Correlates of urinary concentrations of phthalate and phthalate alternative metabolites among reProductive-aged Black women from Detroit, Michigan

In the United States, Black women are more highly exposed to phthalates than other racial/ethnic groups, yet information on predictors of phthalate exposure among Black women is limited. Objective We evaluated the association of demographics, lifestyle, reProductive history, and Personal Care Product use with urinary concentrations of phthalate and phthalate alternative metabolites, using cross-sectional data from a study of 754 Black women from Detroit, Michigan (2010–2012). Methods Women completed questionnaires and provided urine specimens which were analyzed for 16 phthalate and phthalate alternative metabolites. We used linear regression models to estimate mean percentage differences and 95% confidence intervals (CIs) in concentrations across levels of correlates. Results Monoethyl phthalate (MEP) and MBP concentrations were positively associated with Personal Care Product use, particularly nail Products. Educational attainment was positively associated with high molecular weight phthalate concentrations but inversely associated with monobenzyl phthalate (MBzP) concentrations. Parity was positively associated with MBzP concentrations and inversely associated with concentrations of MEP and high molecular weight phthalates. Significance We found that sociodemographics, reProductive characteristics, and use of certain Personal Care Products were associated with urinary phthalate concentrations among Black women. Our results emphasize the importance of examining exposure determinants among multiply marginalized populations.

EDCs have been linked to pregnancy complications such as gestational diabetes (GDM), and PCP use could represent a modifiable source of exposure in this sensitive time window. Yet, to our knowledge, no study has directly evaluated the association between pregnancy use of PCP and late pregnancy glucose levels, established risk factors for complications such as GDM. Methods A total of 233 women from the Environment and ReProductive Health (EARTH) Study had data available on 1st and/or 2nd trimester PCP use, assessed through self-reported use over the previous 24 h, and blood glucose levels after the glucose loading test (GLT), taken at late 2nd trimester. Associations between each individual PCP and total PCP with glucose levels were evaluated in multivariable adjusted linear regression models. Results Both positive and negative differences in glucose levels were observed when comparing users vs. non-users of several PCPs including 2nd trimester use of deodorant (adjusted mean difference: 12.2 mg/dL, 95% CI: −0.6, 24.9); bar soap (6.9 mg/dL, 95% CI: −0.9, 14.7 mg/dL); and liquid soap (−13.3, 95% CI: −26.8, 0.1 mg/dL), and 1st trimester use of sunscreen (−14.6 mg/dL, 95% CI: −27.8, −1.5 mg/dL). Total number of PCPs used in the 2nd trimester was also associated with higher glucose levels, with the largest difference of 20 mg/dL when comparing individuals who used eight vs none PCPs (95% CI: 3–37). Conclusions In a pregnancy cohort of women seeking Care at a fertility clinic, we found the use of several PCPs to be positively or negatively associated with glucose levels in the late second trimester, which may reflect increased risk of GDM and subsequent perinatal outcomes. These results strengthen the role of Product use as a potentially modifiable source of EDCs that may impact glucose levels.

These findings have contributed to the replacement of some chemicals, such as di‑2‑ethylhexyl phthalate (DEHP) and bisphenol A (BPA), in consumer Products. For the current study we quantified urinary concentrations of biomarkers of exposure among 50 pregnant women in Israel to several phthalates, bisphenols and Personal Care Product chemicals, as well as DEHP and BPA alternatives. We detected 14 of the 31 biomarkers in more than 90% of the women. We detected biomarkers of 1,2‑cyclohexane dicarboxylic acid, diisononyl ester (DINCH), bisphenol S, and bisphenol F not as frequently (27–56%). This study is the first to evaluate exposure to triclosan, bisphenols, parabens, and phthalates and BPA alternatives among Israeli pregnant women.

