    Abstract Compatibility of pacu, Piaractus brachypomus with Indian major carps was evaluated in grow-out carp polyculture system for 4 months in 12 large concrete tanks. Four different species combinations were evaluated i.e., control: catla (Catla catla), rohu (Labeo rohita) and mrigal (Cirrhinus mrigala); T1: rohu, mrigal and pacu (P. brachypomus); T2: catla, mrigal and pacu and T3: catla, rohu and pacu. The tanks were stocked at the combined stocking density of 2 fingerlings m−3 with equal species ratio of the component species. Incorporation of pacu in carp polyculture system did not affect survival of any of the three major carps irrespective of species combination. Compatibility was observed between mrigal and pacu in the grow-out system, while catla showed inter-specific competition with the later at a level similar to that it receives from rohu. The study further hinted possible overlapping of the feeding niche of rohu and pacu in which the later gets the advantage.

    Abstract:

    The pirapitinga, Piaractus brachypomus (Characiformes, Serrasalmidae), is a fish from the Amazon basin and is considered to be one of the main native species used in aquaculture production in South America. The objectives of this study were: (1) to perform liver transcriptome sequencing of pirapitinga through NGS and then validate a set of microsatellite markers for this species; and (2) to use polymorphic microsatellites for analysis of genetic variability in farmed stocks. The transcriptome sequencing was carried out through the Roche/454 technology, which resulted in 3,696 non-redundant contigs. Of this total, 2,568 contigs had similarity in the non-redundant (nr) protein database (Genbank) and 2,075 sequences were characterized in the categories of Gene Ontology (GO). After the validation process of 30 microsatellite loci, eight markers showed polymorphism. The analysis of these polymorphic markers in farmed stocks revealed that fish farms from North Brazil had a higher genetic diversity than fish farms from Southeast Brazil. AMOVA demonstrated that the highest proportion of variation was presented within the populations. However, when comparing different groups (1: Wild; 2: North fish farms; 3: Southeast fish farms), a considerable variation between the groups was observed. The FST values showed the occurrence of genetic structure among the broodstocks from different regions of Brazil. The transcriptome sequencing in pirapitinga provided important genetic resources for biological studies in this non-model species, and microsatellite data can be used as the framework for the genetic management of breeding stocks in Brazil, which might provide a basis for a genetic pre-breeding programme.

    The effect of two semen collections performed in the same breeding season on the sperm parameters and physicochemical composition of the seminal plasma of Piaractus brachypomus was evaluated. The semen of nine hormone-induced males with hypophysial carp extract (HCE) was collected in April and May 2014. Changes between semen collections in seminal volume (SV), sperm motility (SM), motility duration (MD), sperm concentration (SC) and biochemical composition (Na+, Ca2+, Mg2+, Cl-, K+, glucose, triglycerides, protein, osmolarity and pH) were evaluated. Significant differences were observed between collections for Na+, triglycerides and osmolarity (p<0.05) and Ca2+ (p<0.01). On the other hand, no significant differences were found in SV, SM, MD and SC between collections. The collection of semen from a hormonally induced male in two occasions in one reproductive season is a viable alternative for P. brachypomus, although plasma components may vary due to sampling.Se evaluó el efecto de dos colectas de semen realizadas en la misma temporada reproductiva sobre los parámetros espermáticos y composición fisicoquímica del plasma seminal de Piaractus brachypomus.     El Triclorfon (TCF) es un insecticida organofosforado ampliamente utilizado en sistemas de produccion agricola y pecuario; sin embargo, su uso indiscriminado ha generado serios problemas en ecosistemas acuaticos afectando organismos que no son blanco directo del compuesto quimico. Por este motivo, el presente estudio tuvo como objetivos evaluar la toxicidad aguda (CL 50) y cronica subletal del TCF en alevinos de cachama blanca ( Piaractus brachypomus ). La toxicidad aguda se evaluo a 96 horas, empleando cuatro concentraciones de TCF (0,01, 0,1, 0,25, 0,5 mg/L) y un grupo control, cada grupo consto de 8 animales (2,5 ± 0,5 g). The LC50 value calculated using the Trimmed Spearman-Karber software was 0.18 mg/L (p<0,05). The evaluation of the effect of chronic exposure to sublethal TCF on the immune response of P. brachypomus was performed by testing respiratory burst, showing significant increase in animals exposed to 0.029 mg / L during the first 10 days of exposure compared to control. Likewise, the plasma bactericidal capacity was evaluated in TCF exposed fish using Aeromonas hydrophila, finding greater of colony forming units on day 20 and 30 of exposure to the concentration 0.018 mg/L as opposed to the control. In addition, a histopathological study was performed, finding gill lesions compatible with epithelial cell hypertrophy and lamellar detachment in fish exposed to 0,05 mg/L on day 30 of exposure. This work showed the cachama blanca as a biomarker in contaminations associated with these xenobiotics.
