    Therapeutic Principle was a rational utilization of intensive multi-reason.On the one hand,Therapeutic Principle contained content of syndromes,etiopathogenisis,pathogenesis and regulation of compatibility and formula,and implicates inherent logicality between syndrome and formula.On the other hand,Therapeutic Principle took an important role in summary and logical classification towards syndrome,prescription and medicine.The theoretical system of theory,Therapeutic Principle,prescription and medicine was developed and improved with the further comprehension in clinical practice and theory.As for the research of inheritance of the famous traditional Chinese medicine(TCM)veteran doctors’clinical experience,consistency in theory,Therapeutic Principle,prescription and medicine was the premise,and combined disease with syndrome was the foundation.It was a significant breakthrough for developing and innovating clinical experience and academic thought of the famous TCM veteran doctors in the research of their clinical experience from the aspect of Therapeutic ways.Theory refining on the basis of above research will be the core driving force of TCM development.

    Abstract:

    Probiotics are nonpathogenic microorganisms mostly of human origin which, when administered in adequate amounts, confer a health benefit on the host and enable to prevent or improve some diseases. Probiotics may be a natural temporary constituent of the resident intestinal microflora, but their concentration is not sufficient for Therapeutic purposes. The microbiota, the intestinal epithelium, and the mucosal immune system constitute the gastrointestinal ecosystem. All three components are essential for complete functional and developmental maturity of the system. The viability of intestinal microflora (including probiotic strains) requires the availability of nutritional substrates (prebiotics), i.e. various types of fiber and oligosaccharides. Prebiotics are cleaved by microbial enzymes to numerous substances (short-chain fatty acids, aminoacids, polyamines, growth factors, vitamins and antioxidants) indispensable for metabolic and functional activities of the intestinal mucosa. The principal probiotics in use include lactobacilli, bifidobacteria, some nonpathogenic strains of Escherichia coli , and Saccharomyces boulardii . These microbiota display favourable effects which qualify them for Therapeutic use. For this purpose, probiotics have to fulfill a series of requirements verifying their efficacy and safety. Experimental and clinical studies examine the prerequisites for the administration of probiotics in digestive diseases, allergic and atopic affections, as well as in some extraintestinal conditions. Future goals of probiotic application include genomic analysis, controlled postnatal colonisation of the digestive tract, the use of probiotics as carriers of peroral vaccines, and recombinant probiotics with in-situ production and targeted application of Therapeutic molecules.

  • Clinical Efficacy of a Novel Therapeutic Principle, Anakoinosis

    Abstract:

    Classic tumor therapy, consisting of cytotoxic agents and/or targeted therapy, has not overcome Therapeutic limitations like poor risk genetic parameters, genetic heterogeneity at different metastatic sites or the problem of undruggable targets. Here we summarize data and trials principally following a completely different treatment concept tackling systems biologic processes: the Principle of communicative reprogramming of tumor tissues, i.e., anakoinosis (ancient greek for communication), aims at establishing novel communicative behavior of tumor tissue, the hosting organ and organism via re-modeling gene expression, thus recovering differentiation, and apoptosis competence leading to cancer control – in contrast to an immediate, “poisoning” with maximal tolerable doses of targeted or cytotoxic therapies. Therefore, we introduce the term “Master modulators” for drugs or drug combinations promoting evolutionary processes or regulating homeostatic pathways. These “master modulators” comprise a broad diversity of drugs, characterized by the capacity for reprogramming tumor tissues, i.e., transcriptional modulators, metronomic low-dose chemotherapy, epigenetically modifying agents, protein binding pro-anakoinotic drugs, such as COX-2 inhibitors, IMiDs etc., or for example differentiation inducing therapies. Data on 97 anakoinosis inducing schedules indicate a favorable toxicity profile: The combined administration of master modulators, frequently (with poor or no monoactivity) may even induce continuous complete remission in refractory metastatic neoplasia, irrespectively of the tumor type. That means recessive components of the tumor, successively developing during tumor ontogenesis, are accessible by regulatory active drug combinations in a Therapeutically meaningful way. Drug selection is now dependent on situative systems characteristics, to less extent histology dependent. To sum up, anakoinosis represents a new substantive therapy Principle besides novel targeted therapies.
