    Abstract:

    Abstract High-speed counter-current chromatography (HSCCC) with a solvent system composed of ethyl acetate–n-butanol–water (2:1:3, v/v/v) was used to isolate and separate two C-glycosylflavones from Patrinia villosa Juss, a traditional Chinese medicine. The separation produced 42.9 mg isovitexin and 20.1 mg isoorientin with purities of 99.3% and 98.5%, respectively as determined by high-performance liquid chromatography (HPLC) in one step elution from 250 mg crude extract, and identification was performed by MS, 1H NMR and 13C NMR. It is the first report of discovering isovitexin and isoorientin from the plant of Patrinia genus.

    Abstract:

    AIM To study the chemical constituents of Patrinia villosa Juss. METHODS Solvent extraction, silica gel column and preparative liquid chromatography were used to separate the chemical constituents, and the chemical structures were elucidated by physico-chemical properties and spectra data. RESULTS Eight compounds were isolated and identified as bolusanthol B (1), (2S)-5, 7, 2', 6'-tetrahydroxy-6,8-di (gamma,gamma-dimethylallyl) flavanone (2), orotinin (3), (2S)-5, 7, 2', 6'-tetrahydroxy-6-lavandulylated flavanone (4), 3'-prenyl-apigenine (5), luteolin (6), quercetin (7) and apigenin (8). CONCLUSION Compound 2 and 4 are new compounds, compounds 1, 3 and 5 were separated from Patrinia genius for the first time, compounds 6, 7 and 8 were isolated from Patrinia vollosa Juss for the first time.

    Abstract:

    OBJECTIVE To investigate the chemical constituents of Patrinia villosa. METHOD The chemical constituents were isolated by silica gel column chromatography and semi-preparative high-performance liquid chromatography, and identified by physicochemical properties and spectral analysis (MS, 1H-NMR and 13C-NMR). RESULT Seven compounds were isolated from ethyl acetate and n-butanol extract and identified as: 5-hydroxyl-7, 3', 4'-trimethoxy flavone (I), 5-hydroxyl-7, 4'-dimethoxy flavone (II), luteolin (III), quercetin (IV), isoorientin (V), isovitexin (VI) and 8-C glucosylprunetin (VII). CONCLUSION Compounds I , II, III, V, VI and VIII were obtained from the plant of genus Patrinia for the first time, compound IV was separated from P. villosa for the first time.

    Abstract:

    Nine compounds including n-dotriacontanoic acid (I), n-dotriacontanol (II), Palmic acid (III), oleanolic acid (IV) aurentiamide acetate (V), daucosterol (VI), beta-sitosterol (VII), 7beta-hydroxysitosterol (VIII) and stigmasterol (IX) were isolated from petroleum ether and chloroform extract of Patrinia villosa Juss. Among them, compounds I, II, V, VIII were obtained from the plant of Patrinia for the first time and compounds VI, VII ,IX were separated from Patrinia villosa Juss for the first time.

    From the roots of Patrinia scabra two new iridoids, 3-methylbutyric acid 7-hydroxy-7-hydroxymethyl-4-(3-methyl-butyryloxymethyl)-6-oxo-1,6,7,7a-tetrahydrocyclopenta[c]pyran-1-yl ester (1) and 6-hydroxy-7-methylhexahydrocyclopenta[c]pyran-3-one (2) have been isolated. Their structures were determined by means of NMR spectra and X-ray crystallographic analysis.

    AIM To study the lignans from Patrinia scabra Bunge. METHODS The constituents were separated and purified by column chromatography with silical gel, RP-silical gel and Sephadex LH-20. Their structures were identified on the basis of spectral data (IR, MS, 1HNMR, 13CNMR, HMQC and HMBC). RESULTS AND CONCLUSION A new lignan was obtained and its structure was elucidated as 4-[1-ethoxyl-1-(4-hydroxy-3-methoxy)benzyl]methyl- 2-(4-hydroxy-3-methoxy)benzyl-3-hydroxymethyl-tetrahydro-furan (2), along with three known lignans, lariciresinol (1), isolariciresinol (3) and nortracheloside (4).

    Various chromatographic techniques were used to separate and purify the constituents. Their structures were elucidated on the basis of chemical evidences and spectral analysis. Four compounds were isolated from the roots of P. scabra. They were identified as hexadecylic acid-#alpha#-monoglyceride (I), oleanolic acid (II), quercetin (III), N-benzoyl-phenylalanine-2-acetylamino-phenylproanol easter (IV). All compounds were isolated from this plant for the first time. Among them, IV is a new alkaloid and named patriscabratine.

    Abstract Five compounds were isolated from the roots of Patrinia scabra Bunge separated and purified on sillica gel column chromatography. Their structures were elucidated on the physico-chemical properties and spectral data as lariciresinol(I), syringaresinol(II), scopoletin(III), quercetin(IV), ferulaic acid(V). All compounds were obtained from P. scabra for the first time. In vitro biological test of these compounds showed that syringaresinol was able to stimulate T and B lymphocyte proliferation.
