    Abstract:

    The muscle relaxing activity of the essential oil of Hyssopus officinalis L. (Lamiaceae) and some of its main components (isopinocamphone, limonene and p-pinene) was studied on isolated preparations of guinea-pig and rabbit intestine. The essential oil and isopinocamphone inhibited the acetylcholine- and BaCl 2 -induced contractions in guinea-pig ileum in a concentration-dependent manner (IC 50 42.4 μg/ml and 61.9 μg/ml to acetylcholine; 48.3 μg/ml and 70.4 μg/ml to BaCl 2 ) whereas limonene or p-pinene left tissue contraction unchanged. In guinea-pig ileum H. officinalis essential oil also blocked the contractions induced by CaCl 2 . In isolated rabbit jejunum the essential oil reduced the amplitude of spontaneous movements and decreased the basal tone; neither haemoglobin, methylene blue, N ω -nitro-L-arginine methyl ester (L-NAME) or propranolol blocked the myorelaxant effect.

    Abstract The in vitro antimicrobial and cytotoxic activity of the essential oils of Hyssopus officinalis L. (HO) and H. officinalis var. decumbens (HOD) was determined using minimum time period required to kill Staphylococcus aureus (ATCC 6538 P) and Streptococcus sanguis (ATCC 10556). Cytotoxicity was evaluated using the brine shrimp (Artemia salina Leach) test by determining the percent of nauplii dead within 24 h. At the minimal concentration tested (0.1% w/v), killing time was 8 min for HOD and 15–30 min for HO. In the brine shrimp test, HOD had a LC50 (the concentration that would kill 50% of the brine shrimps within 24 h) lower than HO (156.03 μg/mL vs 191.06 mg/mL). Linalool, the main component of H. officinalis L. var. decumbens oil, was the most potent of the pure components tested.

    The antimicrobial activity of essential oil of Hyssopus officinalis L. var decumbens (Jordan & Fourr.) Briq. from France (Banon) and Hyssopus officinalis L. from Italy (Piedmont) was studied taking account of their chemical composition determined by GC and GC–MS. Pinocamphone and isopinocamphone are present in H. officinalis (4.4% and 43.3%, respectively), according to the ISO 9841 Standard (1991 E) but they are lacking in var. decumbens, where linalol (51.7%), 1,8-cineole (12.3%) and limonene (5.1%) instead are predominant. The disc diffusion tests carried out on Gram-positive (Staphylococcus aureus and Enterococcus spp.) and Gram-negative bacteria (Klebsiella oxytoca, Escherichia coli, Proteus mirabilis, Pseudomonas spp. and two strains of Salmonella spp.) showed an antimicrobial activity generally negligible for H. officinalis, but broader, and in a few cases more evident (Enterococcus spp. and E. coli), for var. decumbens. All yeasts (seven strains of Candida albicans, C. krusei and C. tropicalis) were strongly inhibited by both species. In liquid medium the MIC of H. officialis was always >1.2% v/v for bacteria and between 0.6 and 1.2% v/v for yeasts, while the MIC of var. decumbens was between 0.15 and 0.6% v/v for the Gram-positive bacteria, 0.3 and 1.2% v/v for the Gram-negative bacteria and 0.15 and 0.3% v/v for the yeasts.     Abstract The essential oils of Hyssopus officinalis L. var. decumbens (HOD) from France (Banon) and H. officinalis (HO) from Italy (Piedmont) were studied by GC and GC/MS. The bicyclic monoterpene ketones, pinocamphone and isopinocamphone, were present in HO (4.4% and 43.3%, respectively, in accordance with the ISO 9841 Standard, 1991 E), but their percentages were very low in HOD, where instead linalool (49.6%), 1,8-cineole (13.3%) and limonene (5.4%) were predominant.

