    Abstract:

    By analyzing stomach content,including the food composition,feeding ecological type,feeding intensity and trophic level of the escaped cultured red drum,Sciaenops ocellatus in Zhejiang sea area was studied.It was shown that red drum belongs to nektivorous fish,which mainly feeds on Pisces,Macrura,Brachyura,Cephalopoda,as well as Stomatopoda and Polychaeta.Their food composition and feeding ecological classification are very different from those populations living in the natural habitat.The feeding intensity of red drum was 80.28%,the highest full stomach coefficient and its average were 60.93 and 10.26,respectively.According to calculation,the feeding niche breadth was 4.09 and the trophic level was 3.43.Red drum in Zhejiang sea area is a kind of raptorial middle level carnivore.

  • Preliminary study on growth characteristics of escaped red drum Sciaenops ocellatus in Zhejiang sea area

    Abstract:

    Study on the main biological characteristics of Sciaenops ocellatus in Zhejiang sea area was conducted on fish samples collected in 2004~2006. We obtained the regression equations of total length-body length, body weight-net weight, and length-weight, as well as the von-Bertalanffy growth equation (BL∞=951.09 mm, k=0.27, t0=0.57;TL∞=1 144.61 mm, k=0.26, t0=0.52; WB∞=14 468.07g, tx=4.514, Φ'BL=5.388) and relative fatness index (Q=1.82) by analyzing measurable characteristics and age. Compared to previous studies, it was concluded that S.ocellatus has adapted well to the sea area of Zhejiang, and achieved similar growth rate as those in the American sea areas. Once the S.ocellatus reproduce massively, it may pose a potential threat to the endemic species of this area.

    article i nfo Article history: A mapping tool for the economically important, aquacultured species red drum (Sciaenops ocellatus )i s described. The tool consists of multiplex panels, each of which contains 3-4 microsatellite markers that span individual linkage groups, with a spacing of ≤25 cM in either male- or female-specific maps or a combined map. A total of 27 newly-designed microsatellite markers were added to a previously generated map and then localized to specific linkage groups, using a de novo single-pair cross, in order to demonstrate the utility of the tool. All 27 markers were successfully localized when 100 progeny were genotyped; 26 of the markers were localized when 25, 50, or 75 progeny were genotyped. The tool will allow researchers without access to tissue from original crosses to localize genes of interest, including quantitative trait loci (QTL), to specific linkage groups (chromosomes), using a subset of microsatellite markers.

    Summary Second-generation, sex-specific genetic linkage maps were generated for the economically important estuarine-dependent marine fish Sciaenops ocellatus (red drum). The maps were based on F1 progeny from each of two single-pair mating families. A total of 237 nuclearencoded microsatellite markers were mapped to 25 linkage groups. The female map contained 226 markers, with a total length of 1270.9 centiMorgans (cM) and an average inter-marker interval of 6.53 cM; the male map contained 201 markers, with a total length of 1122.9 cM and an average inter-marker interval of 6.03 cM. The overall recombination rate was approximately equal in the two sexes ($:# = 1.03:1). Recombination rates in a number of linkage intervals, however, differed significantly between the same sex in both families and between sexes within families. The former occurred in 2.4% of mapped intervals, while the latter occurred in 51.2% of mapped intervals. Sex-specific recombination rates varied within chromosomes, with regions of both female-biased and male-biased recombination. Original clones from which the microsatellite markers were generated were compared with genome sequence data for the spotted green puffer, Tetraodon nigroviridis; a total of 43 matches were located in 17 of 21 chromosomes of T. nigroviridis, while seven matches were in unknown portions of the T. nigroviridis genome. The map for red drum provides a new, useful tool for aquaculture, population genetics, and comparative genomics of this economically important marine species.

    One hundred nuclear-encoded microsatellites from a genomic library of red drum (Sciaenops ocellatus) were isolated and characterized. Eight microsatellites had tetranucleotide motifs; 92 had dinucleotide motifs. The average number of alleles per microsatellite (sample of 22-24 fish) was 17.7 (range = 2-30); gene diversity averaged 0.796 (range = 0.227-1.000). Following Bonferroni correction, genotype frequencies at 90 microsatellites did not deviate significantly from Hardy-Weinberg equilibrium expectations. Occurrence of null alleles was inferred at 15 microsatellites; alleles differing by only a single base were observed at 11 microsatellites. The microsatellites developed should prove useful for population-genetic studies of 'wild' red drum and in construction of a genetic map.

    In this note, we document polymerase-chain-reaction (PCR) primer pairs for 101 nuclear-encoded microsatellitesdesigned and developed from a genomic library for red drum (Sciaenops ocellatus). Details of the genomic library construction, the sequencing of positive clones, primer design, and PCR protocols may be found in Karlsson et al. (2008). The 101 microsatellites (GENBA NK Accession NumbersEU015882-EU015982) were amplified successfully and used to genotype 24 red drum obtained from Galveston Bay, Texas (Table 1). A total of 69 of the microsatellites had an uninterrupted (perfect) dinucleotide motif, and 30 had an imperfect dinucleotide motif; one microsatellite had animperfect tetranucleotide motif, and one had an imperfect and compound motif (Table 1 ). Sizes of the cloned alleles ranged from 84 to 252 base pairs. A ‘blast’ search of the GENBANK database indicated that all of the primers and the cloned alleles were unique (i.e., not duplicated).

