a metabolomics guided exploration of marine natural Product Chemical space
Metabolomics, 2016Co-Authors: Dimitrios J Floros, Paul R Jensen, Pieter C Dorrestein, Nobuhiro KoyamaAbstract:Natural Products from culture collections have enormous impact in advancing discovery programs for metabolites of biotechnological importance. These discovery efforts rely on the metabolomic characterization of strain collections. Many emerging approaches compare metabolomic profiles of such collections, but few enable the analysis and prioritization of thousands of samples from diverse organisms while delivering chemistry specific read outs. In this work we utilize untargeted LC–MS/MS based metabolomics together with molecular networking to inventory the chemistries associated with 1000 marine microorganisms. This approach annotated 76 molecular families (a spectral match rate of 28 %), including clinically and biotechnologically important molecules such as valinomycin, actinomycin D, and desferrioxamine E. Targeting a molecular family produced primarily by one microorganism led to the isolation and structure elucidation of two new molecules designated maridric acids A and B. Molecular networking guided exploration of large culture collections allows for rapid dereplication of know molecules and can highlight producers of uniques metabolites. These methods, together with large culture collections and growing databases, allow for data driven strain prioritization with a focus on novel chemistries.

associations between prenatal maternal urinary concentrations of personal care Product Chemical biomarkers and childhood respiratory and allergic outcomes in the chamacos study
Environment International, 2018Co-Authors: Kimberly Berger, Antonia M. Calafat, Brenda Eskenazi, John R Balmes, Nina Holland, Kim G HarleyAbstract:Abstract Background Personal care Product Chemicals may be contributing to risk for asthma and other atopic illnesses. The existing literature is conflicting, and many studies do not control for multiple Chemical exposures. Methods We quantified concentrations of three phthalate metabolites, three parabens, and four other phenols in urine collected twice during pregnancy from 392 women. We measured T helper 1 (Th1) and T helper 2 (Th2) cells in their children's blood at ages two, five, and seven, and assessed probable asthma, aeroallergies, eczema, and lung function at age seven. We conducted linear and logistic regressions, controlling for additional biomarkers measured in this population as selected by Bayesian Model Averaging. Results The majority of comparisons showed null associations. Mono-n-butyl phthalate (MnBP) was associated with higher Th2% (RR: 10.40, 95% CI: 3.37, 17.92), and methyl paraben was associated with lower Th1% (RR: −3.35, 95% CI: −6.58, −0.02) and Th2% at borderline significance (RR: −4.45, 95% CI: −8.77, 0.08). Monoethyl phthalate was associated with lower forced expiratory flow from 25 to 75% of forced vital capacity (FEF25–75%) (RR: −3.22 L/s, 95% CI: −6.02, −0.34). Propyl paraben (OR: 0.86, 95% CI: 0.74, 0.99) was associated with decreased odds of probable asthma. Conclusions While some biomarkers, particularly those from low molecular weight phthalates, were associated with an atopic cytokine profile and poorer lung function, no biomarkers were associated with a corresponding increase in atopic disease.

urinary concentrations of phthalate metabolites bisphenols and personal care Product Chemical biomarkers in pregnant women in israel
Environment International, 2018Co-Authors: Ronit Machtinger, Russ Hauser, Antonia M. Calafat, Tamar Berman, Michal Adir, Abdallah Mansur, Andrea Baccarelli, Catherine Racowsky, Ravit NahumAbstract:Abstract Mounting evidence suggests possible adverse effects of intrauterine exposure to certain phenols and phthalates, two classes of endocrine disruptor Chemicals, on the developing fetus, with consequences into later life. These findings have contributed to the replacement of some Chemicals, such as di‑2‑ethylhexyl phthalate (DEHP) and bisphenol A (BPA), in consumer Products. For the current study we quantified urinary concentrations of biomarkers of exposure among 50 pregnant women in Israel to several phthalates, bisphenols and personal care Product Chemicals, as well as DEHP and BPA alternatives. We detected 14 of the 31 biomarkers in more than 90% of the women. We detected biomarkers of 1,2‑cyclohexane dicarboxylic acid, diisononyl ester (DINCH), bisphenol S, and bisphenol F not as frequently (27–56%). This study is the first to evaluate exposure to triclosan, bisphenols, parabens, and phthalates and BPA alternatives among Israeli pregnant women.

use of pooled samples to assess human exposure to parabens benzophenone 3 and triclosan in queensland australia
Faculty of Health; Institute of Health and Biomedical Innovation, 2015Co-Authors: Amy Heffernan, Antonia M. Calafat, Christine Baduel, Leisamaree Toms, Peter Hobson, Sara Broomhall, Jochen F MuellerAbstract:Parabens, benzophenone-3 and triclosan are common ingredients used as preservatives, ultraviolet radiation filters and antimicrobial agents, respectively. Human exposure occurs through consumption of processed food and use of cosmetics and consumer Products. The aim of this study was to provide a preliminary characterisation of exposure to selected personal care Product Chemicals in the general Australian population. De-identified urine specimens stratified by age and sex were obtained from a community-based pathology laboratory and pooled (n= 24 pools of 100). Concentrations of free and total (sum of free plus conjugated) species of methyl, ethyl, propyl and butyl paraben, benzophenone-3 and triclosan were quantified using isotope dilution tandem mass spectrometry; with geometric means 232, 33.5, 60.6, 4.32, 61.5 and 87.7. ng/mL, respectively. Age was inversely associated with paraben concentration, and females had concentrations approximately two times higher than males. Total paraben and benzophenone-3 concentrations are significantly higher than reported worldwide, and the average triclosan concentration was more than one order of magnitude higher than in many other populations. This study provides the first data on exposure of the general Australian population to a range of common personal care Product Chemical ingredients, which appears to be prevalent and warrants further investigation.

KADAR PROTEIN DAN SIFAT ORGANOLEPTIK NUGGET RAJUNGAN DENGAN SUBSTITUSI IKAN LELE (Clarias gariepinus) (Protein Levels and Organoleptic Crab Nugget with Substitution Catfish (Clarias gariepinus)
'LPPM Universitas Muhammadiyah Semarang', 2010Co-Authors: Ubaidillah Anas, Hersulistyorini WikanastriAbstract:Crab meat (Second-grade) is a sort of meat produced by crab processing industry which has export quality. Crab meat (second-grade) used to a Product which has value added but has not been optimal. One of effort to develop this Product is substitute the crab meat with catfish meat into crab nugget Product. Catfish is one source of animal protein which is cheaper but it has high nutrient. So the substitution is expected to be an affordable price of processed Product. The research object is a Product with a substitution crab nugget into catfish meat (Clarias gariepinus). The independent variables in this research were variations of substitution and dependent variable is the proportion of protein and flavor in crab nugget Product. Chemical analysis is carried out quantitative analysis of protein and flavor. The design used completely randomized design in three replication. The results showed that there are effects of substitution catfish meat and crab meat on protein crab nugget Product. While flavor of aroma, flavor and texture except color there are not effect on protein crab nugget Product. The highest protein of crab nugget Product is Product with substitution L0: R100 is 10.06% while Product with substitution L95: R5 has little protein about 8.15%. The result showed that favorite flavor of crab nugget Product is a Product which has substitution L65: R35 about 2.95 and substitution of Product with L0: R100 has the smallest value about 2.56. Key words: crab nugget, protein, substitution catfish
