    Abstract:

    Plectranthus bishopianus has recently been treated as a synonym of Plectranthus deccanicus . However, a detailed study of the type materials, recently collected specimens, as well as SEM analysis of pollen and mericarp of both species suggests that P. bishopianus is a distinct species. The specific status of P. bishopianus is thus reinstated, and it can be easily distinguished from P. deccanicus by the stem nature, leaf size and shape, cyme length, number of flowers per cyme, the structure of flowering and fruiting calyx and mericarp surface. Additionally, complete taxonomic descriptions, photographs and conservation assessments, along with notes on distribution and habitat of both species, are provided.

    A new species, Plectranthus sahyadricus is described from Munnar, southern Western Ghats, India. The species shows similarity with Plectranthus beddomei, in its stem and leaf characters but differs mainly by presence of a ring of hairs at young nodes, floral characters like cyme length, number of flowers per cyme, structure of the fruiting calyx tube, anterior corolla lip not fully opened and form an inverted dome over mouth, presence of hairs inside corolla tube and size of the disc. Detailed description, IUCN red list category and other relevant notes are provided.

    Taxonomic identity of Plectranthus montanus , a poorly known species from India is discussed by providing a detailed description, photographs and notes on its distribution. The specific status of Coleus subbaraoi is also discussed here. Critical study reveals that Coleus subbaraoi is conspecific with Plectranthus montanus and it is thus synonymized.

    A new species, Plectranthus anamudianus (Lamiaceae) is described and illustrated from southern Western Ghats, India. The new species is similar to Plectranthus glabratus, in its inflorescence architecture but differs mainly by its habit, internode length, leaf size and shape, petiole length and texture, number of flowers per cyme, bract structure and recurved fruiting calyx. Detailed description, illustration, photographs, IUCN red list category, distribution map, notes on biogeography and other relevant notes are provided. Surface sculpturing pattern of pollen grains and mericarps of the two species are also compared.

    The Plectranthus genus is commonly used in traditional medicine due to its potential to treat several illnesses, including bacterial infections and cancer. As such, aiming to screen the antibacterial and cytotoxic activities of extracts, sixteen selected Plectranthus species with medicinal potential were studied. In total, 31 extracts obtained from 16 Plectranthus spp. were tested for their antibacterial and anticancer properties. Well diffusion method was used for preliminary antibacterial screening. The minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) values of the five most active acetonic extracts (P. aliciae, P. japonicus, P. madagascariensis var. “Lynne”, P. stylesii, and P. strigosus) were determined. After preliminary toxicity evaluation on Artemia salina L., their cytotoxic properties were assessed on three human cancer cell lines (HCT116, MCF-7, and H460). These were also selected for mechanism of resistance studies (on NCI-H460/R and DLD1-TxR cells). An identified compound—parvifloron D—was tested in a pair of sensitive and MDR-Multidrug resistance cancer cells (NCI-H460 and NCI-H460/R) and in normal bronchial fibroblasts MRC-5. The chemical composition of the most active extract was studied through high performance liquid chromatography with a diode array detector (HPLC-DAD/UV) and liquid chromatography–mass spectrometry (LC–MS). Overall, P. strigosus acetonic extract showed the strongest antimicrobial and cytotoxic potential that could be explained by the presence of parvifloron D, a highly cytotoxic diterpene. This study provides valuable information on the use of the Plectranthus genus as a source of bioactive compounds, namely P. strigosus with the potential active ingredient the parvifloron D.

    Plants from the Plectranthus genus are commonly known for their medicinal properties, and their applications are vast in traditional medicine. Often accountable for these properties, bioactive secondary metabolites such as diterpenes are present in these plants, and some possess cytotoxic properties. Abietane diterpenes 7α-acetoxy-6β-hydroxyroyleanone, 7β,6β-dihydroxyroyleanone, 6,7- dehydroroyleanone, and parvifloron D are examples of such class of bioactive diterpenes. These abietanes are obtained from Plectranthus genus plants as their major compounds. In this work, the cytotoxic properties of these abietanes have been studied. This property was evaluated through MTT assays on A549 cells and CCRF-CEM cells. In addition, the level of intracellular Reactive Oxygen Species (ROS) in these cells was also measured. Our study has revealed that on CCRF-CEM cells, there is a slight increase of the ROS levels after exposure to all compounds. Nevertheless, on A549 cells the levels of ROS increase approximately 10-fold, with notorious increase after exposure to 7β,6β-dihydroxyroyleanone, which can induce cytotoxicity due to ROS accumulation on cells. In conclusion, this study proves the therapeutic potential of Plectranthus-derived diterpenes and validates its use in traditional medicine.

