Effect of boldo (Peumus Boldus M.) pretreatment on kinetics of supercritical CO2 extraction of essential oil

Del ValleAbstract:Abstract This work examined the effect of the solid matrix on supercritical carbon dioxide (SC CO 2 ) extraction of essentials oils from boldo leaves ( Peumus Boldus M.) subjected to rapid decompression of a CO 2 -impregnated sample, conventional milling, and low-temperature milling. Low-temperature conditioning prior to milling decreased heat-driven losses of volatile compounds during milling, as attested by a higher extract yield for low-temperature (10.8 g extract/kg extract-free substrate) than conventionally (9.63 g/kg) milled sample. Extract yield was even larger for the rapidly decompressed sample (11.4 g/kg). Results of SC CO 2 extraction experiments carried out at 40 °C and 10 MPa were adjusted to a diffusional model using the effective diffusivity of the extract in the solid matrix ( D e ) and a single partition of essential oils between solid substrate and SC CO 2 as best-fitting parameters. A microstructural factor ( F M ), which was estimated as the ratio between the binary diffusion coefficient of the essential oil in SC-CO 2 under extraction conditions and D e , was used to characterize the effect of sample pretreatment on extraction rates. Values of F M for rapidly decompressed (202) and low-temperature milled (1740) samples were smaller than value for conventionally milled samples (2200), which revealed that the two first treatments disrupted secretory cavities in boldo leaves more effectively than the third. This was confirmed by light microscopy observations. The work included also measurements using oregano bracts ( Origanum vulgare L.) as the substrate to confirm literature reports on the SC CO 2 extraction of pretreated bracts and to serve as a reference to our main results with boldo leaves. Trends with oregano coincided with those of boldo.

Recovery of antioxidants from boldo (Peumus Boldus M.) by conventional and supercritical CO2 extraction

Asencio, José Miguel AguileraAbstract:Abstract Boldo ( Peumus Boldus M.) is a native plant from Chile that has been used is traditional medicine to treat digestive and/or hepatobiliary disorders. An attempt was made in this work to extract antioxidant compounds from boldo using conventional solvents and supercritical carbon dioxide (SCCO 2 ). Yield of soluble solids (SS), boldine, and compounds with antioxidant activity varied depending on part plant, solvent and extraction conditions. Following 24 h extraction, the highest yield was 33.4% (w/w, dry basis) of SS with 70:30 ethanol/water (v/v) mixture at 70 °C. Yield of alcoholic extractions increased as the temperature was raised from 20 to 70 °C, but the total antioxidant activity (free radical DPPH method) was maximal in boldo leaf extracts with aqueous ethanol at 50 °C (1648 TEAC/g dry leaf). Boldine concentration was maximal (12%) in boldo bark extracted with acidulated water, and decreased with extraction time when methanol or ethanol/water mixtures were used. No boldine was detected in boldo leaf extracts. The contribution of boldine to the antioxidant activity of boldo extracts was low, specially when using aqueous ethanol as solvent, and/or boldo leaf instead of boldo bark as substrate. Yield of SCCO 2 extraction was ≈3% SS. Only non-polar compounds with limited antioxidant activity (0.08 TEAC/g extract) were extracted by SCCO 2 .

Los tratamientos con mayor mortalidad mostraron también una baja emergencia de insectos adultos (F 1) y menor pérdida de peso del grano. En el control de estados inmaduros la menor F1 se observó en las concentraciones más altas de polvo. El almacenamiento del polvo en refrigeración no impidió la pérdida en el tiempo de las propiedades insecticidas. Effect of vacuum storage on shelf life of a grain protector based on Peumus Boldus Molina foliage powder and lime against Sitophilus zeamais Motschulsk

Concepción RodríguezAbstract:The maize weevil (Sitophilus zeamais Motschulsky, Coleoptera: Curculionidae) is a key pest of stored grain maize. As an ecological pest control alternative, the use of botanical insecticides, such as powder from Boldus (Peumus Boldus Molina) foliage singly or mixed with lime, has been evaluated. Unfortunately, its shelf life is very short and does not exceed 15 d. The effectiveness of vacuum storage on insecticidal properties of a natural grain protector produced with Boldus powder:lime at proportions of 50:50 and 60:40 against adults of S. zeamais was assessed under laboratory conditions. Treatments were evaluated at 1% and 2% (w/w) for 150 d of storage. All treatments based on Boldus powder kept the level of mortality by contact activity over 80% at 150 d of storage. The highest toxicity, as a fumigant, was observed in treatments 50:50 at 2% and 60:40 at 1% and 2% with mortality over 60%. The grain weight loss was less than 1% and seed germination was not affected. With the exception of 0:100 at 2% without vacuum storage, all treatments were repellent to S. zeamais. Vacuum storage extended shelf life of the grain protector for 150 d.

Smith and Helicoverpa zea Boddie were evaluated under laboratory conditions in concentrations of 0.25, 0.5, 1.0, 2.0, 4.0, and 8.0% (w/w). Spodoptera frugiperda was the most susceptible with 75% mortality at 7 d at 8% P. Boldus concentration, while H. zea had only 30% mortality. LC50 was 2.31 mL kg-1 for S. frugiperda and 16.05 mL kg-1 for H. zea. When the extract concentration increased in the diet, larval size and weight, percentage of pupation and number of adults decreased, and the time required to reach those states was greater. Neonate larvae fed primarily on the diet with the lower extract concentration and the control was preferred by more than 50% of larvae. Inhibition of feeding, growth, weight gain of 3rd instar larvae as well as new biomass production decreased with concentration of the extract.

Bioactivity of Boldo (Peumus Boldus Molina) (Laurales: Monimiaceae) on Spodoptera frugiperda (J.E. Smith) and Helicoverpa zea (Boddie) (Lepidoptera: Noctuidae)

BlancoAbstract:Abstract. Insecticidal properties of powdered boldo, Peumus Boldus Molina, were evaluated against larvae of fall armyworm, Spodoptera frugiperda (J. E. Smith), and corn earworm, Helicoverpa zea (Boddie). Bioassays assessed development, feeding preferences, and mortality of neonate and third instars. For both species, most mortality (65 and 67.5%, respectively) was obtained with 8% boldo concentration incorporated into an artificial insect diet, and the LC50 and LC90 for fall armyworm were 6.8 and 25.9 g boldo kg-1 diet and 3.8 and 35.6 g boldo kg-1 diet for corn earworm. With increased concentration of boldo, larvae were shorter, weighed less, and had anatomical abnormalities, and fewer pupated. Concentrations of 4 and 8% boldo resulted in fewer adults of both species. In feeding preference tests, neonates selected the diet with the least concentration of boldo powder, and larvae fed less with higher concentrations.
