Fabrication of nano-hydroxyapatite on electrospun silk fibroin nanofiber and their effects in osteoblastic behavior.

The morphologies of both nonmineralized and mineralized nanofibers were analyzed using a field-emission scanning electron microscopy (FESEM). The crystallographic phases of the nHA were analyzed using X-ray diffraction (XRD). Fourier transform infrared (FTIR) spectrophotometer and thermogravimetry analyses (TGA) were employed to determine the type of functional groups and the amount of nHA presenting in the silk/nHA biocomposite nanofibers, respectively. The osteoblastic activities of this novel nanofibrous biocomposite scaffold were also investigated by employing osteoblastic-like MC3T3-E1 Cell Line. The Cell functionality such as alkaLine phosphatase (ALP) activity was ameliorated on mineralized nanofibers. All these results indicated that this silk/nHA biocomposite scaffold material may be a promising biomaterial for bone tissue engineering.

In this study, two kinds of biocomposites based on regenerated silk fibroin are fabricated by electrospinning and post-treatment processes, respectively. Firstly, regenerated silk fibroin/tetramethoxysilane (TMOS) hybrid nanofibers with high hydrophilicity are prepared, which is superior for fibroblast attachment. The electrospinning process causes adjacent fibers to 'weld' at contact points, which can be proved by scanning electron microscope (SEM). The water contact angle of silk/tetramethoxysilane (TMOS) composites shows a sharper decrease than pure regenerated silk fibroin nanofiber, which has a great effect on the early stage of Cell attachment behavior. Structure, biodegradation behavior and cytotoxicity of alkali-containing alkaLine-earth phosphosilicate glasses

Tulyaganov, José M.f. FerreiraAbstract:We report on the effect of sodium on the structure, chemical degradation and bioactivity of glasses in the CaO-MgO-SiO2-P2O5-CaF2 system. The (29)Si and (31)P magic angle spinning-nuclear magnetic resonance spectroscopy of melt-quenched glasses with varying Na2O/MgO ratios exhibit a silicate glass network with the dominance of Q(2)(Si) units and phosphorus mainly forming orthophosphate species. Sodium incorporation in the glasses did not induce a significant structural change in the silicate network, while it did influence the phosphate environment due to its lower ionic field strength in comparison with that of magnesium. The apatite forming ability of glasses has been investigated by immersion of glass powders in simulated body fluid (SBF) for time durations varying between 1h and 7 days while their chemical degradation has been studied in Tris-HCl in accordance with ISO-10993-14. Increasing Na(+)/Mg(2+) ratio caused a decrease in the chemical durability of glasses and in the apatite forming ability especially during initial steps of interaction between glass and SBF solution. The Cellular responses were observed in vitro on bulk glass samples using mouse-derived pre-osteoblastic MC3T3-E1 Cell Line. The preliminary study suggested that the increasing alkali-concentration in glasses led to cytotoxicity in the Cell culture medium.

Synthesis, mechanical and biological characterization of ionic doped carbonated hydroxyapatite/β-tricalcium phosphate mixtures.

FerreiraAbstract:The influence of ionic substituents in calcium phosphates intended for bone and tooth replacement biomedical applications is an important research topic, owing to the essential roles played by trace elements in biological processes. The present study investigates the mechanical and biological evaluation of ionic doped hydroxyapatite/β-tricalcium phosphate mixtures which have been prepared by a simple aqueous precipitation method. Heat treating the resultant calcium phosphates in a carbonated atmosphere led to the formation of ionic doped carbonated hydroxyapatite/β-tricalcium phosphate mixtures containing the essential ions of biological apatite. The structural analysis determined by Rietveld refinement confirmed the presence of hydroxyapatite as the main phase, together with a considerable amount of β-tricalcium phosphate. Such phase assemblage is essentially due to the influence of substituted ions during synthesis. The results from mechanical tests proved that carbonate substitutions are detrimental for the mechanical properties of apatite-based ceramics. In vitro proliferation assays of osteoblastic-like Cells (MC3T3-E1 Cell Line) to powders revealed that carbonate incorporation can either delay or accelerate MC3T3 proliferation, although reaching the same proliferation levels as control Cells after 2 weeks in culture. Further, the powders enable pre-osteoblastic differentiation in a similar manner to control Cells, as indirectly measured by ALP activity and Type-I collagen medium secretion.

Biological responses of brushite-forming Zn- and ZnSr- substituted beta-tricalcium phosphate bone cements.

FerreiraAbstract:The core aim of this study was to investigate zinc (Zn)- and zinc and strontium (ZnSr)-containing brushite-forming beta-tricalcium phosphate (TCP) cements for their effects on proliferation and differentiation of osteoblastic-like Cells (MC3T3-E1 Cell Line) as well as for their in vivo behaviour in trabecular bone cylindrical defects in a pilot study. In vitro proliferation and maturation responses of MC3T3-E1 osteoblastic-like Cells to bone cements were studied at the Cellular and molecular levels. The Zn- and Sr-containing brushite cements were found to stimulate pre-osteoblastic proliferation and osteoblastic maturation. Indeed, MC3T3-E1 Cells exposed to the powdered cements had increased proliferative rates and higher adhesiveness capacity, in comparison to control Cells. Furthermore, they exhibited higher alkaLine phosphatase (ALP) activity and increased Type-I collagen secretion and fibre deposition into the extraCellular matrix. Proliferative and collagen deposition properties were more evident for Cells grown in cements doped with Sr. The in vivo osteoconductive propertiesof the ZnCPC and ZnSrCPC cements were also pursued. Histological and histomorphometric analyses were performed at 1 and 2 months after implantation, using carbonated apatite cement (Norian SRS) as control. There was no evidence of cement-induced adverse foreign body reactions, and furthermore ZnCPC and ZnSrCPC cements revealed better in vivo performance in comparison to the control apatite cement. Additionally, the presence of both zinc and strontium resulted in the highest rate of new bone formation. These novel results indicate that the investigated ZnCPC and ZnSrCPC cements are both biocompatible and osteoconductive, being good candidate materials to use as bone substitutes.

