    Abstract:

    Abstract Ethnopharmacological relevance Phyllanthus amarus Schum. & Thonn. belongs to the family Euphorbiaceae is a small herb well known for its medicinal properties and widely used worldwide. P. amarus is an important plant of Indian Ayurvedic system of medicine which is used in the problems of stomach, genitourinary system, liver, kidney and spleen. It is bitter, astringent, stomachic, diuretic, febrifuge and antiseptic. The whole plant is used in gonorrhea, menorrhagia and other genital affections. It is useful in gastropathy, diarrhoea, dysentery, intermittent fevers, ophthalmopathy, scabies, ulcers and wounds. Materials and methods The present review covers a literature across from 1980 to 2011. Some information collected from traditional Ayurvedic texts and published literature on ethanomedicinal uses of Phyllanthus amarus in different countries worldwide. Results Phytochemical studies have shown the presence of many valuable compounds such as lignans, flavonoids, hydrolysable tannins (ellagitannins), polyphenols, triterpenes, sterols and alkaloids. The extracts and the compounds isolated from P. amarus show a wide spectrum of pharmacological activities including antiviral, antibacterial, antiplasmodial, anti-inflammatory, antimalarial, antimicrobial, anticancer, antidiabetic, hypolipidemic, antioxidant, hepatoprotective nephroprotective and diurectic properties. Conclusion The present review summarizes information concerning the morphology, ecology, ethnopharmacology, phytochemistry, biological activities, clinical applications and toxicological reports of P. amarus . This review aims at gathering the research work undertaken till date on this plant in order to provide sufficient baseline information for future works and commercial exploitation.

    Chromatographic fractions obtained from Phyllanthus amarus were tested for toxicity on the serum biochemistry of rats. The results revealed that some fractions of P. amarus had potentially deleterious effects on the blood and therefore caution should be exercised in the use of P. amarus as a medicinal plant.

    The leaves of two plants in the family Euphorbiaceae; Phyllanthus amarus Schum and Thonn and Euphorbia hirta Linn, which are commonly used for medicinal purposes and also occurring among Nigerian pasture were analyzed using spectrophotometric and titrimetric methods to ascertain their levels of inorganic substances such as nitrate, nitrite, cyanide; and metallic constituents such as copper, manganese, magnesium, iron, zinc, calcium, sodium and potassium. High levels of Mn (189 and 465 ppm), Cu (30.5 and 21.8 ppm), Zn (151.9 and 75.1 ppm), and NO 3 (4,600 and 7,200ppm) were found in the leaves of E. hirta and P. amarus respectively. No traces of cyanide and nitrite were found in the leaves of these two plants. Varying proportions of Fe (2,225 and 1,563ppm), Mg (5,500 and 5,800ppm), K (33,000 and 32,000ppm), Ca (21,000 and 26,500ppm), and Na (558.1 and 125 ppm) were found in the leaves of E. hirta and P. amarus respectively. The levels of chemicals are high enough to constitute a source of toxicosis to animals consuming them and their use for medicinal purpose should also be a source for concern most especially if the fresh leave decoctions are used.   • Evaluation of Phyllanthus amarus and Phyllanthus maderaspatensis as agents for postexposure prophylaxis in neonatal duck hepatitis B virus infection

    The therapeutic potential of plant extracts of Phyllanthus amarus and Phyllanthus maderaspatensis for postexposure prophylaxis against infection by Hepadnaviruses was studied in ducklings infected by the duck hepatitis B virus (DHBV). Forty-four Pekin ducklings were inoculated intraperitoneally with DHBV at 24 hr posthatch. They were treated by intraperitoneal injection of Phyllanthus amarus (aqueous extract) (100 mg/kg body weight) or Phyllanthus mad eraspatensis (alcoholic extract) (100 mg/kg body weight) for a period of 4 weeks. Infected ducklings treated with saline served as controls. Weekly serum samples obtained before, during, and after treatment were analysed for the presence of DHBV DNA in serum by dot blot hybridisation using α 32P-labelled probes. Liver tissue was collected after killing the ducks at various time intervals and was studied for replicative status of the viral DNA and liver histopathology; 17 of 21 ducks were viraemic on completion of treatment with Phyllanthus amarus. At 16 week posttreatment follow-up four of seven animals remained viraemic. Similar results were obtained with Phyllanthus maderaspatensis. There was no alteration in DHBV replication in the liver. No toxicity was observed with this treatment. These observations suggest that Phyllanthus amarus and Phyllanthus maderaspatensis are not useful as therapeutic agents for postexposure prophylaxis against DHBV infection. © 1993 Wiley-Liss, Inc.

    A simple, specific and precise high performance liquid chromatographic method has been developed and validated for the estimation of Phyllanthin, an important bitter lignan present in Phyllanthus amarus. Furthermore, the developed method was used to successfully quantify the Phyllanthin in the plant extract also. The mobile phase optimized for the RP-HPLC was methanol-water 66:34 (%v/v) and that was very simple and cost effective. The detection is carried out using variable wavelength UV–VIS detector set at 229 nm. Linearity for the developed method was found over the concentration range 1 – 50 μg / ml with a correlation coefficient of 0.999. Method was validated according to ICH guidelines and found to be suitable for rapid quantification of Phyllanthin in bulk and in plant extracts.     Abstract Phyllanthus amarus Schum. et Thonn. (Bhuia amla; Euphorbiacae) is a herb common to central and southern India. It is an ayurvedic herb and has a wide range of traditional uses in different diseases. The aim of this work was to evaluate the hepatoprotective effect of ethanolic extract of Phyllanthus amarus (Phyllanthus amarus) on aflatoxin B1-induced liver damage in mice using different biochemical parameters and histopathological studies. Aflatoxin was administered orally (66.6 μg kg−1 BW 0.2 ml−1 day−1) to the mice of each group except control to which normal saline and ascorbic acid (0.1 g kg−1 BW 0.2 ml−1 day−1) were given, respectively. Ethanolic extract of Phyllanthus amarus (0.3 g kg−1 BW 0.2 ml−1 day−1) was given to all groups except control groups (gp. I and gp. V) after 30 min of aflatoxin administration. The entire study was carried out for 3 months and animals were sacrificed after an interval of 30 days till the completion of study. Phyllanthus amarus extract was found to show hepatoprotective effect by lowering down the content of thiobarbituric acid reactive substances (TBARS) and enhancing the reduced glutathione level and the activities of antioxidant enzymes, glutathione peroxidase (GPx), glutathione-S-transferase (GST), superoxide dismutase (SOD) and catalase (CAT). Histopathological analyses of liver samples also confirmed the hepatoprotective value and antioxidant activity of the ethanolic extract of the herb, which was comparable to the standard antioxidant, ascorbic acid. The overall data indicated that Phyllanthus amarus possesses a potent protective effect against aflatoxin B1-induced hepatic damage, and the main mechanism involved in the protection could be associated with its strong capability to reduce the intracellular level of reactive oxygen species by enhancing the level of both enzymatic and non-enzymatic antioxidants.
