    Abstract:

    The paper deals with the imposition of local stress constraints in topology optimization. The aim of the work is to analyze the performances of an alternative methodology to the ε -relaxation introduced in Cheng and Guo (Struct Optim 13:258–266, 1997 ), which handles the well-known stress singularity problem. The proposed methodology consists in introducing, in the SIMP law used to apply stress constraints, suitable penalty exponents that are different from those that interpolate stiffness parameters. The approach is similar to the classical one because its main effect is to produce a relaxation of the stress constraints, but it is different in terms of convergence features. The technique is compared with the classical one in the Context of stress-constrained minimum-weight topology optimization. Firstly, the problem is studied in a modified truss design framework, where the arising of the singularity phenomenon can be easily shown analytically. Afterwards, the analysis is extended to its Natural Context of topology bidimensional problems.

    Limited research has explored how to best train caregivers to support their child with autism spectrum disorder (ASD) despite caregivers being well suited to promote generalization and maintenance of their child’s skills in the Natural environment. Children with ASD have been shown to benefit from social skill training, which is not always conducted in the Natural Context. This research examined the efficacy of behavioral skills training (BST) with, and without in situ training (IST), for teaching caregivers how to also use BST to support their child’s Context-specific social skills. Although caregivers met mastery criterion within BST sessions, their skills did not generalize to the Natural environment until IST was introduced. The implications of the findings are discussed.

    Abstract Keratocan, along with lumican and mimecan, represent the keratan sulfate-containing proteoglycans of the vertebrate cornea that play a key role in development and maintenance of corneal transparency. In this study, we cloned 4.1 kb of the human Kera 5′-flanking region and characterized the promoter structure. Using primer extension and ribonuclease protection assay, we identify two major transcriptional start sites in the first exon. Using luciferase reporter gene transfection analysis of 5′-deletion and mutation constructs, we demonstrate positive and negative regulatory elements within a 1.3 kb upstream sequence. Comparison of human and bovine 5′-flanking sequences reveals three highly conserved regions: a 450 bp region in the first exon, a 92 bp promoter proximal conserved regulatory region identified as an enhancer in the Natural Context, and a 223 bp promoter distal conserved regulatory region identified as a silencer both in the Natural Context and in a heterologous promoter system. In addition, a conserved CArG-box residing 851 bp upstream of the first transcription start site also can lead to the repression of Kera expression in cultured corneal keratocytes. DNaseI footprinting and electrophoretic mobility shift assay demonstrate that cell type-specific factors bind to regulatory elements located in the conserved regions. Competition experiments indicate that the CTC factor and a protein that binds to the CAGA motif are likely to be among the multiple factors involved in the transcriptional regulation of the human Kera gene.

    Abstract:

    Protein complex purification represents a powerful approach to identify novel players in plant innate immunity. However, the identification of interacting protein partners within a Natural Context has been a challenge for researchers. In this chapter, we describe a method of immunoaffinity chromatography using purified, antibodies to isolate native protein complexes from wild-type tissue. We detail the antibody purification and immobilization steps in addition to the co-immunoprecipitation protocol. In addition, a method to prepare protein samples for mass spectroscopy analysis is described. This straightforward protocol has been used to isolate and identify novel components of Arabidopsis immunity-associated protein complexes.

    Publisher Summary This chapter concerns the inter-relationships between hormones, brain, and behavior that mediate the occurrence of reproductive cycles and reproductive behavior among adults in avian species. The chapter ranges from a consideration of the neuroendocrine basis of seasonal reproduction to a description of the neural, and endocrine mechanisms that regulate male and female sexual behavior and parental behavior. Many of the individual sections of this chapter correspond to entire chapters in this multivolume work on hormones, brain, and behavior. A consideration of the neuroendocrine basis of reproductive cycles and reproductive behavior in bird species is potentially particularly beneficial. Many of the pioneering studies concerning different aspects of the inter-relations among hormones, brain, and behavior are conducted on birds. Therefore, the neuroendocrinology of reproductive behavior in birds can be considered more easily in a Natural Context than in many other taxa.
