    Abstract:

    This study had been carried out at the Department of Ornamental HortiCulture, Faculty of Agic., Cairo University. The Experiments had been done during the period from 1999 to 2004 at the Central Laboratory for Development of Date Palm Research, Giza, Egypt and Laboratory of Tissue Culture, El-Zohreya Botanical Garden, Horticultural Services Unit, Agricultural Research Center (A.R.C.), with the aim of investigating the factors affecting the different stages of in vitro propagation of date palm (Phoenix dactylifera L.) cv. Sewei. Seven experiments were conducted. The best results (Free of contamination, healthy explants) were obtained by using Clorox at 80%, and mercuric chloride at 1 mg/l. The greatest formation of callus was obtained on shoot tip explants grown on an MS medium supplemented with 100 mg 2, 4-D + 3 mg 2iP/l.Results indicated that a high strength of the MS medium (full or 3⁄4 strength) and a long Culture duration (18 weeks) were the most favourable conditions for friable callus formation. The highest number of somatic embryos was obtained when sucrose was added at 30 g/l, combined with the longest Culture duration (24 weeks).The longest shoots were those formed on explants Cultured on an MS medium containing 40 g sucrose /l for a period of 18 weeks. Culturing date palm explants in a full strength medium supplemented with 1.5 mg NAA/l gave the highest values for number of roots and root length. The best results, in terms of number of roots and root length, were recorded when NAA was used at 1.5 mg/l, combined with the highest charcoal concentration (9 g/l).

    Abstract:

    Over 100 years ago, Haberlandt envisioned the concept of plant Tissue Culture and provided the groundwork for the cultivation of plant cells, Tissues and organs in Culture. Initially plant Tissue Cultures arose as a research tool and focused on attempts to Culture and study the development of small, isolated cells and segments of plant Tissues. At the peak of the plant Tissue Culture era in the 1980s, in a relatively short time, many commercial laboratories were established around the world to capitalize on the potential of micropropagation for mass production of clonal plants for the hortiCulture industry. Today plant Tissue Culture applications encompass much more than clonal propagation. The range of routine technologies has expanded to include somatic embryogenesis, somatic hybridization, virus elimination as well as the application of bioreactors to mass propagation. Perhaps the greatest value of these Tissue Culture technologies lies not so much in their application to mass clonal propagation but rather in their role underpinning developments and applications in plant improvement, molecular biology and bioprocessing, as well as being a basic research tool. Plant Tissue Culture Technique though an underutilized tool in Nigeria, it can be extensively applied in hortiCulture to increase crop production. This paper highlights some of the applications of plant Tissue Culture to hortiCulture, the achievements and limitations of Tissue Culture and some insights into current and possible future developments. With rapid population growth, the total acreages of fruits, vegetables and various ornamental plants have not been able to meet the needs of people in the developing countries.

    Medicinal plants have been the subject of man's curiosity since the time of human survival. A considerable majority of people of the world's population still rely on the traditional medicine for their primary health care necessities. Currently demand of herbal medicines has been enhanced; and it is very difficult to fulfill the demand from field plants. Hence, in vitro propagation of plant by Tissue Culture can be used to fulfill the requirement of medicinal plants. The Techniques applied in Tissue Culture are expensive; hence we try to summarize the data for low cost method for in vitro micropropagation of plant through Tissue Culture Technique. This can help the scholar working in developing of Tissue Culture methods for in vitro propagation of medicinal plant.

    This study was conducted during 2011 and 2012 seasons to compare growth, flowering and fruit setting behaviour, yield as well as physical and chemical characteristics of some dry date palm varieties (Melkaby, Gendiala and Sakkoti) propagated through seeds and Tissue Culture Techniques under Aswan region climatic conditions. This study clarified that major differences were observed on vegetative growth characters as well as flowering and fruiting aspects among the two propagation methods namely seeds and Tissue Culture as well as among the three date palm cvs Melkaby, Gendiala and Sakkoti. All the positive effects on the behaviour of growth and fruiting aspects were attributed to using propagation via seeds. The best date palm cvs successfully grown under Aswan environmental conditions are Gendiala, Melkaby and Sakkoti, in descending order based on their higher yields and fruit quality. We need in the future for evaluation of all date palm cvs produced from Tissue Culture grown in all environmental conditions under Egypt to survey the different problems and to select the best way for alleviating these problems especially at the older ages.
