    Abstract:

    Objective To investigate the effect of mesalazine treatment on the expression of NF-κB p65 in the Spleen Tissue of rats with 2,4,6-trinitrobenzenesulfonic acid( TNBS)-induced ulcerative colitis.Methods Forty-two SD rats were randomly divided into the control group,colitis model group and mesalazine group( 14 rats in each group).Apart from the control group,the rats in the other two groups were induced to ulcerative colitis by 2,4,6-trinitrobenzenesulfonic acid.And 2 days later,rats in the control and colitis model groups were administered intragastrically with distilled water at a dose of 3 mL,while rats in the mesalazine group was intragastrically given suspension of mixed mesalazine and distilled water at a dose of 3 mL.All rats were treated for 15 d.Spleen Tissue samples were taken to detect the expression of mRNA of NF-κB p65 by RT-PCR and the protein by Western blot.Results The expression of the mRNA and protein of NF-κB p65 were significantly higher in the model group than those in the control group( all P 0.05).Treatment with mesalazine significantly decreased the expression of the mRNA and protein of NF-κB p65( all P 0.05).Conclusion Mesalazine plays an important role in the treatment of ulcerative colitis by down-regulating the expression of the mRNA and protein of NF-κB p65 in the Spleen Tissue of rats with TNBS-induced ulcerative colitis.

    Objective To observe the effect of Wumeiwan on the expression of β-arrestin1 in the Spleen Tissue of rats with 2,4,6-trinitrobenzenesulfonic acid(TNBS)-induced ulcerative colitis.Methods Fifty-six SD rats were randomly divided into the control group,colitis model group,mesalazine group and Wumeiwan group(14 rats in each group).Apart from the control group,rats in the other three groups were induced to experimental colitis by 2,4,6-trinitrobenzenesulfonic acid.And 2 days later,rats in the control group and colitis model group were administered intragastrically with normal saline at a dose of 3 mL,while rats in the mesalazine group and Wumeiwan group were intragastrically given mesalazine(50 g/L) and Wumeiwan(0.51 g/L) at a dose of 3 mL,respectively.All rats were treated for 15 d.Spleen Tissue samples were taken to detect the expression of mRNA of β-arrestin1 by RT-PCR and the expression of β-arrestin1 protein by Western blot.Results Compared with the control group,the expression of β-arrestin1 mRNA and β-arrestin1 protein was significantly increased in the colitis model group(all P 0.05);compared with the colitis model group,the expression of the mRNA and protein of β-arrestin1 in the Wumeiwan and mesalazine groups was significantly decreased(all P 0.05).Conclusion Wumeiwan can down-regulate the expression of the mRNA and protein of β-arrestin1 in the Spleen Tissue of rats with TNBS-induced ulcerative colitis.

    Abstract:

    Aridity and heat are significant environmental stressors that affect sheep adaptation and adaptability, thus influencing immunity, growth, reproduction, production performance, and profitability. The aim of this study was to profile mRNA expression levels in the Spleen of indigenous Kazakh sheep breed for comparative analysis with the exotic Suffolk breed. Spleen histomorphology was observed in indigenous Kazakh sheep and exotic Suffolk sheep raised in Xinjiang China. Transcriptome sequencing of Spleen Tissue from the two breeds were performed via Illumina high-throughput sequencing technology and validated by RT-qPCR. Blood cytokine and IgG levels differed between the two breeds and IgG and IL-1β were significantly higher in Kazakh sheep than in Suffolk sheep (p < 0.05), though Spleen Tissue morphology was the same. A total of 52.04 Gb clean reads were obtained and the clean reads were assembled into 67,271 unigenes using bioinformatics analysis. Profiling analysis of differential gene expression showed that 1158 differentially expressed genes were found when comparing Suffolk with Kazakh sheep, including 246 up-regulated genes and 912 down-regulated genes. Utilizing gene ontology annotation and pathway analysis, 21 immune- responsive genes were identified as Spleen-specific genes associated with adaptive traits and were significantly enriched in hematopoietic cell lineage, natural killer cell-mediated cytotoxicity, complement and coagulation cascades, and in the intestinal immune network for IgA production. Four pathways and up-regulated genes associated with immune responses in indigenous sheep played indispensable and promoting roles in arid and hot environments. Overall, this study provides valuable transcriptome data on the immunological mechanisms related to adaptive traits in indigenous and exotic sheep and offers a foundation for research into adaptive evolution.

