Dynamics of the striped bass (Morone Saxatilis) ovary proteome reveal a complex network of the translasome.

SullivanAbstract:We evaluated changes in the striped bass (Morone Saxatilis) ovary proteome during the annual reproductive cycle using label-free quantitative mass spectrometry and a novel machine learning analysis based on K-means clustering and support vector machines. Modulated modularity clustering was used to group co-variable proteins into expression modules and Gene Ontology (GO) biological process and KEGG pathway enrichment analyses were conducted for proteins within those modules. We discovered that components of the ribosome along with translation initiation and elongation factors generally decrease as the annual ovarian cycle progresses toward ovulation, concomitant with a slight increase in components of the 26S-proteasome. Co-variation within more than one expression module of components from these two multi-protein complexes suggests that they are not only co-regulated, but that co-regulation occurs through more than one sub-network. Isolation and characterization of 149 novel microsatellite DNA markers for striped bass, Morone Saxatilis, and cross-species amplification in white bass, Morone chrysops, and their hybrid

SullivanAbstract:To support detailed genetic analysis of striped bass ( Morone Saxatilis ) and white bass ( Morone chrysops ), we isolated 153 microsatellite loci from repeat-enriched striped bass DNA libraries. Of these, 147 markers amplified in striped bass (average 4.7 alleles per locus) and 133 in white bass (average 2.2 alleles per locus). One hundred twenty-two markers amplified in their hybrid. Development of new microsatellite markers will facilitate evaluations of genetic structure in wild populations and will support pedigree analysis and linkage mapping for selective breeding.

Activity imposes two constraints: maximum swimming speed limits habitats that can be occupied for short periods of time, while the metabolic cost of swimming limits the habitats that are suitable for long-term residence. We measured the energetic consequences of activity and maximum swimming speeds of young-of-the-year striped bass (Morone Saxatilis), a species that overwinters in tidal estuaries. The energetic cost of swimming was determined from energy changes in unfed fish forced to swim at various speeds, while energy changes in fed fish provided a measure of their ability to offset swimming costs through feeding. In high-velocity treatments, mortality was size-dependent and appeared to be related to fatigue rather than to depletion of energy reserves. Newly hatched larvae were obtained from six native anadromous stocks spanning most of the natural range of striped bass (Florida to Nova Scotia). Growth experiments were conducted under conditions of unlimited food at three temperatures (17, 21, 28°C) and commenced after larvae had exhausted maternal energy resources (i.e., yolk and oil) and had begun to metamorphose. Mixed-model nested ANOVA demonstrated that length and dry weight growth differed significantly among latitudes in most comparisons. A neomycin dose response (0, 1, 2 and 5 mmol l(-1)) was evaluated for neuromast ablation of bass aged 10, 13, 17 and 20 dph. Using these same age groups, the ability of bass to catch Artemia salina prey in both dark and light tank-based feeding trials was compared between larvae with neuromasts ablated using neomycin (5 mmol l(-1)) and controls. Transcriptome annotation and marker discovery in white bass (Morone chrysops) and striped bass (Morone Saxatilis)

Adam Fuller, Eric PeatmanAbstract:Summary Striped bass (Morone Saxatilis) and white bass (Morone chrysops) are the parental species of the hybrid striped bass, a major U.S. aquaculture species. Currently, genomic resources for striped bass, white bass, and their hybrid lag behind those of other aquaculture species. Current resources consist of a medium-density genetic linkage map and a well-annotated ovarian transcriptome. A well-annotated transcriptome from across striped bass and white bass tissues is needed to advance both broad-based RNA-seq studies of gene expression as well as aid in more targeted studies of important genes and pathways critical for reproductive physiology and immunity. Here, we carried out Illumina-based transcriptome sequencing and annotation in both species utilizing the trinity and trinotate packages. The assembled Moronid reference transcriptomes and identified SSRs and SNPs should advance ongoing studies of reproduction, physiology, and immunology in these species and provide markers for broodstock management and selection.
