Sing (FV), Lentinus edodes (LE)and Agaricus bispo-rus Sing (AB) on the proliferation of human hepatoma SMMC-7721 cells in vitro and on mouse implanted S-180 tumors in vivo.METHODS:The polysaccharide extracts were isolated from the fruit bodies of FV, LE and AB by the methods of hot-water extraction,Sevag's removal of proteins, ethanol precipitation, trypsin digestion and ethanol fractional precipitation. Human hepatoma SMMC-7721 cells were treated with 50mg/L polysaccharide extracts, and the Mitosis Index, mitochondria activity and cell proliferation were detected at different times in both control and experimental groups. The mice with S-180 implanted tumors were injected with the polysaccharide extracts at 24mg/kg body weight for 9d and the tumor weight was measured on the 15th day.RESULTS:The Mitosis Index of hepatoma cells in vitro could be significantly decreased by treatment with the polysaccharide extracts from the three kinds of edible fungi (P< 0.005). The cell numbers and mitochondria activity of SMMC-7721 cells treated with polysaccharide extracts were lower than those in control groups (P<0.005). The inhibition rates of polysaccharide extracts against implanted S-180 tumors in mice were 52.8%, 56.6% and 51.9% respectively compared with that in control groups.CONCLUSION:The polysaccharide extracts from the three kinds of edible fungi could inhibit not only the cultured malignant cells in vitro but also implanted S-180 tumor in vivo.

Presently, it is unknown whether BIO can influence the derivation of porcine embryonic germ (EG) cells. In this study, porcine primordial germ cells (PGCs) were isolated from gonads of 24- and 28-day embryos, and were then treated with BIO either individually or in combination with other cytokines (stem cell factor (SCF), leukemia inhibitory factor (LIF), and fibroblast growth factor (FGF); abbreviated as '3F'), and the effects of the treatment on the proliferation ability of porcine PGCs at early stage were examined using 5-bromo-2-deoxyuridine (Brdu) immunostaining assay. After continuous culture, the effects on the efficiency of porcine undifferentiated EG cells in the third passage and differentiated EG cells from embryoid bodies were examined as well. The results obtained through the observation of the Brdu-labeled PGCs indicated that BIO in combination with 3F resulted in a significant increase in the Mitosis Index, and also indicated that the BIO in combination with 3F had a higher efficiency in promoting the formation of porcine EG colony derived from porcine day 24 PGCs than BIO used either individually or in combination with LIF. In addition, BIO in combination with 3F exhibited the apparent anti-differentiation activity by reversing the differentiated EG cells to the undifferentiated status. Our results demonstrate that BIO in combination with SCF, LIF, and FGF could significantly contribute to the establishment of a porcine EG cell colony and maintain the undifferentiated status.

2009Co-Authors: Elena Bonciu, P IancuAbstract:The proportionality cell division intensity to different genotypes is kept constant along the different ontogenetic phases, therefore, the mitotic activity determination in the 1-2 cm sunflower radicules (2-3 days after germination), could contribute to cell division particularities revealing to homozygote and heterozygote sunflower genotypes and their correlation with the heterosis. In order to determine sunflower mitotic activity and the mitotic Index phases, were investigated four genotypes. They were represented by two Romanian hybrids (Saturn and Minunea) and two foreign hybrids (Flavia and PR63A82). The mitotic activity analyzed to these genotypes, expressed by mitotic Index, was variable from 14,8% to 23,0% limits. From this point of view, it was found superiority of the mitotic Index percentage to Saturn hybrid with the 35,7% bigger than Minunea hybrid, with the 27,8% bigger than Flavia hybrid and with the 16% more than the PR63A82 sunflower hybrid.
