Journal of Ethnopharmacology, 2016Co-Authors: Paul Owusu Donkor, Ying Chen, Liqin Ding, Feng QiuAbstract:Abstract Ethnopharmacological relevance Ligusticum species (Umbelliferae) have been widely used in traditional Chinese medicine, Korean folk medicine and Native American medicine for their medicinal and nutritional value. Decoctions of the rhizomes are used in treatment and prophylaxis of migraine, anemia and cardiovascular conditions including stroke. Aim of study This review is intended to fully compile the constituents of locally and traditionally used Ligusticum species, present their bioactivities and highlight potential leads for future drug design, and thus, provide a reference for further research and application of these species. Emphasis is also placed on current trends in the pharmacokinetic studies of the major constituents. Methods The literature discussed is derived from readily accessible papers spanning the early 1990 s to the end of 2015. Information was collected from journals, books and online searches (Google Scholar, PubMed, ScienceDirect, SciFinder, Springerlink and CNKI). Results The major phytoconstituents, 154 of which are presented in this review, include alkaloids, phthalides and phenolic acids. The crude extracts and isolated constituents have exhibited a wide range of in vitro and in vivo pharmacologic effects, including cardioprotective, antioxidant, anti-inflammatory and neuroprotective activities. The bioactive alkaloid tetramethylpyrazine (TMP) has attracted the most attention for its potent effect on calcium channels, anti-platelet as well as anti-inflammatory effects. Pharmacokinetic studies of major constituents have also been summarized. Conclusion The pthalides, organic acids and alkaloids of Ligusticum species have emerged as a good source of traditional medicines for the management of cardio- and cerebrovascular conditions, inflammation and neurogenerative disorders. The species discussed in this review have demonstrated wide pharmacological actions and have great potential to yield multipotent drugs if challenges such as poor bioavailability, solubility and toxicological profiles are addressed. Apart from the rhizomes, pharmacological activities of other botanical parts also need to be studied further. Expansion of research to cover other species in the Ligusticum genus would provide more opportunities for the discovery of new bioactive principles.

(Umbelliferae) is a traditional Chinese medicine (TCM) herb for the prevention and treatment of inflammatory and cardiovascular diseases. However, the role of phthalide lactones from Ligusticum chuanxiong in the therapeutic actions is not yet fully understood. In the present study, two phthalide lactones from the herb, Z-ligustilide and senkyunolide A, were identified and characterized as inhibitors of lipopolysaccharide (LPS)-induced TNF-alpha production in monocytes. The results of gene expression studies showed that the observed TNF-alpha suppression was related to their inhibitory activity on TNF-alpha mRNA transcription. Furthermore, the two phthalides exhibited significant suppressive effects on TNF-alpha-mediated nuclear factor-kappaB (NF-kappaB) activation in reporter gene assays. Taken together, the results suggest that Z-ligustilide and senkyunolide A may have potential applications in the treatment of inflammation and related diseases based on their inhibitory activity on TNF-alpha production and TNF-alpha bioactivity. New insights into the therapeutic basis of the TCM herb, Ligusticum chuanxiong, are presented.

Isolation and Identification of Antagonistic Endophytic Actinomycetes Against Root Rot Disease in Ligusticum chuanxiong
Journal of Chinese medicinal materials, 2016Co-Authors: Chan-chun Lin, Zhu-yun Yan, Hai Wang, Wei LiuAbstract:Objective To get an understanding of the microflora of endophytic actinomycetes in Ligusticum chuanxiong,and to obtain the resource of antagonistic strains against Ligusticum chuanxiong root rot disease. Methods Actinomycetes in stem nodes and rhizomes of Ligusticum chuanxiong were isolated and purified by a series of means,namely tissue block method, homogenate technique and plat streaking method. Further, dual culture and inhibition zone method were employed to test the antagonistic activity of these strains. To identify strains with potential precisely,both microscopic observation and DNA sequencing were conducted. Results 83 strains of Ligusticum chuanxiong endophytic actinomycetes were identified, which consisted of 13 species groups. Among all of them,species identified as Streptomyces scopuliridis( KF600747. 1), Streptomyces griseorubiginosus( AB706352. 1) and Streptomyces agglomeratus( LC055413. 1) showed antagonistic activities against four kinds of pathogenic fungi of Ligusticum chuanxiong root rot disease. Conclusion Actinomycetes which belong to Griseorubroviolaceus groups of streptomyce are potential biocontrol microbes to Ligusticum chuanxiong root rot,showing importance to the production of Chuanxiong Rhizoma.

Identification of Pathogens Causing Root Rot Disease on Ligusticum chuanxiong in Sichuan
Journal of Chinese medicinal materials, 2015Co-Authors: Zhu-yun Yan, Yin Lan, Shen Xiaofeng, Hai WangAbstract:OBJECTIVE To identify the pathogens causing root rot disease on Ligusticum chuanxiong from the main producing areas in Sichuan, and to provide gist of management of this disease in the future. METHODS The diseased rhizomes of Ligusticurn chuanxiong with root rot symptom were sampled in two main producing areas in Sichuan. According to Koch's postulate, the pathogenicity on Ligusticum chuanxiong rhizomes of the isolates was confirmed. The representative isolates were identified based on their morphological characteristics and ribosomal DNA internal transcribed spacer(rDNA-ITS) sequences. RESULTS The isolates(f2-16, 3-2, f4-19 and f5-7)which caused root rot disease held high homology reached 99%, 99%, 100% and 100% respectively, when compared with GenBank sequences GQ229075, JQ796755, JN232136 and FJ481024. CONCLUSION The causing agents of root rot disease on Ligusticum chuanxiong belong to Fusarium solani, Fusarium oxysporum, Plectosphaerella cucumerina and Phoma glomerata. Pathogens from different regions are varied.