    Genetic eiects on cold-stress tolerance were assessed for red drum (Sciaenops ocellatus), an economically important sciaenid ¢sh in the southern USA. Thirty- ¢ve families were generated via 'natural'spawning of multiple sets of ¢ve breeders (three damstwo sires) in individual brood tanks. Oispring from the 35 families were transferred abruptly from an accli- mation temperature of � 24 1 Ct o 5.71C and main- tained subsequently at an average temperature of 6.5 1C. Oispring were assigned a posteriori to indivi- dual brood¢sh (dam and sire) based on genotypes at nuclear-encoded microsatellites. Heritability of the survival^time probability function was estimated using a proportional hazard approach and an animal- additive model. The estimated heritability was 0.20 (95% CI: 0.07^0.40), indicating a signi¢cant genetic component to acute cold-stress tolerance in red drum.

    Cathepsin S is a member of cysteine cathepsins and belongs to the cathepsin L-like family. In mammals, it is known to participate in various physiological processes and host immune defense. In teleost fish, the function of cathepsin S is less investigated. In the present work, we characterized a cathepsin S homologue (SoCatS) from red drum (Sciaenops ocellatus), a commercially valuable fish in Chinese mariculture. Like all cathepsin S, SoCatS possesses a peptidase domain with four catalytically essential residues (Gln140, Cys146, His285, and Asn305) conserved in the cathepsin S of different organisms. SoCatS shares 60-90% overall sequence identities with known teleost cathepsin S. Phylogenetic profiling indicated that SoCatS is evolutionally close to the cathepsin S of other teleost fish, especially Miichthys miiuy, a member of Sciaenidae family like red drum. SoCatS expression was detected in various tissues and was enhanced by bacterial infection. Purified recombinant SoCatS exhibited apparent peptidase activity with maximum at 50 degrees C and pH 7.5. This activity depended on the catalytic residue Cys146 and was severely reduced by the cathepsin inhibitor E-64.   • A Genetic Assessment of a Red Drum, Sciaenops ocellatus, Stock Enhancement Program

    The South Carolina Department of Natural Resources began a stock enhancement research program for red drum, Sciaenops ocellatus, as a possible management strategy for augmenting the availability of juveniles for recreational harvest in South Carolina estuaries. Between 1999 and 2011, approximately 6 million juveniles and 260 million larvae were stocked into the Charleston Harbor (CH) estuary, and proportions of stocked fish were found through standardized sampling as high as 49.6% within a subadult year-class and up to 12% within the spatially separate spawning population. This study evaluated the genetic influences of stocking on the spawning population, using microsatellite loci, and examined any changes in genetic diversity metrics over the course of the stock enhancement program. Percent contribution of stocked fish among subadult and adult collections was compared against measures of genetic diversity. No statistically significant correlations were found among genetic diversity metrics, suggesting that responsible stocking of red drum as small juveniles has not altered the genetic diversity of adults spawning offshore the CH. Life-history characteristics likely play a large role in mitigating changes in genetic diversity, despite high contributions of stocked fish in some years, because red drum exhibit a long adult life span and benefit from overlapping generations.

    We investigated the metabolic effects of four different commercial soy-based protein products on red drum fish (Sciaenops ocellatus) using nuclear magnetic resonance (NMR) spectroscopy-based metabolomics along with unsupervised principal component analysis (PCA) to evaluate metabolic profiles in liver, muscle, and plasma tissues. Specifically, during a 12 week feeding trial, juvenile red drum maintained in an indoor recirculating aquaculture system were fed four different commercially available soy formulations, containing the same amount of crude protein, and two reference diets as performance controls: a 60% soybean meal diet that had been used in a previous trial in our lab and a natural diet. Red drum liver, muscle, and plasma tissues were sampled at multiple time points to provide a more accurate snapshot of specific metabolic states during the grow-out.     In the present study, quantitative data were collected to clarify the relationship between calling, call structure and eggs produced in a captive population of red drum Sciaenops ocellatus. Sciaenops ocellatus were held in four tanks equipped with long-term acoustic loggers to record underwater sound throughout a simulated reproductive season. Maximal sound production of captive S. ocellatus occurred when the photoperiod shifted from 13·0 to 12·5 h of light, and the water temperature decreased to c. 25° C. These captive settings are similar to the amount of daylight and water temperatures observed during the autumn, which is the primary spawning period for S. ocellatus. Sciaenops ocellatus exhibited daily patterns of calling with peak sound production occurring in the evenings between 0·50 h before dark and 1·08 h after dark. Spawning occurred only on evenings in which S. ocellatus were calling, and spawning was more productive when S. ocellatus produced more calls with longer durations and more pulses. This study provides ample evidence that sound production equates to spawning in captive S. ocellatus when calls are longer than 0·8 s and contain more than seven pulses. The fact that more calling, longer calls and higher sound pressure levels are associated with spawns that are more productive indicates that acoustic metrics can provide quantitative information on spawning in the wild.