In 2009–2010, 80 pregnant women from Ottawa Canada participated in the Plastics and Personal-Care Product Use in Pregnancy (P4) Study. A subset of women (n = 31) who provided multiple spot urine samples (n = 542) collected over two 24-h periods had their samples analyzed for methylparaben (MP), n-propylparaben (PP), ethylparaben (EP), butylparaben (BP), isobutylparaben (IBP), and benzylparaben (BzP). These parabens were also measured in breast milk samples collected at approximately 3 months postpartum (n = 56 women). The existing literature is conflicting, and many studies do not control for multiple chemical exposures. Methods We quantified concentrations of three phthalate metabolites, three parabens, and four other phenols in urine collected twice during pregnancy from 392 women. We measured T helper 1 (Th1) and T helper 2 (Th2) cells in their children's blood at ages two, five, and seven, and assessed probable asthma, aeroallergies, eczema, and lung function at age seven. We conducted linear and logistic regressions, controlling for additional biomarkers measured in this population as selected by Bayesian Model Averaging. Results The majority of comparisons showed null associations. Mono-n-butyl phthalate (MnBP) was associated with higher Th2% (RR: 10.40, 95% CI: 3.37, 17.92), and methyl paraben was associated with lower Th1% (RR: −3.35, 95% CI: −6.58, −0.02) and Th2% at borderline significance (RR: −4.45, 95% CI: −8.77, 0.08). Monoethyl phthalate was associated with lower forced expiratory flow from 25 to 75% of forced vital capacity (FEF25–75%) (RR: −3.22 L/s, 95% CI: −6.02, −0.34). Propyl paraben (OR: 0.86, 95% CI: 0.74, 0.99) was associated with decreased odds of probable asthma. Conclusions While some biomarkers, particularly those from low molecular weight phthalates, were associated with an atopic cytokine profile and poorer lung function, no biomarkers were associated with a corresponding increase in atopic disease.

Human exposure occurs through consumption of processed food and use of cosmetics and consumer Products. The aim of this study was to provide a preliminary characterisation of exposure to selected Personal Care Product chemicals in the general Australian population. De-identified urine specimens stratified by age and sex were obtained from a community-based pathology laboratory and pooled (n= 24 pools of 100). Concentrations of free and total (sum of free plus conjugated) species of methyl, ethyl, propyl and butyl paraben, benzophenone-3 and triclosan were quantified using isotope dilution tandem mass spectrometry; with geometric means 232, 33.5, 60.6, 4.32, 61.5 and 87.7. ng/mL, respectively. Age was inversely associated with paraben concentration, and females had concentrations approximately two times higher than males. Total paraben and benzophenone-3 concentrations are significantly higher than reported worldwide, and the average triclosan concentration was more than one order of magnitude higher than in many other populations. This study provides the first data on exposure of the general Australian population to a range of common Personal Care Product chemical ingredients, which appears to be prevalent and warrants further investigation.

Calafat, Russ HauserAbstract:Parabens and phthalates are potential endocrine disruptors frequently used in Personal Care/beauty Products, and the developing fetus may be sensitive to these chemicals. We measured urinary butyl-paraben (BP), methyl-paraben, propyl-paraben, mono- n -butyl phthalate (MBP), and monoethyl phthalate (MEP) concentrations up to three times in 177 pregnant women from a fertility clinic in Boston, MA. Using linear mixed models, we examined the relationship between self-reported Personal Care Product use in the previous 24 h and urinary paraben and phthalate metabolite concentrations. Lotion, cosmetic, and cologne/perfume use were associated with the greatest increases in the molar sum of phthalate metabolite and paraben concentrations, although the magnitude of individual biomarker increases varied by Product used. For example, women who used lotion had BP concentrations 111% higher (95% confidence interval (CI): 41%, 216%) than non-users, whereas their MBP concentrations were only 28% higher (CI: 2%, 62%). Women using cologne/perfume had MEP concentrations 167% (CI: 98%, 261%) higher than non-users, but BP concentrations were similar. We observed a monotonic dose–response relationship between the total number of Products used and urinary paraben and phthalate metabolite concentrations. These results suggest that questionnaire data may be useful for assessing exposure to a mixture of chemicals from Personal Care Products during pregnancy.

