    Abstract:

    In this study we determined the effect of insulin injection and /or short fasting on the induction of Pregnancy Toxemia in Zariabi goats. Eighteen twin-bearing Zaraibi goats (around 130 days of gestation) were divided into control group (Group C, n = 6) fed ad labtium, feed deprived only group (Group A, n = 6) and feed deprived with insulin injection group (Group B, n = 6). Clinical signs of Pregnancy Toxemia appeared after 24 and 72 h in group B and A, respectively. Refeeding and 50 ml of 500 gm dextrose /l were injected after appearance of the clinical signs every 12 h for two successive doses. Plasma concentrations of glucose and β-hydroxybutyrate were significantly (p 0.1) at 48 h of refeeding and injection of dextrose in both experimentally induced groups. There was significant (P < 0.05) decrease in the length of gestation in both experimental groups (A & B) than that of control one. The lengths of gestation were 140.3 ± 1.1, 139.2 ± 0.8 and 149.1 ± 1.4 days in group A, B and C, respectively. Simple protocol of short fasting combined with insulin injection can induce Pregnancy Toxemia and this protocol might be used for valuable studies for Pregnancy Toxemia in twin-bearing Zaraibi goats.

    Abstract:

    Hyperketonemia and hypoglycemia are more common obvious biochemical features of Pregnancy Toxemia as well as liver and kidney may be involved in the pathogenesis of Toxemia. Fifteen pregnant goats with twins (3-4 years, 20-27 kg body weight and 120-130 days of gestation) were classified into two groups, control one consists of six goats and experimental group consists of nine goats for induction of Pregnancy Toxemia by the stress of fasting with access of water until the symptoms of Pregnancy Toxemia were appeared within 72 hours. Serum samples were obtained and analyzed for βhydroxybutyrate, glucose, total protein, albumin, globulin, urea, creatinine, AST, ALT, total lipids, cholesterol, calcium, magnesium, phosphorus, sodium and potassium using kits, while insulin, cortisol, T3, T4, growth hormone were measured by radioimmunoassay. Hematological profile was investigated on whole blood samples. β-hydroxybutyrate, AST, ALT, urea, creatinine, cortisol and insulin hormones were significantly higher in toxemic goats than those of control ones while glucose, sodium, potassium, calcium, magnesium, T4, total protein, globulin, albumin, cholesterol and total lipids values were significantly lower in toxemic goats than those of control ones while, there were no significant changes in phosphorous, T3 and growth hormone. A significant increase in leucocytes, hemoglobin, packed cell volume and neutrophils and a significant decrease in lymphocytes were observed in Pregnancy toxemic goats than those of control ones. Clinical chemistry of Pregnancy Toxemia can affect hormonal, electrolytes and mineral balance as well as immune and hematological picture in goats.

    Pregnancy Toxemia was induced in nine pregnant goat does with twins by the stress of fasting with access to water in late Pregnancy to investigate the effect of Pregnancy Toxemia on immunoglobulins (IgA, IgM, and IgG), cortisol, insulin, thyroid, and growth hormones and their correlations with the plasma levels of glucose and β-Hydroxybutyrate. Plasma samples were collected at 0, 12, 24, 36, 48, and 72 hours after induction of Pregnancy Toxemia. The result revealed that experimental animals developed neurological findings with convulsions and acetone odor from the mouth with recumbency after 72 hours. Laboratory findings showed a significant increase in β-Hydroxybutyrate, cortisol, and insulin while there were significant decreases in glucose, thyroid, and immunoglobulins (IgA, IgM, and IgG). Plasma glucose concentrations had significant negative correlations with β-hydroxybutyrate, cortisol, and insulin while the correlations were significantly positive with immunoglobulins and thyroid hormone. Plasma β-hydroxybutyrate concentration was significantly positively correlated with cortisol and negatively correlated with immunoglobulins, insulin, and thyroid hormone. From this study we can conclude that Pregnancy Toxemia might affect humoral immune responses as well as insulin, cortisol, and thyroid hormones. Moreover, insulin might have a compensatory role to increase suppressive effect on ketogenesis in experimentally pregnant toxemic goats.

