The methanol extract of P. incarnata exhibited significant aphrodisiac behaviour in male mice at all doses, i.e. 75, 100 and 150 mg/kg. Amongst these, the highest activity was observed with the 100 mg/kg dose when the mountings were calculated about 95 min after the administration of the test extracts.

Despite a very long history of use, the healing potential and the mode of action of this plant was never described clearly, perhaps due to the controversial identity of P. incarnata with Passiflora edulis Sims. In many references, both these plants were reported and considered to be synonyms of each other and were named passion flower, maracuja or maypops. The key differential parameters reported by the authors enable the selection of the authentic plant, i.e., P. incarnata for the purpose of systematic biological studies. In the present investigations, the methanol extract of leaves of P. incarnata was evaluated for various CNS effects in experimental animals. The methanol extract of leaves of P. incarnata exhibited significant sedative, anticonvulsant and CNS-depressant activities at a dose of 200 mg/kg in mice. The extract also exhibited analgesic and anti-inflammatory activities against acetic acid-induced pain and carrageenan-induced edema, respectively, when evaluated in experimental animals.

The 20 mg kg(-1) dose of BZF exhibited better results. 4 Upon discontinuation of THC, treatment with BZF (10 and 20 mg kg(-1) p.o.) also facilitated the early restoration chronic-THC-induced decline in libido, sperm count and sexual fertility within 7 days.

In the light of the established usefulness of the benzoflavone moiety in counteracting the withdrawal effects of substances like morphine, cannabinoids and nicotine by the authors, the bioactive benzoflavone moiety (BZF) has been tested in mice treated with an addictive dose (2 g/kg, bid for 6 days) of ethyl alcohol, in order to evaluate its effectiveness in countering alcohol dependence. In a 7-day regimen, different groups of mice were administered vehicle, alcohol and alcohol+three doses (10, 20 and 50 mg/kg of the benzoflavone moiety) of P. incarnata; all treatments (chronic) being administered orally, twice daily for 6 days. Similarly, three other groups of mice were rendered addicts upon alcohol by administration of the addictive dose (2 g/kg, bid for 6 days) of ethyl alcohol, and a single acute administration of 10, 20 and 50 mg/kg dose of benzoflavone moiety was given on the 7th day. In both, chronic and acute administrations, the benzoflavone moiety prevented significantly the expression of withdrawal effects of alcohol as there was a significant decrease in anxiety oriented behavior in mice that received benzoflavone moiety of P. incarnata. The chronic administration of P. incarnata with alcohol had better preventive effects than the single acute treatment with P. incarnata in alcohol-dependent mice.

Despite the worldwide use of P. incarnata, the pharmacological work on this plant had been inadequate, inconclusive and wage as the earlier reports were unable to infer the mode of action of the plant as well as the phytoconstituents responsible for the much acclaimed anxiolytic and sedative effects of P. incarnata. An attempt has been made to isolate and identify the bioactive phytomoiety of P. incarnata by resorting to bioactivity directed fractionation and chromatographic procedures. A fraction derived from the methanol extract of P. incarnata has been observed to exhibit significant anxiolytic activity at a dose of 10 mg/kg in mice using elevated plus-maze model of anxiety. This fraction comprises mainly two components which are visible as blue and turquoise colored fluorescent spots at 366 nm of the UV light. The possibility of a phytoconstituent having benzoflavone nucleus as the basic moiety being responsible for the bioactivity of P. incarnata is highly anticipated.

Phytochemical Analysis, 1995Co-Authors: Anne Rehwald, Otto Sticher, Beat H MeierAbstract:Seventeen different samples of Passiflora incarnata were screened for harman alkaloids by a reversed-phase high performance liquid chromatographic (HPLC) method. Only one sample yielded a possible harman content of approximately 0.1 ppm which was just above the level of detection limit. The method comprises extraction of the plant material, purification and concentration of the alkaloid fraction by combined solid-phase extractions on Extrelut and cation-exchange cartridges, and subsequent HPLC analysis using diode-array and fluorescence detection. The method was found to be sensitive and selective and revealed good recovery. The efficiency of the method was demonstrated with Peganum harmala seeds.

The methanol extracts of leaves, stems, flowers, and whole plant exhibited anxiolytic effects at 100, 125, 200 and 300 mg/kg, respectively. The roots were practically devoid of anxiolytic effects. These results show that roots and flowers of P. incarnata act as natural adulterants by causing a significant increase in the anxiolytic dose. Therefore, separation of these parts is recommended prior to any pharmacological, phytochemical and standardization studies on P. incarnata.