    Abstract:

    The iron-specific magnetic susceptibility of Tissue iron deposits is used in the field of non-invasive measurement of Tissue iron concentrations. It has generally been assumed to be a constant for all Tissue and disease types. The iron-specific magnetic susceptibilities chi(Fe) for Spleen Tissue samples from 7 transfusion dependent beta-thalassaemia (beta-thal) patients and 11 non-transfusion dependent beta-thalassaemia/Haemoglobin E (beta/E) patients were measured at 37 degrees C. Both groups of patients were iron loaded with no significant difference in the distribution of Spleen iron concentrations between the two groups. There was a significant difference between the mean chi(Fe) of the Spleen Tissue from each group. The non-transfusion dependent beta/E patients had a higher mean (+/-standard deviation) Spleen chi(Fe) (1.55+/-0.23 x 10(-6) m(3)/kg Fe) than the transfusion dependent beta-thal patients (1.16+/-0.25 x 10(-6) m(3)/kg Fe). Correlations were observed between chi(Fe) of the Spleen Tissue and the fraction of magnetic hyperfine split sextet in the (57)Fe Mossbauer spectra of the Tissues at 78 K (Spearman rank order correlation r=-0.54, p=0.03) and between chi(Fe) of the Spleen Tissue and the fraction of doublet in the spectra at 5 K (r=0.58, p=0.02) indicating that chi(Fe) of the Spleen Tissue is related to the chemical speciation of the iron deposits in the Tissue.

    Patients with β-Thalassemia major are transfusion dependent and become iron loaded owing to the large number of blood transfusions they receive. Patients with β-Thalassemia/Hb E become iron loaded even in the absence of regular blood transfusions because of increased erythropoeisis and stimulated dietary iron absorption. In both cases, iron accumulates in Tissues such as the liver, Spleen, and heart in the form of insoluble iron(III) oxyhydroxide nanoparticles found in ferritin and hemosiderin. The body iron burden is usually estimated by measurement of the liver iron concentration (LIC). Non-invasive techniques for measuring LIC often exploit the large magnetic susceptibility of Tissue iron, taken to be 1600 x 10−9 m3/kgFe at body temperature. The current study examined Spleen Tissue samples from Australian β-Thalassemia major patients (n = 7) who had undergone chelation and transfusion therapy and Thai β-Thalassemia/Hemoglobin E disease patients (n = 11) who had received very few transfusions and no chelation therapy. Magnetometry measurements of the iron specific magnetic susceptibility of the Spleen Tissue samples ranged from 800 x 10−9 m3/kgFe to 1800 x 10−9 m3/kgFe. The mean iron specific magnetic susceptibility for the β-Thal major Spleen Tissue was approximately 25% lower than that for the β-Thal/Hb E samples (p = 0.007 Student’s t test). To investigate the underlying cause of this variability the physico-chemical properties of the Tissue iron deposits were probed using cryogenic 57Fe Mossbauer spectroscopy. Spectra recorded at 78K showed that the β-Thal major Tissues contained iron with a greater fraction in the form of a more crystalline chemical species of iron oxyhydroxide than the β-Thal/Hb E Tissues (p =0.01 Wilcoxon-Mann-Whitney test). Significant correlations (Spearman rank correlation tests) were identified between the iron specific magnetic susceptibility measured at body temperature and the Mossbauer spectral parameters recorded at both 5 K (p = 0.02) and at 78 K (p = 0.03). This relationship between the iron specific magnetic susceptibility and the chemical speciation of iron oxyhydroxide Tissue deposits implies that the magnetic behaviour of Tissue iron deposits is determined by the physical and chemical nature of the iron deposits. Variability in the iron specific magnetic susceptibility of Tissues could impact on the precision of non-invasive techniques of Tissue iron measurement based on magnetic susceptibility measurements.

    Abstract:

    Mossbauer spectra of 12 normal human Spleen and 12 normal human liver samples (post mortem) from Australia and Thailand have been recorded at 78 K. The spectra show the presence of iron in the form of ferrihydrite, together with some deoxyhemoglobin and methemoglobin in some samples. The spectra were used in conjunction with elemental analysis to calculate the non-heme iron concentrations in the Tissues. The mean non-heme iron concentration in the Thai livers was significantly less than that for the Australian samples. The goethite-like form of hemosiderin that has been observed in some pathological Tissues was not detected.
