the study This is the first case study reporting the beneficial effects of an oily extract of St. John's wort in the treatment of pressure sores in a intensive care unit (ICU) patient. Material and methods The oily extract of St. John's wort was applied to a volunteer patient at ICU daily for forty successive days for wound care and treatment. Healing status was monitored macroscopically by measuring the wound size and stages at certain intervals as well as histopathological evaluation of the tissue sections taken at the initial and final dates of treatment. Results Evaluation of the results obtained from the macroscopical and histopathological experimentation have shown that oily extract of St. John's wort provided significant efficacy for the treatment of pressure sore wounds. Conclusion St. John's wort oily extract may be suggested as a cost-effective option for the prevention or treatment of pressure sores in ICU patients.
