    Abstract:

    Malaria transmission in Gunung Jati Village, Banjarmegara needs serious attention because this village is malaria endemic area. This study aimed to investigate the biotic and abiotic environmental conditions at Anopheles spp. breeding sites in Gunung Jati village, Banjarnegara. Anopheles spp. larvae collection, measurement of abiotic factors and observation of microbiota on water samples were conducted on May 2015. Anopheles spp. larva was found  in puddles on the banks of the river with larvae density was 5 larvae per dip. Water temperature conditions in all breeding sites ranged from 26-32 oC, the degree of acidity was neutral, with varying light intensity. Synedra are the most commonly found algae from all observed breeding sites. The environmental conditions were in accordance with the optimum conditions that support the development of Anopheles spp. larva

    Abstract:

    ABSTRAK Potensi penularan malaria di Desa Gunung Jati, Banjarnegara perlu diwaspadai mengingat daerah tersebut merupakan daerah endemis malaria. Penelitian ini bertujuan untuk menggambarkan kondisi lingkungan biotik dan abiotik pada tempat perkembangbiakan Anopheles spp. yang ditemukan di Desa Gunung jati, Banjarnegara. Kegiatannya meliputi penangkapan larva, pengukuran faktor abiotik dan pengamatan biota mikro pada sampel air pada bulan Mei 2015. Larva Anopheles spp. ditemukan pada genangan air di pinggiran sungai dengan kepadatan larva rata-rata 5 ekor larva per cidukan. Kondisi suhu air di semua  tempat perkembangbiakan yang ditemukan berkisar antara 26-32 °C, derajat keasaman netral, dengan intensitas pencahayaan bervariasi. Mikro alga Synedra paling sering ditemukan dari semua tempat perkembangbiakan yang diamati.     Abstract:

    Algal-bacterial associations were studied in unialgal xenic cultures of Synedra acus subsp. radians, Asterionella formosa and Fragillaria crotonensis planktonic diatoms from Lake Baikal, using epifluorescent and scanning electron microscopy. It was found that rod- and ovoid-shaped bacteria colonized cell walls of diatoms. Cloning and sequencing of fragments of 16S rRNA gene in diatom cultures revealed members of Gammaproteobacteria (Pseudomonas sp.), Betaproteobacteria (Janthinobacterium sp., Hydrogenophaga sp., Methylophilus sp.), Bacteroidetes (Flavobacterium sp., Pedobacter sp.), and Acinobacteria (Nocardioides sp.).

    Abstract:

    Complete chloroplast genome of diatom alga Synedra acus possesses canonical quadripartite structure with two inverted repeats containing ribosomal RNA gene loci that separate small and large single-copy regions. Chloroplast genome maps as a circular molecule of 116 251 bp. It encodes 27 tRNAs, three rRNAs, two small RNA genes, and 128 protein-coding genes. Comparison of the genic features across diatom chloroplast genomes reveals the absence of an overlap between atpD and atpF gene coding sequences that is present in other plastid genomes of diatom origin. This feature is a clear synapomorphy of S. acus plastid genome that is likely a result of either relaxed constraints or extensive selection forces acting upon atpF gene. We also characterized nuclear-encoded acyl-carrier protein gene with chloroplastic targeting in S. acus . The transfer of acpp gene into the nuclear host genome is hypothesized to have occurred independently in several lineages of diatoms.

    Abstract:

    Production of axenic cultures of diatoms is a highly relevant task, as these widespread organisms are objects of research in different fields of biology, from ecology to whole-genome studies, and promising as producers of various organic molecules, including biologically active substances. However, culturing of diatoms isolated from natural communities is accompanied by the development of associated microorganisms. We propose a procedure for establishing an axenic culture of the freshwater diatom Synedra asus subsp. radians (Kutz.) Skabitsch. from Lake Baikal. Such a culture has been produced and maintained by subculturing in our laboratory. The procedure involves filtration (to remove free bacteria), detergent treatment (to remove microorganisms associated with diatom frustules), selection of effective antibiotics and their concentrations nontoxic for diatoms (by the disk diffusion method), cell treatment with the selected antibiotic (ciprofloxacin), repeated filtration, and monoclonal culturing of diatom cells. Axenity of the culture was verified microscopically (after DAPI staining) and by molecular biological methods.

    Alaş, Ali (Aksaray, Yazar)In this study, alimentary canal contents of 188 tench (Tinca tinca L., 1758) caught from Beyşehir Lake (Turkey) between April 2004 and March 2005 were examined for feeding habits. Phytoplanktonic and zoooplanktonic organisms, insects, detritus (animal and vegetable) and two most common parasites of tench, Ligula intestinalis and Asymphylodora tincae were determined in observed in the alimentary canal of fish. There was a significant difference between the frequency of the existance of Chlorella, Coscinidiscus, Fragilaria, Mougetia, Phytoconis, Pinnularia, Rivularia, Synedra, Ulotrix, Brachionus, Gammarus sp., and Ligula intestinalis among seasons (df=3, P<0.05). Only 6 of these genera (Chaetophora, Coscinidiscus, Phytoconis, Surirella, Synedra, Daphnia) had a significant difference among age classes (df=5, P<0.05). Additionally, it was found that only 5 of the 188 tench (2.66%) were observed to have almost empty alimentary canal. Algae and macrophytes were found to be the most considerable food types in the present study, which is followed by zooplankton and insect larvae. Konu Alanı:Bu çalışmada, beslenme alışkanlıklarını belirlemek için Nisan 2004 ve Mart 2005 tarihleri arasında Beyşehir Gölü (Türkiye)’nden yakalanan 188 adet Kadife Balığı (Tinca tinca L., 1758)’nın sindirim kanalı incelenmiştir. Balıkların sindirim kanalında fitoplanktonik ve zooplanktonik organizmalar, böcekler, detritus (bitkisel ve hayvansal) ve kadife balığında en yaygın iki parazit türü olan Ligula intestinalis ve Asymphylodora tincae tespit edilmiştir.