Fabrication of core-sheath structured fibers for model drug release and tissue engineering by emulsion electrospinning

Here we present a novel and effective emulsion electrospinning method for obtaining fluorescein isothiocyanate-dextran (FITC-dextran)/poly(lactic-co-glycolic acid) (PLGA) and type I collagen/PLGA fibrous composite scaffolds. Core-sheath structured fibers with average diameters of 665 nm for FITC-dextran/PLGA and 567 nm for collagen/PLGA were successfully fabricated. In vitro-release profile shows sustained release of encapsulated FITC-dextran from FITC-dextran/PLGA fibers for as long as 7 weeks. The osteoblastic activity of the collagen/PLGA nanofibrous scaffold was investigated employing the osteoblastic-like MC3T3-E1 Cell Line. The results of the lactate dehydrogenase assay suggested exCellent cytocompatibility. Cell proliferation and alkaLine phosphatase activity were also ameliorated on this emulsion-electrospun collagen/PLGA fibrous scaffold. All the results indicated that this composite scaffold could support the early stages of osteoblast behavior as well as the immediate/late stages. The emulsion electrospinning process has potential for application in drug-release devices and as a 3-D scaffold in bone regeneration.

The effect of strontium ranelate on titanium particle-induced periprosthetic osteolysis regulated by WNT/β-catenin signaling in vivo and in vitro.

The wear particles trigger a chronic inflammation response in the periprosthetic tissue and turn over the bone balance to bone resorption. The present study aimed to investigate the possible effect and mechanism of strontium ranelate (SR), a clinically safe drug for osteoporosis, on particle-induced periprosthetic osteolysis. Thirty-six female C57BL/6j mice underwent tibial Ti-nail implantation to establish an animal model of aseptic loosening. After 12 weeks, micro-CT results showed that strontium ranelate could inhibit periprosthetic bone resorption. In vitro, Ti particles were used to stimulate RAW264.7 Cell Line to collect conditioned medium, and co-culture MC3T3-E1 Cell Line with conditioned medium to establish a Cell model of aseptic loosening. The results of alkaLine phosphatase (ALP) detection, immunofluorescence, and flow cytometry demonstrated that strontium ranelate could regulate the expression of OPG/RANKL, promote differentiation and mineralization, and inhibit apoptosis in osteoblasts. Moreover, we revealed that SR's exerted its therapeutic effect by down-regulating sclerostin, thereby activating the Wnt/β-catenin signal pathway. Therefore, this research suggests that strontium ranelate could be a potential drug for the prevention and treatment of particle-induced aseptic loosening post-TJA.

Heterodimeric BMP-2/7 Antagonizes the Inhibition of All-Trans Retinoic Acid and Promotes the Osteoblastogenesis
PloS one, 2013Co-Authors: Yuanna Zheng, Xiao Zhang, Jing GuoAbstract:Hypervitaminosis A and alcoholism can result in a low mineral density and compromised regenerative capacity of bone, thus delaying implant osteointegration. The inhibitory effect of all-trans retinoic acid on osteoblastogenesis is considered to be one of the mechanisms. We hypothesized that heterodimeric bone morphogenetic protein-2/7 could antagonize all-trans retinoic acid and enhance osteoblastogenesis, with an aim to accelerate and enhance bone regeneration and implant osteointegration. We applied 5 ng/ml or 50 ng/ml bone morphogenetic protein-2/7 to restore the osteoblastogenesis of pre-osteoblasts (MC3T3-E1 Cell Line) that was inhibited by 1 µM all-trans retinoic acid. We evaluated the efficacy by assessing Cell numbers (proliferation), alkaLine phosphatase activity (a marker for early differentiation), osteocalcin (a marker for late differentiation), calcium deposition (a marker for final mineralization) and the expression of osteoblastogenic genes (such as Runx2, Collagen Ia, alkaLine phosphatase and osteocalcin) at different time points. All-trans retinoic acid significantly inhibited the expression of all the tested osteoblastogenic genes and proteins except alkaLine phosphatase activity. In the presence of ATRA, 50 ng/ml bone morphogenetic protein-2/7 not only completely restored but also significantly enhanced all the osteoblastogenic genes and proteins. On the 28(th) day, mineralization was completely inhibited by all-trans retinoic acid. In contrast, 50 ng/ml BMP-2/7 could antagonize ATRA and significantly enhance the mineralization about 2.5 folds in comparison with the control treatment (no ATRA, no BMP2/7). Heterodimeric bone morphogenetic protein-2/7 bears a promising application potential to significantly promote bone regeneration and implant osteointegration for the patients with hypervitaminosis A and alcoholism.
