Thorley-lawsonAbstract:IB4 is a prototype, latently Epstein-Barr virus (EBV)-infected, lymphoblastoid cell line. We show here that IB4 contains only integrated EBV genomes. Episomal EBV DNA is not detected by Gardella gel analysis or in situ hybridization. Restriction Enzyme Mapping indicates that the EBV genomes first circularized and then integrated into and deleted part of the BamHI C fragment. IB4 is therefore the only lymphoblastoid cell line described to date that lacks episomal EBV and has integrated EBV genomes with joined ends. Thus, the detection of joined EBV termini on Southern blots is not as reliable as the Gardella gel system for detecting episomal EBV DNA, and IB4 is not an ideal prototype cell line for the study of latent infection by EBV.

CarletonAbstract:The isolation and characterization of a hu- man T-cell lymphotropic retrovirus related to human T-cell lymphotropic virus type I (HTLV-I) from cerebrospinal fluid of a Jamaican patient with tropical spastic paraparesis is de- scribed. The virus isolate is a typical type C retrovirus as seen by electron microscopy and is related to prototype HTLV-I isolated from patients with adult T-cell leukemia but is not identical to this prototype HTLV-I as seen by Restriction Enzyme Mapping. Human T-cell lymphotropic virus type I (HTLV-I) has been shown to be the etiological agent of tropical spastic para- paresis (TSP) in several parts of the world. We have com- pared a strain isolated from the cerebrospinal fluid of a Jamaican TSP patient with a prototype HTLV-I isolated from a patient with adult T-cell leukemia (ATL), by Restriction Enzyme Mapping. TSP is a neurological disorder characterized by paraparesis and spasticity of the lower limbs, which is sometimes associated with posterior column and superficial sensory change. Endemic TSP occurs in diverse geographical regions (1), and recently IgG antibodies to HTLV-I were found in several Caribbean islands (2-4), South America (3, 5, 6), the Seychelles islands (7), parts of Africa (8), and also in patients in the more temperate climate of southern Japan, where an identical syndrome called HTLV- I-associated myelopathy (HAM) has been described (9). Molecular cloning and Restriction Enzyme Mapping of avian adenovirus type 8 DNA

Krell, Éva NagyAbstract:Avian adenovirus (AAV) type 8 was cultured in an avian hepatoma cell line designated CH-SAH and the viral DNA extracted and purified. Restriction Enzyme analysis of viral DNA using the endonucleases ApaI, EcoRI, HindIII, KpnI, NotI, SpeI, StuI and XbaI was carried out, and fragments representing the entire genome were cloned. According to the Restriction Enzyme fragments, the size of the AAV type 8 genome was calculated to be 44.7 kb. Subcloning of viral DNA fragments and hybridization studies using selected viral DNA fragments facilitated the construction of the physical map of AAV type 8 DNA.
