Microcystin production by Radiocystis fernandoi (Chroococcales, Cyanobacteria) isolated from a drinking water reservoir in the city of Belém, PA, Brazilian Amazonia region

This non-axenic strain was submitted to a toxicity bioassay with mice and microcystin production analyzed by HPLC-DAD. The species was identified based on cultured and natural preserved material. Morphometric, developmental and reproductive characteristics were analyzed. The strain was cultured in liquid ASM-1 medium, at 25^1 8C, at an incident irradiance of 20 mmol photon m 22 s 21 and constant aeration. At the end of the exponential growth phase, cells were lyophilized and submitted to toxicity tests. The strain showed high toxicity to mice, by Intraperitoneal Route, with an approximate LD 100 of 60 mg kg 21 of body weight, producing characteristic symptoms of hepatotoxicity. Analyses performed by HPLC-DAD confirmed the production of microcystins, in a concentration of 3.83 mg mg 21 of lyophilized cells. This is the first reference related to the toxicity of the genus Radiocystis

Logambal, B V Aruna, Dinakaran R MichaelAbstract:The Indian major carps Catla catla, Labeo rohita and Cirrhinus mrigala were immunised against the potent bacterial fish pathogen, Aeromonas hydrophila by the Intraperitoneal Route, in field conditions. Two different polyvalent antigen preparations namely, whole cell and extracellular products (ECP) were used. Immunisation schedule consisting of a single booster dose given 28 days after priming resulted in a good agglutinating antibody response in the carps. Kinetics of the response were similar in the three carp species with both whole cell and ECP. Upon challenge with virulent strains, relative percent survival as high as 80-90% was recorded. Indian major carps are the most important freshwater species cultured in India and it is essential that they are protected against infections of A. hydrophila, a scourge of freshwater fish worldwide.

Journal of Virology, 2003Co-Authors: Thomas J Chambers, Yan Liang, Deborah A Droll, Jacob J Schlesinger, Andrew D Davidson, Peter J Wright, Xiaoshan JiangAbstract:Two yellow fever virus (YFV)/dengue virus chimeras which encode the prM and E proteins of either dengue virus serotype 2 (dengue-2 virus) or dengue-4 virus within the genome of the YFV 17D strain (YF5.2iv infectious clone) were constructed and characterized for their properties in cell culture and as experimental vaccines in mice. The prM and E proteins appeared to be properly processed and glycosylated, and in plaque reduction neutralization tests and other assays of antigenic specificity, the E proteins exhibited profiles which resembled those of the homologous dengue virus serotypes. Both chimeric viruses replicated in cell lines of vertebrate and mosquito origin to levels comparable to those of homologous dengue viruses but less efficiently than the YF5.2iv parent. YFV/dengue-4 virus, but not YFV/dengue-2 virus, was neurovirulent for 3-week-old mice by intracerebral inoculation; however, both viruses were attenuated when administered by the Intraperitoneal Route in mice of that age. Single-dose inoculation of either chimeric virus at a dose of 10 5 PFU by the Intraperitoneal Route induced detectable levels of neutralizing antibodies against the homologous dengue virus strains. Mice which had been immunized in this manner were fully protected from challenge with homologous neurovirulent dengue viruses by intracerebral inoculation compared to unimmunized mice. Protection was associated with significant increases in geometric mean titers of neutralizing antibody compared to those for unimmunized mice. These data indicate that YFV/dengue virus chimeras elicit antibodies which represent protective memory responses in the mouse model of dengue encephalitis. The levels of neurovirulence and immunogenicity of the chimeric viruses in mice correlate with the degree of adaptation of the dengue virus strain to mice. This study supports ongoing investigations concerning the use of this technology for development of a live attenuated viral vaccine against dengue viruses.

Immunostimulatory effect of leaf extract of Ocimum sanctum Linn. in Oreochromis mossambicus (Peters)

Dinakaran MichaelAbstract:Effect of leaf extract of Ocimum sanctum on (i) the specific and non-specific immune responses and (ii) disease resistance against Aeromonas hydrophila was investigated in Oreochromis mossambicus . Sheep red blood cells (SRBC) and Heat aggregated bovine serum albumin (HA-BSA) were used as antigens for specific and non-specific immune response studies, respectively. Antigens were administered through Intraperitoneal Route. Anti-SRBC antibody titres were determined by direct haemagglutination and anti-bacterial antibodies were determined by bacterial agglutination. Nitroblue tetrazolium (NBT) assay was used to determine neutrophil activity. Disease resistance was measured in terms of relative percent survival (RPS). The immunostimulatory effect of the leaf extract of O. sanctum , when administered through Intraperitoneal and oral Routes was obvious. Leaf extract of O. sanctum , when administered Intraperitoneally, stimulated both antibody response and neutrophil activity. Dietary intake of O. sanctum also enhanced the antibody response and disease resistance against A. hydrophila . Possibility of using O. sanctum as immunostimulant in the maintenance of finfish health in intensive freshwater aquaculture is suggested.

Evidence of anti-inflammatory effects of Passiflora edulis in an inflammation model

Most of the pharmacological investigations of Passiflora edulis have been addressed to its Central Nervous System activities, such as anxiolytic, anticonvulsant and sedative actions. Otherwise, there are few reports about the anti-inflammatory activity of the Passiflora species. The aim of this study was to investigate the mechanism of the anti-inflammatory effect of aqueous lyophilized extract obtained from leaves of Passiflora edulis var. flavicarpa Degener (Passifloraceae) in the mouse model of pleurisy induced by carrageenan (Cg), bradykinin, histamine or substance P, observing the effects upon leucocytes migration, myeloperoxidase (MPO), nitric oxide (NO) concentrations and tumor necrosis factor-alpha (TNFα) and interleukin-1 beta (IL-1β) levels. Results Passiflora edulis (250 mg/kg) administered by Intraperitoneal Route (i.p.) inhibited the leukocyte, neutrophils, myeloperoxidase, nitric oxide, TNFα and IL-1β levels (P Conclusion Several mechanisms, including the inhibition of pro-inflammatory cytokines (TNFα, IL-1β), enzyme (myeloperoxidase) and mediators (bradykinin, histamine, substance P, nitric oxide) release and/or action, appear to account for Passiflora edulis's actions.
