  • Preliminary results on the conservation of Lamyropsis Microcephala (Moris) Dittrich & Greuter (Asteraceae), a threatened endemic species of the Gennargentu massif, Sardinia (Italy)
    Flora Montiberica, 2020
Co-Authors: Gianluigi Bacchetta, Giuseppe Fenu, Efisio Mattana, Tiziana Ulian
    Abstract:

EnglishLamyropsis Microcephala (Moris) Dittrich & Greuter (Compositae) is a perennial species, narrow endemic of Sardinia (Italy) and known only in two sites of the Gennargentu massif. The aims of this study are to investigate characteristics of the ecology and biology of the species which might be important for its conservation and reassess its conservation status. Here we show the preliminary results of this study, with a focus on the abundance and distribution of the species, its reproductive capacity and the levels of threats. From the data collected in the field it appears that the population had been fragmented into two subpopulations. The total population covered an area of around 12 hectares and consisted of around 2500 individuals. The reproductive biology of the species, in particular the low production of fertile seeds and absence of plantulae (recruitment) in the field, seems to be one of the main biological causes of the threatened status of the species. Increases in extensive grazing and tourism, linked to the skiing and trekking activities, have been recognized as the major anthropogenic threats to the species. The preliminary results of this study confirm the conservation status of L. m-icrocephala as Critically Endangered.

  • the essential oil composition of cunila Microcephala and cunila fasciculata
    Phytochemistry, 1997
Co-Authors: Sergio A L Bordignon, Eloir Paulo Schenkel, Volker Spitzer
    Abstract:

Abstract The essential oils of cultivated and wild-growing Cunila Microcephala and C. fasciculata from southern Brazil were analysed by GC, GC-mass spectrometry and 13 C NMR spectroscopy. The main constituent in both species was found to be menthofuran (82.3–85.1% in C. Microcephala and 71.6–76.4% in C. fasciculata ). Other compounds in significant amounts were limonene (2.1–3.8% in C. Microcephala and 8.6–11.2% in C. fasciculata ) and β-caryophyllene (3.3–3.9% in C. Microcephala and 3.5–4.2% in C. fasciculata ). The chemotaxonomic and toxicologic aspects of menthofuran accumulation are discussed.

  • Neuropädiatrische Differenzialdiagnostik der Mikrozephalie im Kindesalter
    medizinische genetik, 2016
Co-Authors: Maja Von Der Hagen, Horst Von Bernuth, Julia B Hennermann, Rainer John, Birgit Spors, Angela M. Kaindl
    Abstract:

Microcephaly affects 2–3 % of the population and is often associated with intellectual disability. The underlying reduction in brain volume can result from various exogenous factors or genetic causes. Microcephaly remains a poorly defined condition lacking both uniform diagnostic approaches and classification. A definite etiological diagnosis is the key to predict the prognosis, identify co-morbidities and offer genetic counseling. In addition, the identification of the underlying cause increases our knowledge of brain development and the clinical spectrum of microcephaly. We propose a diagnostic approach for children with microcephaly from a pediatric neurologist point of view.

  • novel alternative splice variants of mouse cdk5rap2
    PLOS ONE, 2015
Co-Authors: Nadine Kraemer, Ethiraj Ravindran, Gerda Neubert, Lina Issajahns, Olaf Ninnemann, Shyamala Mani, Angela M. Kaindl
    Abstract:

Autosomal recessive primary microcephaly (MCPH) is a rare neurodevelopmental disorder characterized by a pronounced reduction of brain volume and intellectual disability. A current model for the microcephaly phenotype invokes a stem cell proliferation and differentiation defect, which has moved the disease into the spotlight of stem cell biology and neurodevelopmental science. Homozygous mutations of the Cyclin-dependent kinase-5 regulatory subunit-associated protein 2 gene CDK5RAP2 are one genetic cause of MCPH. To further characterize the pathomechanism underlying MCPH, we generated a conditional Cdk5rap2 LoxP/hCMV Cre mutant mouse. Further analysis, initiated on account of a lack of a microcephaly phenotype in these mutant mice, revealed the presence of previously unknown splice variants of the Cdk5rap2 gene that are at least in part accountable for the lack of microcephaly in the mice.

  • diagnostic approach to microcephaly in childhood a two center study and review of the literature
Developmental Medicine & Child Neurology, 2014
Co-Authors: Maja Von Der Hagen, Mark Pivarcsi, Juliane Liebe, Horst Von Bernuth, Nataliya Didonato, Julia B Hennermann, Christoph Buhrer, Dagmar Wieczorek, Angela M. Kaindl
    Abstract:

Aim The aim of this study was to assess the diagnostic approach to microcephaly in childhood and to identify the prevalence of the various underlying causes/disease entities. Method We conducted a retrospective study on a cohort of 680 children with microcephaly (399 males, 281 females; mean age at presentation 7-8mo, range 1mo-5y) from patients presenting to Charite - University Medicine Berlin (n=474) and University Hospital Dresden (n=206). Patient discharge letters were searched electronically to identify cases of microcephaly, and then the medical records of these patients were used to analyze parameters for distribution. Results The putative aetiology for microcephaly was ascertained in 59% of all patients, leaving 41% without a definite diagnosis. In the cohort of pathogenetically defined microcephaly, genetic causes were identified in about half of the patients, perinatal brain damage accounted for 45%, and postnatal brain damage for 3% of the cases. Microcephaly was associated with intellectual impairment in 65% of participants, epilepsy was diagnosed in 43%, and ophthalmological disorders were found in 30%. Brain magnetic resonance imaging revealed abnormalities in 76% of participants. Interpretation Microcephaly remains a poorly defined condition, and a uniform diagnostic approach is urgently needed. A definite aetiological diagnosis is important in order to predict the prognosis and offer genetic counselling. Identifying gene mutations as causes of microcephaly increases our knowledge of brain development and the clinical spectrum of microcephaly. We therefore propose a standardized initial diagnostic approach to microcephaly.

  • What’s the hype about CDK5RAP2?
Cellular and Molecular Life Sciences, 2011
Co-Authors: Nadine Kraemer, Lina Issa, Stefanie C. R. Hauck, Olaf Ninnemann, Shyamala Mani, Angela M. Kaindl
    Abstract:

Cyclin dependent kinase 5 regulatory subunit-associated protein 2 (CDK5RAP2) has gained attention in the last years following the discovery, in 2005, that recessive mutations cause primary autosomal recessive microcephaly. This disease is seen as an isolated developmental defect of the brain, particularly of the cerebral cortex, and was thus historically also referred to as microcephalia vera . Unraveling the pathomechanisms leading to this human disease is fascinating scientists because it can convey insight into basic mechanisms of physiologic brain development (particularly of cortex formation). It also finds itself in the spotlight because of its implication in trends in mammalian evolution with a massive increase in the size of the cerebral cortex in primates. Here, we provide a timely overview of the current knowledge on the function of CDK5RAP2 and mechanisms that might lead to disease in humans when the function of this protein is disturbed.