    The spasmolytic activity of the essential oil from Hyssopus officinalis L. var. decumbens (HOD) and its major pure components, linalool, 1,8-cineole and limonene was studied on isolated guinea-pig ileum contracted by acetylcholine and BaCl2. HOD and linalool inhibited the acetylcholine- and BaCl2-induced contractions in a concentration-dependent manner (IC50 values: HOD, 37 μg/mL and 60 μg/mL; linalool 10 μg/mL and 51 μg/mL). The antagonism appeared non-competitive. In contrast, 1,8-cineole and limonene showed only a weak spasmogen action. © 1998 John Wiley & Sons, Ltd.

Linalol, and in a lesser way, 1,8-cineole, may contribute to the greater antimicrobial activity of var. decumbens in comparison with H. officinalis, while limonene may be responsible for the antimycotic action observed in both oils, as suggested by results of the disc diffusion tests carried out on the pure reference substances. © 1998 John Wiley & Sons, Ltd.

    Abstract:

    Introduction: Hyssopus officinalis (L) (Hyssop, Family: Lamiaceae), one of the endemic Iranian perennial herb with a long history of medicinal use, was studied to detect some biologically active chemical constituents of the plant. Methods: The flavonoids of the hydromethanolic extract of the aerial parts of Hyssopus officinalis (L.) were studied by VLC and crystalisation of the major compound in subsequent fractions. Furthermore, the composition of its essential oil, total phenolic content and antioxidant activities were studied by GC-MS, Folin–Ciocalteau and DPPH reagents respectively. Results: Apigenin 7-O-β-D-glucuronide was isolated as the major flavonoid. All structural elucidation was performed by spectral means. A total of 20 compounds representing 99.97% of the oil have been identified. Myrtenylacetate , Camphor, Germacrene, Spathulenol were the main compounds The total phenol content of the n-butanol and ethylacetate extracts were determined spectrophotometrically according to the Folin–Ciocalteau procedure to be 246 mgGAE g-1 and 51 mgGAE g-1 in the aerial parts of Hyssopus officinalis. The antioxidant activities of apigenin 7-O-β-D-glucuronide, ethylacetate and n-butanol extracts were also determined by DPPH radical scavenging assay with IC50 values of 116×10_3, 103×10_3, 25×10_3 mg mL-1 respectively. The purified flavonoid showed weak radical scavenging activity (IC50 = 116×10_3mg mL_1). N-butanol extract, because of the highest content of total phenolic compounds (246 mgGAE100_1g) had the best antioxidant activity (IC50 = 25mg mL_1). Conclusion: On the whole, the findings of the study revealed that Hyssop possesses valuable antioxidant properties for culinary and possible medicinal use.

    Abstract:

    Hyssopus officinalis L. (Hyssop) is one of the most popular herbal preparations, mainly distributed in the East Mediterranean to central Asia. The plant has been used traditionally for medicinal purposes; generally, these therapeutic uses and health benefits of hyssop are largely based on folklore rather than on scientific substantiation, making it a good candidate to gather documentations, including the phytochemical content, in vitro experiments, animal models and human studies available in the recent scientific studies. A literature review on the chemical and biological aspects of the plant indicates that the main constituents of H. officinalis include several polyphenolic compounds, primarily the flavonoids apigenin, quercetin, diosmin, luteolin and their glucosides followed by other phenolic compounds chlorogenic, protocatechuic, ferulic, syringic, p-hydroxybenzoic and caffeic acids. Reports on the essential oils extracted from aerial parts of H. officinalis revealed several principal components, including terpenoids pinocamphone, isopinocamphone and β-pinene. Hyssop has moderate antioxidant and antimicrobial activity against Gram positive and negative bacteria activities together with antifungal and insecticidal antiviral properties in vitro. Animal model studies indicate myorelaxant, antiplatelet and α-glucosidase inhibitory activities for this plant. However, human studies, adverse reactions and clinical trials examining the reported properties of hyssop are absent and needs more attention to determine whether biological differences in findings of the studies reflect the different isolation procedures, different types of plant material used, collection time, locations or different chemotypes.   Key words: Hyssopus officinalis L., phenolic compounds, essential oil, extract.