    BACKGROUND Pregnancy Toxemia is a common metabolic disease of periparturient small ruminants. Information on its effects on metabolism and perinatal adaptation of newborn lambs and kids is lacking. OBJECTIVES Evaluate differences in morbidity, mortality, and common biochemical and hematologic variables between Pregnancy Toxemia kids (PT) and control kids (CON). ANIMALS Sixteen kids born to does being treated at the hospital for Pregnancy Toxemia (blood beta-hydroxybutyrate concentration [BHB] > 1.2 mmol/L) and 12 kids from healthy dams (dam BHB < 1.2 mmol/L) that kidded at the hospital. METHODS In this cohort study, serial measurements of blood L-lactate, glucose, and BHB concentrations, arterial blood gases, hematocrit, total protein concentrations, nonesterified fatty acids (NEFAs) concentrations, and body weight were compared between groups over the first 72 hours of life. Long-term follow-up was performed after 3 months. RESULTS Pregnancy Toxemia kids were more likely to require tube feeding at 0 and 12 hours (relative risk 7.7 [1.13, 52.45] and 2.8 [1.39, 5.65]). Pregnancy Toxemia kids were more acidemic (7.26 ± 0.069 vs 7.34 ± 0.079, P = .003) and hyperlactatemic (8.17 ± 2.57 vs 5.48 ± 2.71, P = .003) at birth than CON kids. Control kids were 1.1 [1.01, 1.77] times more likely to survive to discharge and 2.2 [1.15, 4.20] times more likely to survive to 3 months than PT kids. CONCLUSIONS AND CLINICAL IMPORTANCE Pregnancy Toxemia kids had higher short- and long-term mortality and were more likely to require perinatal intervention. Weight loss in the first few days could be a useful predictor of nonsurvival.

    Abstract:

    Pregnancy Toxemia (PT) is considered one of the most common metabolic diseases with high impact on the production of small ruminants. The objective of this study was investigate possible myocardial damage in goats affected with PT by the determination of serum myocardial biomarkers CK-MB and cTnI. A total of 44 goats affected with PT, and 10 apparently healthy goats (control group or CG) were used in the study. In goats with PT, the serum concentrations of cTnI (0.43 ng/mL) were significantly higher than that in CG goats (0.06 ng/mL). Although CK-MB showed no significant difference, it was approximately three times higher in animals with PT. The serum concentrations of insulin were significantly lower in PT goats (5.03 ppmol/L) compared to CG goats (10.66 pmol/L). The serum concentrations of cortisol in PT goats (155.41 nmol/L) were significantly higher than that in CG goats (36.58 nmol/L). Results of this study indicate that a clinically significant myocardial damage might occur in goats affected with PT leading to significant elevations in values of cTnI and CK-MB. Therefore, these parameters could be used as a potential prognostic indicator in goats affected with this important disease.

    Abstract:

    ABSTRACT: The objective of the present study was to characterize the biochemical, hormonal, and mineral profile, and histopathology of the liver and kidneys, related to the severity of natural cases of Pregnancy Toxemia (PT) in sheep. A total of 45 sheep with PT were analyzed at the “Clínica de Bovinos”, Campus Garanhuns-UFRPE. The animals were submitted to clinical examination, followed by the collection of blood and urine. A necropsy was performed on thirteen animals that died and a histopathological examination was performed on samples of liver and kidneys. Increased creatinine, urea, glucose, fructosamine, non-esterified fatty acids, β-hydroxybutyrate, cortisol, chlorine, amylase, aspartate aminotransferase, gamma-glutamyltransferase, and folic acid were found, whereas insulin, potassium, and total and ionizable calcium presented low values for the species. Total protein, albumin, globulin, sodium, magnesium, and vitamin B12 remained within the normal range. In the macroscopic analysis of the liver, it was possible to observe an increase in organ and border size, yellowish coloration and parenchyma that varied from firm to friable. Vacuolation of the hepatocytes and renal tubular cells was observed. Metabolic disorders in sheep associated with hepatic and renal lesions are more apparent in overweight animals. With the greater impairment in hepatic and renal function, the clinical prognosis of animals with a high body score should be considered reserved.

    Abstract:

    The aims of the present study were to determine serum levels of triiodothyronine (T3) and thyroxine (T4) in ewes diagnosed with Pregnancy Toxemia (PT) and evaluate the participation of these hormones in this type of metabolic disorder. Analyses were performed of the clinical cases of 24 ewes diagnosed with PT and treated at the Bovine Clinic of the University Federal Rural of Pernambuco, Campus Garanhuns, from 2007 to 2012. The animals were submitted to clinical and ultrasound exams. Blood and urine were collected for the laboratory exams. The main abnormalities were apathy, elevated body temperature, congested mucosa, dehydration, edema in the limbs, amaurosis, tachycardia, inappetence or anorexia and diminished ruminal motility. The mortality rate was 50%. Among the animals that died, 58.33% were hyperglycemic. The study of ketone bodies revealed ketonuria. Three conditions of glycemia were found in the ewes with PT. The laboratory findings revealed high levels of non-esterified fatty acids and β-hydroxybutyric acid. Serum levels of T3 (3.21 nmol/L) and T4 (69.16 nmol/L) were low. In conclusion, Pregnancy Toxemia in ewes is characterized by endocrine alterations that reflect a negative energy balance..
