Mechanism of modulation through PI3K-AKT pathway about Nepeta cataria L.'s extract in non-small cell lung cancer

But it is still a vital mission for the current medical and researchers that hunting a natural medicine which have little side effects and high-efficiency against the NSCLC on account of the shortcomings on current drugs. Nepeta cataria L. plays an important role in anti-cancer treatment according to the reports which was recorded in the Chinese Pharmacopoeia of version 2015 and belongs to one of the Traditional Chinese medicine (TCM). Microfluidic chip technology is widely used in scientific research field due to its high-throughput, high sensitivity and low cost with the continuous progress of science and technology. In this study, we investigate the effect of total flavonoid extracted from Nepeta cataria L. (TFS) through human lung cancer cell line A549 based on the microfluidic device and Flow Cytometry. So we detected the mRNA expression of MicroRNA-126 (miR-126), VEGF, PI3K, PTEN and proteins expression respectively to explore the partial PI3K-AKT pathway molecular mechanisms through Quantitative Real-time PCR (qRT-PCR) and Western Blot. The results showed that TFS can disturb the expression of miR-126 and regulate the PI3K-AKT signaling pathway to meet the effect of anti-cancer. Taking all these results into consideration we can draw a conclusion that TFS may be used as a novel therapeutic agent for NSCLC in the near future.

Repellency Assessment of Nepeta cataria Essential Oils and Isolated Nepetalactones on Aedes aegypti

Natural products are often used when DEET cannot be afforded or accessed and when consumers choose not to use a synthetic repellent. The essential oils from two newly bred Nepeta cataria (catnip) plants representing two different chemotypes and their respective isolated Nepetalactone isomers were evaluated as mosquito repellents against Aedes aegypti mosquitoes that transmit the Zika and Dengue virus in a one choice landing rate inhibition assay. A dose response curve was generated for each treatment and a time course analysis of repellency was performed over 24 hours with a N. cataria essential oil sample. The results indicate that all essential oil samples and their respective purified Nepetalactone isomers were able to achieve greater than 95% repellency. Between two and four hours, the ability to repel more than 95% of the mosquitoes diminished. At the lowest concentrations tested, the Nepetalactones and crude essential oil samples were more effective than DEET at reducing the number of mosquito landings.

Two major Nepetalactones, Z,E-Nepetalactone and E,Z-Nepetalactone, were successfully identified and quantified. The linearity range for Z,E-Nepetalactone was determined as 0.00655–0.655 mg/mL with a correlation coefficient of 0.9999, and the linearity range of E,Z-Nepetalactone was found to be 0.00228–0.456 mg/mL with a correlation coefficient of 0.9999, under UV detection at 228 nm. The linearity ranges were from 0.00164 to 0.0328 mg/mL, with a correlation coefficient of 0.9999, for Z,E-Nepetalactone and 0.00114–0.0228 mg/mL, with a correlation coefficient of 0.9999, for E,Z-Nepetalactone by MS detection with selected ion monitoring of ion peak m/z 167. The MS detection was found to be more sensitive than UV detection and this method was validated as simple, reliable and sensitive for catnip Nepetalactone analysis. Nepeta cataria l var citriodora becker increases penile erection in rats

Aim of the study Because sexual behavior is involved in pleasure, the effect of NC on sexual behavior and penile erection was evaluated in male rats that were acutely fed chow enriched with 10% NC leaves. Further, yawning was monitored because we previously demonstrated that NC modifies dopaminergic-related behaviors and that sexual behavior is closely linked with the dopaminergic system. The general activity and the motor coordination were examined to investigate the possible motor and emotional interferences of the sexual performance. Material and methods Male rats of the NC group received for a 4 h period the chow enriched with 10% NC leaves while the control groups received regular chow. Fifteen min after the end of the 4 h period of NC feeding the sexual behavior, apomorphine-induced penile erection and motor coordination were observed; the general activity in the open field was assessed 0, 15, 30 and 60 min after treatment. Results NC treatment increased male rat's penile erection. A slightly facilitation on male rat sexual behavior and a decreased in general activity of NC treated rats were observed. No effects on motor coordination and yawning episodes were detected by the NC treatment. Conclusions It was suggested that NC increases penile erection and slightly improves male rat sexual behavior by an action on dopaminergic systems.

Antidepressant-like effects of an apolar extract and chow enriched with Nepeta cataria (catnip) in mice

Catnip has also been used in the treatment of some depressive disorders. In this paper, we studied the antidepressant, anxiogenic, and motor activity effects of acute and repeated feeding of chow enriched with 10% N. cataria leaves and the acute and repeated administration of apolar and polar extracts of N. cataria leaves in male mice. The results showed that repeated feeding and acute and repeated administration with the apolar extract reduced immobility in the behavioral despair test but did not alter elevated plus maze and open-field parameters. Acute feeding and the acute and repeated administration of the polar extracts of N. cataria leaves did not alter the behavior of mice. These data suggest that N. cataria has antidepressant properties. Moreover, this antidepressant activity was present in the apolar extract.

This study was performed to analyze the anti-nociceptive and anti-inflammatory effects of N. cataria essential oil (NCEO) in female mice. Methods – Phytochemical analyses of NCEO were performed. In addition, female mice treated with the oil were observed in an open field for its general activity and to investigate the dose and time responses. The anti-nociceptive effects were evaluated by tail immersion and acetic acid writhing reflex tests. The anti-inflammatory oil properties were investigated by the carrageenan-induced edema test. Results – The results showed that 0.0005 and 0.001 mL/kg i.p. doses of NCEO increased the general activity of female mice, and the 0.0005 mL/kg dose reduced their immobility. Moreover, NCEO (0.0005 mL/kg) has anti-nociceptive properties, as the treated animals exhibited an increased latency of tail withdrawal and reduced acetic acid-induced abdominal constrictions. Furthermore, NCEO (0.0005 mL/kg) presented peripheric anti-inflammatory properties by reducing the induced edema after carrageenan injection. Conclusions – These effects may be due to the Nepetalactone trans-trans and trans-cis Nepetalactone isomers, which were detected as the predominant active components in the phytochemical analysis. It was suggested that the main effect of NCEO occurs on the central nervous system mechanism of pain. Behavioral effects of acute and long-term administration of catnip (Nepeta cataria) in mice.

We investigated the effects of acute and long-term administration of the plant on some behaviors of mice. The plant was fed as 10% of the normal diet for 2 h/d for 1 or 7 d. Acute and long-term dosing increased both rearing and locomotion frequencies observed in an open field. Acute exposure to catnip increased stereotyped behavior and susceptibility to seizures, did not interfere with haloperidol-induced catalepsy, and decreased sleeping time after sodium pentobarbital administration. Long-term exposure induced tolerance to stereotypic behavior, catalepsy and sleeping time, and increased the susceptibility to seizures induced by picrotoxin and strychnine. An amphetamine-like effect of catnip was suggested to explain the acute effects, while dispositional and functional adaptative changes were considered involved with the long-term effects.