Generic delimitation follows a recently published molecular phylogeny which identified Coleus as the sister of the remaining genera of Subtribe Plectranthinae; Plectranthus as sister to Tetradenia Benth. and Thorncroftia N.E.Br., and a separate phylogenetically distinct genus Equilabium comprising species previously placed in Plectranthus. In this treatment, 294 species of Coleus, 42 of Equilabium, and 72 of Plectranthus are recognized. All but one of the combinations in Equilabium are new as only the genus and type species have been previously published. Two-hundred and twelve names are changed to combinations in Coleus from Plectranthus, Pycnostachys Hook. and Anisochilus Benth.

    Lamiaceae subtribe Plectranthinae, a palaeotropical group of just over 450 species with mainly zygomorphic flowers and stamens that are contiguous at the point of insertion at the base of the lower corolla lip, include the medicinally and horticulturally important genus Plectranthus. Plectranthus currently includes the formerly recognized Coleus and Solenostemon. A phylogenetic analysis of the group is presented based on rps16, trnL-F and trnS-G regions of the plastid genome. Plectranthus as currently recognized is paraphyletic; a clade containing the type of Coleus and including Solenostemon, Pycnostachys and Anisochilus is sister to the rest of the group. Three endemic and monotypic Madagascan genera, Dauphinea, Madlabium, Perrierastrum and the Madagascan Capitanopsis belong to a single clade and are recognized under Capitanopsis; the new combinations are made here. Plectranthus s.s. is sister to a clade comprising Thorncroftia and Tetradenia. Tetradenia, unlike any other members of Plectranthinae, has actinomorphic corollas and is usually dioecious. A group of other species previously recognized as Plectranthus form a clade separate from Plectranthus s.s. and is recognized as Equilabium gen. nov. Estimates of clade age suggest that the genera begin to diversify from the mid to late Miocene. Plectranthinae are found in dry woodlands, montane grasslands and evergreen forest margins. Shifts between habitats occur in most clades, although significantly fewer than if the changes were random. The distribution of the clades in the major habitats is examined. Migration in Plectranthinae was from Africa to Madagascar and Asia, and there is no evidence of migration back to Africa. The phylogenetic pattern of medicinal use in Plectranthinae is weak, and issues surrounding this are discussed.

    Abstract The chemical composition of the essential oils of Plectranthus cylindraceus and Plectranthus arabicus grown in Saudi Arabia were analyzed using gas chromatography techniques (GC–MS, GC–FID, Co-GC, LRI determination, and database and literature searches) using two different stationary phase columns (polar and nonpolar). The analysis led to the characterization of a total of 157 different compounds from both oils. In the oil derived from P. cylindraceus , 79 compounds were identified, whereas 132 compounds were identified in the oil derived from P. arabicus ; these compounds account for 95.2% and 98.4% of the total oil compositions, respectively. The major constituents of P. cylindraceus oil were patchouli alcohol (55.5 ± 0.01%), 1,8-cineole (6.0 ± 0.01%) and valerianol (3.8 ± 0.18%), whereas, the main compounds of the P. arabicus oil were 1,8-cineole (50.5 ± 1.37%), β- pinene (7.0 ± 0.08%), camphor (6.3 ± 0.19%) and β -myrcene (4.1 ± 0.10%). To the best of our knowledge, patchouli alcohol found in high concentration in the P. cylindraceus oil has never been reported from the genus Plectranthus . Moreover, this is the first phytochemical study of P. arabicus.
