First clinical case of cutaneous leishmaniasis due to Leishmania (Viannia) braziliensis in a domestic cat from French Guiana

The Infected Animal had a cutaneous ulcer on the nose and nodules of different sizes in the ears. The diagnosis was confirmed by molecular analysis of cutaneous samples that detected the presence of Leishmania parasites and allowed identifying the Leishmania (Viannia) braziliensis species. The discovery of a cat Infected by L. (V.) braziliensis suggests the possibility that cats could be potential secondary reservoirs of Leishmania parasites in French Guiana. Thus, it would be important to investigate the possible epidemiological role of domestic cats in domestic foci of Leishmania in this region.

The Journal of Infectious Diseases, 2007Co-Authors: Toya Nath Baral, Patrick De Baetselier, Frank Brombacher, Stefan MagezAbstract:Very recent reports have documented that Trypanosoma evansi, the etiological agent of the livestock disease "surra," can cause human trypanosomiasis. In contrast to trypanosomes causing human African trypanosomiasis, T. evansi has a wide geographic distribution and host range, yet information about the immunobiological aspects of T. evansi trypanosomiasis is limited. Here, we show that, although T. evansi causes the induction of tumor necrosis factor (TNF), interferon-gamma, and nitric oxide during the early stage of infection, none of these molecules are crucial for parasitemia control and survival of the Infected Animal. However, TNF and TNF receptor 2 affect the induction of late-stage anemia. Using B cell- and immunoglobulin M (IgM)-deficient mice, we identified IgM as being crucial for parasitemia control and host survival. Collectively, our results show that, compared with other trypanosomes, T. evansi displays a distinct host-parasite interaction profile, give that, despite an infection-associated induction of proinflammatory molecules, only IgM antibodies contribute significantly to parasite control.

Characterization of a pathogenic CCR5 (R5)-tropic SIV/HIV chimeric virus (SHIV) molecular clone (SHIVAD8-EO) revealed that eight of eight Infected Animals developed cross-reactive neutralizing antibodies (NAbs) directed against an envelope glycoprotein derived from the heterologous HIV-1DH12 strain. A panel of plasmas, collected from monkeys inoculated with either molecularly cloned or uncloned SHIVAD8 stocks, exhibited cross-neutralization against multiple tier 1 and tier 2 HIV-1 clade B isolates. One SHIVAD8-Infected Animal also developed NAbs against clades A and C HIV-1 strains. In this particular Infected macaque, the cross-reacting anti–HIV-1 NAbs produced between weeks 7 and 13 were directed against a neutralization-sensitive virus strain, whereas neutralizing activities emerging at weeks 41–51 targeted more neutralization-resistant HIV-1 isolates. These results indicate that the SHIVAD8 macaque model represents a potentially valuable experimental system for investigating B-cell maturation and the induction of cross-reactive NAbs directed against multiple HIV-1 strains.

Virus-binding serum antibody titres were correlated strongly with disease progression (P < 0.005) and were predictive of disease outcome by 7 weeks after inoculation. Low virus-binding serum antibody responses to SIV occurred in Animals that also showed acute depletion of circulating CD20+ B cells. Acute damage to the CD4+ T cell and CD20+ B cell populations rendered some Animals incapable of mounting virus-specific antibody responses and these macaques became the rapidly progressing cases comprising approximately 20-30% of Infected Animal cohorts.

Two mitochondrial genes (the cytochrome c oxidase subunit I and the NADH subunit I) were tested by PCR. Four genotypes were identified: G1 (sheep strain), G2 (Tasmanian sheep strain), G3 (buffalo strain), and G5 (cattle strain). Fertile cysts were detected only in the lungs of 4.5% of the total G1 lung cysts, of 9.4% of the total G3 lung cysts, and in the only G5 Infected Animal. This is the first report of Echinococcus ortleppi (genotype G5) in Italy.
