effect of oxymetazoline Nose Drops on acute sinusitis in the rabbit

Arfors, Marcos IntagliettaAbstract:This investigation was designed to evaluate the effect of the common topical nasal decongestant oxymetazoline hydrochloride on the early local tissue defense in an experimental bacterial infection. For that purpose, a bilateral infection of the maxillary sinus was induced in 14 rabbits. Nose Drops (oxymetazoline) were instilled in one nasal cavity, and placebo in the other. After 48 hours, the degree of infection was judged by using a semiquantitative inflammatory score to evaluate histologic preparations of the maxillary sinuses. On the oxymetazoline-treated side, we found a significantly higher degree of inflammation. We conclude that oxymetazoline Nose Drops, commonly used in acute rhinitis and sinusitis, interfere with the normal defense mechanisms during bacterially induced sinusitis, possibly by a decrease in mucosal blood flow.

Nose Drops induce vasomotion in the microcirculation of the sinus mucosa of the rabbit.

Arfors, Marcos IntagliettaAbstract:Periodic oscillations of blood in the microcirculation of the sinus mucosa in the rabbit were studied with laser-Doppler flowmetry after the application of Nose Drops (oxymetazoline, xylometazoline and phenylephrine) to the nasal mucosa. All of these drugs induced a dose-dependent decrease in the blood flow in the mucosa. Flowmotion was found in 9 of 21 experiments after an approximately 50% reduction in blood flow had been induced. The concentrations of the drugs which caused vasomotion corresponded to those normally administered as Nose Drops to man.

Effect of oxymetazoline Nose Drops on vascular permeability of the nasal mucosa in the rabbit after provocation with leukotriene B4

Vascular permeability (125I-albumin) was 53% higher in the leukotriene B4-provoked nostril (LTB4) compared with the vehicle-treated con-tralateral nostril (p 4-induced increase in permeability was decreased by 22 % when oxymetazoline was introduced (p 4-induced increase in vascular permeability was not attenuated by the monoclonal antibody IB4 directed against the neutrophil adhesion complex CD 11/CD18. The latter suggests that LTB4-induced vascular permeability does not require CD18-mediated neutrophil adherence in the nasal mucosa.

Effect of Tong Qiao Drops on the expression of eotaxin, IL-13 in the nasal mucosa of rats with allergic rhinitis

Studies have found that numerous herbs and their constituent compounds can significantly alleviate allergic symptoms and are effective treatments for allergic rhinitis. The purpose of this study was to examine the modulatory effect of Tong Qiao Nose Drops on allergy symptoms and the expression of cytokines in the nasal mucosa of rats with ovalbumin-induced allergic rhinitis. Methods Sixty healthy male Sprague Dawley rats were randomly divided into three groups ( n = 20): negative, control, and experimental. Rats in the control or experimental groups were sensitized with ovalbumin to induce allergic rhinitis. The sensitized rats in the experimental group were subsequently exposed to Tong Qiao Nose Drops, whereas the sensitized control rats were given saline Nose Drops. Negative control rats were only treated with saline. Allergic symptoms and the pathologic features of the nasal mucosa were observed. The expression of eotaxin in the mucous membrane of rat nasal septums was detected by immunohistochemical staining, and the expression levels of interleukin (IL)-5 and IL-13 were measured by enzyme-linked immunosorbent assay. Results The symptom scores for the experimental group were significantly lower than those of control rats ( p p Conclusion Tong Qiao Nose Drops alleviated the symptoms of allergic rhinitis in a rat model and lowered the expression levels of eotaxin, IL-5, and IL-13.

We present two cases of childhood Cushing's syndrome secondary to prolonged use of intranasal betamethasone. ### Case 1 >A boy aged 7 years was referred to the endocrine clinic with a history of growth failure associated with obesity. Over the past two years his weight had increased from the 50th to the 97th centile with height falling from the 10th to the 3rd centile. He had no history of note apart from mild atopy and chronic catarrh, for which he had been prescribed 0.1% betamethasone nasal Drops. On examination he was cushingoid with normal prepubertal genitalia. His blood pressure was 165/75 mm Hg (>95th centile for age).
