    Abstract:

    Lycopus lucidus Turcz has been used as a traditional phytomedicine for Menstrual Disorder, amenorrhea, Menstrual cramps, inflammation and cardiovascular diseases. However, there is not enough information about identification and quantification for the chemical constituents of L. lucidus Turcz. In this work, a simple, rapid and sensitive UHPLC-Q-TOF-MS method was developed for characterization and identification of the phytochemical compositions in L. lucidus Turcz in negative ion mode. A total of 37 compounds, including 15 phenolic acids, 12 flavonoids, three triterpenoids and seven organic acids were tentatively characterized and identified by means of the retention time, accurate mass and characteristic fragment ions. Thirteen compounds were reported for the first time in L. lucidus Turcz. Among of them, 11 compounds were further quantified by multiple reactions monitoring. The results showed good performance with respect to linearity (r > 0.9959), repeatability (RSD < 2.6%), intra- and inter-day precision (RSD < 3.2%), recovery (93.1–104.9%), and lower limit of quantification (5–50 ng/mL). Subsequently, the results were analyzed and classified by hierarchical cluster analysis. The research could be applied for identification and quality evaluation for L. lucidus Turcz.

    Abstract Lycopus lucidus Turcz has been used as a kind of edible and medicinal material in eastern Asian countries. It has various bioactivities, including treatment of Menstrual Disorder, amenorrhea, Menstrual cramps, inflammation, and cardiovascular diseases. However, the in vivo metabolism of L. lucidus Turcz extract is still not well described. In this study, L. lucidus Turcz extracts were administered to rats. Urine and fecal samples were collected at the difference periods (0–12 h, 12–24 h, and 24–36 h). Ultra-high performance liquid chromatography coupled with electrospray ionization tandem quadrupole time-of-flight mass spectrometry (UHPLC−Q-TOF-MS) method was developed to characterize and identify the metabolites. A total of 17 metabolites in feces and 19 metabolites in urine were tentatively identified by means of accurate mass and characteristic fragment ions. The results show that glucuronidation and sulfation are the major metabolic reactions. This study is the first reported analysis and characterization of the metabolites and the proposed metabolic pathways of bioactive components might provide further understanding of the metabolic fate of the chemical constituents after oral administration of L. lucidus Turcz extract in rats.

    Abstract:

    Oocyte donation or egg donation is a process in which one woman donates eggs (oocytes) to another women in order to conceive. The process involves in vitro fertilization and it is seen as third party reproduction in the assisted reproductive technology. The oocytes from the donor are combined with the spermatozoa from the recipient to produce embryos. However, the treatment has some risk factors that afterwards need to be taken care of. One of the complications is delayed or stopped period and the acupuncture treatment can efficiently solve this problem. The treated patient is a 34 year old woman with 3 months delay in the period after receiving hormone therapy for egg donation. The patient was treated with normal needle acupuncture on the meridians of spleen, kidney, pericardium, stomach, liver, large intestine, bladder, ren mai and du mai. Treatments were made indoor, with duration of 35-40 minutes both sides of the body. In the treatment were used fine sterile disposable needles. After only one treatment the period was back and normalized. The patient has made 6 treatments. After the 6thtreatment the second period was normal again. Acupuncture treatment as part of the 5000 years old Traditional Chinese Medicine, as in many other Menstrual Disorders, it is as well successful treatment in Menstrual Disorder after egg donation and gives very satisfying and positive results in a very short time

    Abstract:

    A high prevalence of Menstrual disturbance has been reported among teenage girls, and research shows that there are delays in diagnosis of endometriosis among young girls. Using data from the Menstrual Disorder of Teenagers Survey (administered in 2005 and 2016), we propose a Gaussian copula model with graphical lasso prior to identify cohort differences in Menstrual characteristics and to predict endometriosis. The model includes random effects to account for clustering by school, and we use the extended rank likelihood copula model to handle variables of mixed-type. The graphical lasso prior shrinks the elements in the precision matrix of a Gaussian distribution to encourage a sparse graphical structure, where the level of shrinkage is adaptable based on the strength of the conditional associations among questions in the survey. Applying our proposed model to the Menstrual Disorder data set, we found that Menstrual disturbance was more pronouncedly reported over a decade, and we found some empirical differences between those girls with higher risk of developing endometriosis and the general population.