Science of The Total Environment, 2020Co-Authors: Shushen Liu, Fu Chen, Zejun WangAbstract:Abstract Hormesis describes a specific phenomenon in a biphasic concentration-response curve: low concentrations stimulate a response, while high concentrations suppress it. Hormesis could be influenced by several environmental factors, e.g. pH. In this study, the concentration-response/bioluminescence inhibition profiles (CRPs) of six components in Personal Care Products to Vibrio qinghaiensis sp.-Q67 were measured at five different pH levels. When the exposure lasted for 0.25 h, CRPs of the six components at various pH levels were S-shaped, except ascorbic acid 2-glucoside (AA2G) at pH 10.5. When it lasted for 12 h, the CRPs were J-shaped, except AA2G at pH 6.5, 7.5, and 9.5. To rationally explain these changes in hormesis expressed by J-shaped CRP, four characteristic parameters, the minimum effect (Emin) and its corresponding concentration (ECmin), the median effective concentration (EC50), and the zero effect concentration point (ZEP, where the effect is 0 and the concentration is ZEP), were used to quantify the J-shaped CRP. The results indicated that these parameters vary with pH. Additionally, ZEP showed an excellent linear relationship with EC10 (R2 = 0.9994) at all pH levels, indicating that EC10 could replace the no-observed effective concentration (NOEC) in ecological risk assessment. Furthermore, to elucidate the possible mechanism of hormesis, the binding of the components to the luciferase receptors was analyzed using molecular docking technology. The results showed that the components displaying hormesis bind more easily to the α subunit of luciferase than to the β subunit.

In this study, 23 common and widely used toner, skin water, and make-up water (TSM) commodities were directly taken as mixture samples, and Vibrio qinghaiensis sp.-Q67 (Q67) was used as the test organism. The toxicities of the TSMs to Q67 were determined via microplate toxicity analysis at 0.25 h and 12 h. Each TSM commodity can be regarded as a complicated mixture (relative concentration is 1). It was shown that the concentration-response curves (CRCs) of 23 TSMs are monotonic sigmoid-shaped (S-shaped) at 0.25 h, the CRCs of six TSMs are also S-shaped but the other 17 TSMs are non-monotonic hormetic or J-shaped at 12 h. In addition, to effectively characterize the nature of the stimulation and inhibition phases, it is suggested that five parameters such as the ECL (the median stimulation effective concentration (left)), Emin (the maximum stimulation effect), ECmin (the maximum stimulation effective concentration), ZEP (zero effect point where the effect is 0 and the concentration is ZEP), and EC50 can depict the non-monotonic CRC. To the best of our knowledge, this is the first study about the hormetic CRCs of commercial PCP mixtures.

We utilized questionnaire data from the National Health and Nutrition Examination Survey 2009-2012 cycles to examine the associations between use of sunscreen and mouthwash and urinary concentrations of phthalate metabolites and phenols in a nationally representative population of US adults (n=3529). Compared with individuals who reported "Never" using mouthwash, individuals who reported daily use had significantly elevated urinary concentrations of mono-ethyl phthalate, methyl and propyl parabens, and BP3 (28%, 30%, 39%, and 42% higher, respectively). Individuals who reported "Always" using sunscreen had significantly higher urinary concentrations of triclosan, methyl, ethyl, and propyl parabens, and BP3 (59%, 92%, 102%, 151%, and 510% higher, respectively) compared with "Never" users of sunscreen. Associations between exposure biomarkers and sunscreen use were stronger in women compared with men, and associations with mouthwash use were generally stronger in men compared with women. These results suggest that sunscreen and mouthwash may be important exposure sources for EDCs.
