    Abstract:

    Abstract Head lice are exclusive human parasitic blood-sucking insects. Distributed worldwide among school-age children, this parasitosis generates scalp irritation and sometimes social prejudice. Understanding how head lice detect and perceive their human hosts is crucial to control transmission. Here, we describe the sensory structures present on the mouthparts of Pediculus humanus capitis and their possible contribution to the feeding decision-making process. On the anterior zone of the clypeus around the haustellum two morphological types of sensilla, invariable in location and number, were identified: fourteen short clypeus bristles (SCB) and six long clypeus bristles (LCB). During feeding these structures contact the host skin but not its blood. Located antero-dorsally on the everted haustellum and between the epipharyngeal teeth, a third sensillar type was identified: about four short peg epipharyngeal (SPE) sensilla. These structures are bathed with the incoming blood, when head lice feed, so may have a gustatory role. In behavioural experiments antennectomy of lice did not interfere with feeding behaviour, suggesting that the sensory structures on the mouthparts could be involved in host assessment.

    Abstract:

    Human pediculosis is caused by Pediculus humanus humanus (Linnaeus 1758) and Pediculus humanus capitis (De Geer 1767). We studied the response of body lice to immersion in water and ethanol 70% and determined the optimal times for measuring knockdown and mortality. After immersion in water, all lice remained alive from 5 min to 22 h for both times of exposure. A low proportion of lice were affected after 2 min of immersion in ethanol in the 10-min exposure test, but recovered completely after 5 min. Different proportions of lice were affected between 2 and 7 h after immersion in ethanol, depending on the immersion time. However, a high proportion of lice recovered after 22 h. The results suggest that the optimal times for measuring early knockdown effects of insecticides are the 5-min to 7-h interval for water and 5-min to 1-h interval for ethanol. On the other hand, the best time for measuring mortality is 22 h after immersion. These results should improve the interpretations of the effects of pediculicides in immersion bioassays.

    During the past decades, chemical control against the head louse Pediculus humanus capitis De Geer has been based in the application of products containing permethrin. The repetitive overuse of pediculicides has resulted in the development of high levels of resistance to one or more of these products worldwide. Essential oils obtained from aromatic plants like Eucalyptus are good and safe alternatives due to their low toxicity to mammals and easy biodegradability. In the present study, we reported the chemical composition of Eucalyptus dunnii and Eucalyptus gunni, and the fumigant activity of five Eucalyptus essential oils and their main compounds against permethrin-resistant head lice from Argentina. The most effective essential oils were Eucalyptus sideroxylon, Eucalyptus globulus ssp globulus, and Eucalyptus globulus ssp maidenii, with knockdown time 50% (KT50) values of 24.75, 27.73, and 31.39 min. A linear regression analysis between percentage of 1,8-Cineole and KT50 values of the essential oils showed a significant correlation at a p < 0.01. Since Eucalyptus essential oils showed to be effective against head lice and are classified as safer compounds, they can be employed into pediculicide formulations.

    Abstract:

    The improper use of pediculicides containing permethrin has led to the development of resistance. Thus, new alternatives for control are needed. Plant-derived insecticides are attractive alternatives to common chemical insecticides because most of them are environmentally friendly and non-toxic to mammals. The toxic activity of 23 monoterpenoids belonging to several chemical classes was tested against the eggs of permethrin-resistant head lice, Pediculus humanus capitis De Geer (Phthiraptera: Pediculidae). Significant differences in ovicidal action were observed among the tested substances. The most effective chemicals were hydrocarbons and ethers, followed by ketones, alcohols, phenols and esters. A linear relationship between egg mortality and knockdown time (KT(50)) on adults by the tested components revealed that most of the components were effective on both egg and adult stages. The monoterpenoids described herein are good candidates as effective pediculicides.

It has recently been considered re-emerging among homeless populations in developed countries. In Italy, it was last reported in 1945. Pediculus humanus is a vector of highly relevant human pathogens. Methods In October 2018, a woman found small insects on a T-shirt bought second-hand in a local street market in a village 35 km south of Rome (central Italy). Insects were identified both morphologically and by molecular analysis. Moreover, they were analyzed molecularly for the presence of Rickettsia prowazekii , Borrelia recurrentis , Bartonella quintana , Coxiella burnetii and Yersinia pestis . Results Morphological and molecular analyses of the insects identified them as 26 lice (12 females, 10 males and 4 nymphs) of the species P. humanus . Many nits were found on the T-shirt seams. DNA of the investigated pathogens was not detected in any of the lice. Conclusions The exceptionality of the described case lies both in the report of P. humanus from a country where it had not been reported since 1945, and in its finding from second-hand clothes for sale in a market, constituting a potential source of infection for people buying this type of goods. The question arises, how did adults and nits of P. humanus infest clothes for sale on a market stall in a country where it had not been reported for decades. Hence, it must be presumed that people infected with the human body louse are present in Italy.     New alternative insecticides are necessary for the chemical control of head lice. In this study the fumigant knockdown time 50% (KT50) and repellency index (RI) of three aliphatic lactones was compared with two essential oils and DDVP, against permethrin-resistance Pediculus humanus capitis from Argentina. In the fumigant assay, none of the lactones were effective compared to the highest activity of eucalyptus (KT50 15.53 m). In the repellency test, the three lactones were equally or more effective (RI ranging from 60.50 to 76.68) than the positive control (piperonal). These lactones are promising as head lice repellents.

    Abstract The effectiveness of 1-octanol, 1-nonanol, 1-decanol, 1-undecanol, and 1-dodecanol was evaluated by immersion method against susceptible and permethrin-resistant head lice, Pediculus humanus capitis De Geer, from Buenos Aires, Argentina. All the tested alcohols showed knockdown effect at 10 min and mortality 18 h after treatment. The highest activity was found for the 1-dodecanol (KC50 2.55%, LC50 2.28%) and the lowest for 1-octanol (KC50 8%, LC50 4.46%). The toxicity to the head lice systematically increased with the increase in carbon atoms in the n-aliphatic alcohol moiety, and with the octanol:water coefficient (r2 = 0.94). The pediculicidal activity of 1-dodecanol was not correlated with resistance to permethrin, because no significant difference was observed between toxicity parameters in the susceptible (MAR) and the permethrin-resistant populations which had different resistant levels (RR 5.77× for E49 population, RR 9.5× for HL population and RR > 35.3× for GH population).
