On the relationships of Paecilomyces sect. Isarioidea species.

Isarioidea species were analysed using the β-tubulin gene and ITS rDNA. Maximum parsimony analyses showed that the section does not form a natural taxonomic group and is polyphyletic within the Hypocreales. However, a group was recognized, designated as the Isaria clade, to be monophyletic comprising of the following Paecilomyces species: P. amoeneroseus, P. cateniannulatus, P. cateniobliquus, P. cicadae, P. coleopterorus, P. farinosus, P. fumosoroseus, P. ghanensis, P. javanicus and P. tenuipes. Some of these species have teleomorphs in Cordyceps.

The polyphyletic nature of Paecilomyces sensu lato based on 18S-generated rDNA phylogeny
Mycologia, 2004Co-Authors: J Jennifer Luangsa-ard, Nigel L Hywel-jones, Robert A SamsonAbstract:Nuclear-encoded small-subunit ribosomal DNA was used to examine phylogenetic relationships in Paecilomyces sensu lato. Phylogenetic analysis of the 18S nr DNA demonstrates that Paecilomyces is polyphyletic across two subclasses, Sordariomycetidae and Eurotiomycetidae. The type species, Paecilomyces variotii, and thermophilic relatives belong in the order Eurotiales (Trichocomaceae), while mesophilic species related to Paecilomyces farinosus are in the order Hypocreales (Clavicipitaceae and Hypocreaceae). One species, Paecilomyces inflatus, had affinities for the order Sordariales. Within the Eurotiales, Paecilomyces is monophyletic. Within the Hypocreales, species of Paecilomyces are polyphyletic, although the data failed to fully resolve these relationships.

Thermotolerant and thermoresistant Paecilomyces and its teleomorphic states isolated from Thai forest and mountain soils

The predominant species of Paecilomyces that had been identified was Paecilomyces variotii, the type species of the genus . Oatmeal Agar was used to induce the teleomorph at 37∞C. Other species isolated belong to Paecilomyces or its teleomorphic states Byssochlamys, Talaromyces and Thermoascus included Byssochlamys nivea, Byssochlamys fulva, Talaromyces byssochlamydoides and Thermoascus crustaceus.

We present the case of an 84-year-old immunocompromised patient with interstitial lung disease, who developed pulmonary mycetoma with isolation of P. variotii in bronchoalveolar lavage fluid culture. We found one similar case in the literature, but Paecilomyces species was not identified. Therefore, this is the first described case of pulmonary mycetoma caused by P. variotii. LEARNING POINTS Paecilomyces are rare causes of fungal infection in humans and occur more frequently in immunocompromised patients. Paecilomyces variotii is one of the most common pathogens of this species and is associated with peritonitis, pneumonia, osteomyelitis, endophthalmitis and fungaemia. Amphotericin B, itraconazole, and echinocandins showed poor activity against Paecilomyces lilacinus, while the new triazoles were active against it. Paecilomyces variotii exhibited a different susceptibility pattern, being susceptible to most antifungal agents apart from voriconazole and ravuconazole.

The diagnosis was established by isolation of the fungus in culture from specimens obtained by percutaneous transthoracic needle aspiration in an immunocompetent individual. The isolate was atypical, as it failed to produce fruiting structures on routine mycological media. Identification was achieved by sequencing polymorphisms of the internal transcriber spacer regions of the ribosomal DNA. The antifungal susceptibility profile was also determined. This report underscores the increasing importance of Paecilomyces species in human infections. Paecilomyces should be included among the etiological agents of pulmonary mycetoma.
