CameronAbstract:Comparative global proteome analyses were performed on Leptospira interrogans serovar Copenhageni grown under conventional in vitro conditions and those mimicking in vivo conditions (iron limitation and serum presence). Proteomic analyses were conducted using iTRAQ and LC-ESI-tandem mass spectrometry complemented with two-dimensional gel electrophoresis and MALDI-TOF mass spectrometry. A total of 563 proteins were identified in this study. Altered expression of 65 proteins, including upregulation of the L. interrogans virulence factor Loa22 and 5 novel proteins with homology to virulence factors found in other pathogens, was observed between the comparative conditions. Immunoblot analyses confirmed upregulation of 5 of the known or putative virulence factors in L. interrogans exposed to the in vivo-like environmental conditions. A new serovar in serogroup Manhao of Leptospira interrogans-Serovar Hevan
Chinese journal of zoonoses, 2004Co-Authors: Yang Wen-yingAbstract:A strain of Leptospira interrogans isolated from blood of a patient in Yunnan province in 1990 was assayed and classified by using the classic serotyping methods and the pulsed-field gel electrophoresis (PFGE) in order to determine its serovar. This strain was identified as serogroup Manhao of L. interrogans, and it was different with the 5 known serovars in this serogroup. It is a new serovar in serogroup Manhao of L. interrogans, and named as Serovar Heyan with a representative strain L231.

Sequence analysis of the gene encoding ompL_1 and flaB_2 from Leptospira interrogans serovar lai and Leptospira interrogans serovar australis
Chinese journal of microbiology and immunology, 2002Co-Authors: Dai BaomiAbstract:Objective To construct recombinant plasmids from Leptospira interrogans serovar lai strain 017 and Leptospira interrogans serovar australis strain 607 consisted of an outer membrane protein gene (ompL 1) and a flagellin B gene(flaB 2). The ompL 1 and flaB 2 gene were analyzed by DNA sequencing. Methods The DNA fragments of ompL 1 and flaB 2 were respectively amplified by PCR, then about cloned into the down stream of a T7 promoter of vector pcDNA3.1/Myc His(+), recombinant plasmids of the antigen gene were constructed, and analyzed by DNA sequencing. Results Analysis of the DNA sequence of ompL 1 from L.interrogans lai strain 017 and L.interrogans australis strain 607 showed that the similarity of nucleotide sequence was 98.85%(949/960), variation was 1.15%(11/960); About flaB 2, the similarity was 96.94%(823/849), variation was 3.06%(26/849), the sequences of each were highly conserved. Conclusion Sequences analysis of ompL 1 and flaB 2 from Leptospira interrogans serovar lai strain 017 respectively revealed a high level of identity with that from Leptospira interrogans serovar australis strain 607. Isolation and identification of a serovar canicola of Leptospira interrogans
Medicial Journal of National Defending Forces in Southwest China, 2005Co-Authors: Fan Quan-shuiAbstract:Objective: To isolate and identify the pathogen of a dog like canicola faver, so as to provide some scientific basis for the diagnosis. Methods: The dog was anatomized and examined. Its urine was inoculated into medium and cultured, Leptospira was isolated and identified. Then the genic primer of outer membrane proteins of the Leptospira was used by PCR. Results: A strain of Leptospira interrogans was isolated from dog's urine and detected by PCR. This strain of Leptospira interrogans was classified as serovar canicola in serogroup canicola. Conclusion: A strain of serovar canicola in serogroup canicola of Leptospira interrogans was isolated from a dog.

First isolation and characterization of Leptospira interrogans serogroup Australis from swine in Brazil

Loureiro, Sylvie Brémont, Marco Alberto Medeiros, Pascale Bourhy, Walter LilenbaumAbstract:The purpose of this study was to report the first recovery and characterization of Leptospira interrogans (serogroup Australis) from urine of swine in Brazil. The isolate was studied by serogrouping, MLVA, PGFE, and partial sequencing of rrs and secY. It was serogrouped as serogroup Australis, probably serovar Bratislava (titre 1,600), and sequenced as Leptospira interrogans. The MLVA and PGFE profiles also suggested the isolate as serovar Bratislava, since they were indistinguishable from reference strains Balico and Jez Bratislava. This is the first Leptospira interrogans serogroup Australis isolate, probably serovar Bratislava, obtained in Brazil.
