  • Antifungal Activities of Some Leaf Extracts on Seed-Borne Fungi of African Yam Bean Seeds, Seed Germination and Seedling Emergence

    Abstract:

    The efficacy of leaf extracts of basil (Ocimum basilicum), bitter leaf (Vernonia amygdalina), lemon grass (Cymbopogen citratus), neem (Azadirachta indica) and paw-paw (Carica papaya) on major Seed-Borne Fungi: Aspergillus niger, Aspergillus flavus, Botryodiplodia theobromae and Fusarium moniliforme of African yam bean ( Sphenostylis stenocarpa ) seeds, and on seed germination and seedling emergence were studied in vitro and in vivo. All the plants leaf extracts (crude and aqueous) significantly (P . 0.05) reduced the incidence of Seed-Borne Fungi tested and increased seed germination and seedling emergence except lemon grass leaf extract when compared with the untreated control. Neem extract was the most effective while lemon grass extract was the least. Crude extracts from all the plant leaves tested increased seed germination and seedling emergence of African yam bean seeds and gave significant (P. 0.05) reduction of mycelial growth of all the Fungi tested when compared with their aqueous extracts. Leaf extracts of neem, basil, bitter leaf and paw-paw, which are cheap and environmentally safe, are promising for protecting African yam bean seeds against major Seed-Borne Fungi and in the improvement of the crop.

    Abstract:

    The efficiency of four seed health testing techniques namely the blotter, deep- freezing blotter, ragdoll and agar plate methods in detecting Seed-Borne Fungi of African yam bean,  Sphenostylis stenocarpa (Hochst ex. A. Rich) Harms seeds was evaluated. The blotter method was found to be the most suitable testing technique for detecting Aspergilus flavus, A niger, A terreus, A. ochraceus, Botryodiplodia theobromae, Colletoritchum dematium, C. gloeosporiodes and Curvularia lunata in seeds pre-treated with 1 % available chlorine for 5 minute as well as in untreated seeds. The deep-freezing blotter method was superior to the other methods in detecting Fusarium moniliforme, F. semitectum, Chaetomium sp. and Cephalsorium sp. The incidence of Macrophomina phaseolina and Penicillium spp. were recorded more in the agar plate method than in the other Fungi recorded but gave highest percentage seed germination in both untreated and chlorine pre-treated seeds in all the four sampled tested.     Experiments were conducted under laboratory condition to examine the efficacy of Chitosan and Yeast Elicitor to suppress the growth of Seed-Borne Fungi of cucurbitaceous vegetables. Seeds of bottle gourd, sweet gourd, snake gourd, wax gourd and cucumber were collected from seed traders of Mymensingh districts and different seed borne Fungi were isolated, purified and identified. Fourteen fungal species belonging to twelve genera consisting of Aspergillus flavus, Aspergillus niger, Botrytis cinerea, Fusarium moniliforme, Fusarium oxysporum, Phoma exigua, Rhizopus stolonifer , Macrophomina phaseolina, Penicillium spp. , Curvularia lunata, Chaetomium spp., Colletotrichum spp., Cercospora spp. and Alternaria alternata were isolated and identified. Four concentrations of Chitosan and Yeast Elicitors solutions (200, 500, 1000 & 2000 ppm) including one positive control Vitavax-200 WP (0.35%) were evaluated for controlling Seed-Borne Fungi. Among the seed treating agents Chitosan (2000 ppm) and Yeast Elicitor (2000 ppm) showed better performance in suppressing the Seed-Borne Fungi. Chitosan (2000 ppm) showed superior performance than Yeast Elicitor (2000 ppm).   • Health and Quality of Wheat Seed Samples Collected from Sadar Upazilla of Thakurgaon District and Control of Seed-Borne Fungi

    Abstract:

    Wheat seed samples were collected from ten unions of sadar upazilla of Thakurgaon district in wheat growing season of 2011. Seeds were tested by blotter method at Seed Pathology Center (SPC), Bangladesh Agricultural University, Mymensingh during the months April to November 2011 for recording and identifying the Seed-Borne Fungi associated with wheat seeds. The health status of 20 seed samples were determined whereas five genera Fungi were identified from a total of six fungus. The Fungi were Bipolaris sorokiniana, Alternaria tenuis, Fusarium spp, Penicillium sp, Aspergillus flavus and Aspergillus niger. Prevalence of the total as well as the individual Seed-Borne fungal infections that were recorded varied significantly with respect to wheat varieties and sources of seed collection. Seed samples collected from Jagonathpur and Gorea unions of sadar upazilla showed highest percentage of Seed-Borne infection compared to the samples collection from other unions for both varieties. The Seed-Borne fungal infection in Hazar-8 and Satabdi collected from other unions showed lowest percentage of Seed-Borne fungal infection than that of Jagonathpur and Gorea unions. Seed germination also varied significantly depending on the varieties and the seed sources and a positive correlation between seed germination and Seed-Borne fungal infections were observed. Three seed treating agents viz., neem leaf extract 1:2, hot water and Provax were evaluated for controlling Seed-Borne Fungi associated with wheat seeds. Among the seed treating agents, Provax was found superior to reduce the Seed-Borne infection of wheat. The results also showed that neem leaf extract at 1:2 dilutions was observed to be the most effective in reducing Seed-Borne Fungi with highest percentage of germination followed by hot water treatment.
