    The efficacy of the cholinergic agonist pilocarpine to evoke generalized Seizures in rats was examined over the 24-h photocycle. Both Seizure latency and severity during the 24-h period exhibited marked diurnal variation. Seizure Activity generalized more rapidly throughout the dark phase and was often characterized by wild running clonus and tonic extensor convulsions. In contrast, electrographic Seizures during the light phase were mainly associated with facial/forelimb clonus and rearing typical of pilocarpine-induced Seizures. These data suggest that the sensitivity of the rodent brain to pilocarpine exhibits day-night variation and Seizure Activity induced during the dark phase may generalize via the brainstem.

    Anti-convulsant effects of physical exercise and lipoic acid (LA), also referred to as thioctic acid with antioxidant Activity, were investigated using chemical induced Seizure model. We investigated the synergic effect of physical exercise and LA on kainic acid-induced Seizure Activity caused by oxidative stress. After 8 weeks of swimming training, body weight decreased and endurance capacity increased significantly compared to sedentary mice. Kainic acid (30 mg/kg, i.p.) evoked Seizure Activity 5 min after injection, and Seizure Activity peaked approximately 80 min after kainic acid treatment. In addition, the synergic effect of exercise and LA was found in PGx Activity compared to separated treatment ("LA + EX + KA": 37.3 ± 1.36; p < 0.05 vs. "LA + KA" and "EX + KA" group). These results indicate that physical exercise along with LA could be a more efficient method for modulating Seizure Activity and oxidative stress.

    The synergic effect of regular exercise and resveratrol, a polyphenolic compound with potent antioxidant Activity, was investigated against kainate-induced Seizures and oxidative stress in mice. After 6 weeks of swimming training, the total body weight decreased and the blood concentration of lactate stabilized statistically in comparison with the sedentary mice, indicate that the training program increased the aerobic resistance of mice. Kainate (30 mg/kg) evoked Seizure Activity 5 min after injection, and Seizure Activity was measured Seizure rating scores every 5 min up to 2 h. As previously well known experiments, regular exercise and resveratrol (40 mg/kg, daily supplementation for 6 weeks) have an inhibitory effect on kainate-induced Seizure Activity and oxidative stress. In particularly, a synergistic cooperation of regular exercise and resveratrol was observed in Seizure Activity, mortality and oxidative stress especially in SOD Activity. These results suggest that regular exercise along with an anti-convulsant agent such as resveratrol could be a more efficient method for the prevention of Seizure development than exercise alone.

    Status epilepticus is a life-threatening form of Seizure Activity that represents a major medical emergency associated with significant morbidity and mortality. Protein Kinase A is an important regulator of synaptic strength that may play an important role in the development of status epilepticus-induced neuronal pathology. This study demonstrated an increase in PKA Activity against exogenous and endogenous substrates during later stages of SE. As SE progressed, a significant increase in PKA-mediated phosphorylation of an exogenous peptide substrate was demonstrated in cortical structures. The increased Activity was not due to altered expression of either regulatory or catalytic subunits of the enzyme. Through the use of phospho-specific antibodies, this study also investigated the effects of SE on the phosphorylation of the GluR1 subunit of the AMPA subtype of glutamate receptor. After the onset of continuous Seizure Activity, an increase in phosphorylation of the PKA site on the GluR1 subunit of the AMPA receptor was observed. These data suggest a potential mechanism by which SE may increase neuronal excitability in the cortex, potentially leading to maintenance of Seizure Activity or long-term neuronal pathology.

    Electrographic Seizures are a feature of organophosphate anticholinesterase intoxication. Clinical studies of pesticide poisonings suggest that Seizures are more common in children than in adults. Since flaccid paralysis, a characteristic sign of organophosphate poisoning, can mask convulsions, the most reliable indicator of Seizures is the electroencephalogram, but this has not been widely used in clinical studies. Seizures can rapidly progress to status epilepticus, contributing to mortality and, in survivors, to neuronal damage and neurological impairment. Anticonvulsant drugs can significantly reduce the lethal and toxic effects of these compounds. A benzodiazepine, usually diazepam, is the treatment currently indicated for control of Seizures. Animal studies have indicated that the early phase of Seizure Activity (0-5 min after Seizure onset) is purely cholinergic, predominantly involving muscarinic mechanisms. Seizure Activity subsequently progresses through mixed cholinergic and noncholinergic modulation (5-40 min) into a final noncholinergic phase. Neuropathology caused by Seizures is most likely associated with glutamatergic excitotoxicity. Future prospects for improved treatments include new benzodiazepines, glutamate receptor antagonists, antimuscarinics with additional antiglutamatergic Activity and adenosine receptor antagonists.
